Abstract

Propagation of mangosteen is challenging for many reasons, including limited seed set, slow rate of seedling growth, and difficulty with root formations. The objective of this research was to find the best combination of medium and plant growth regulator for in vitro growth and rooting of mangosteeen seed. Various types of explant (a whole seed; seed divided into 2, 3, and 4 cross sections; seed divided into 2, 3, and 4 longitudinal sections) were treated with five concentrations of benzyl amino purine (BAP; 0, 2.5, 5, 7.5, 10 mg/L) for shoot induction in ½ Nitrogen (N) Murashige and Skoog (MS) medium. The shoots were rooted on MS and woody plant medium (WPM) media with several combinations of indole butyric acid (IBA) and naphtalene acetic acid (NAA). Treatments for root induction were applied as follows: (i) low dose, given during induction of rooting, (ii) soaking the base of the shoots in medium treated with a high dose of auxin for 5 days, and then growing the shoots in MS ½ N with 1 mg/L NAA +  1 mg/L BAP medium. Our result show that BAP positively affected mangosteen bud growth. The best medium for mangosteen shoot regeneration was found to be  MS ½ N  + 5 mg/L BAP. This medium induced  the highest number of shoots from the seed explant cut into four cross sections. We found the best medium to induce in vitro rooting of mangosteen shoot was MS ½ N + 3 mg/L indole butiric acid (IBA) + 4 mg/L NAA medium. Some treatment negatively affected growth. Soaking the mangosteen shoot base in a medium with an overly high dose of auxin seemed to disrupt and inhibit growth of the mangosteen shoot.

Abstract

The objectives of this research were to composed organ from callus culture and to found the best concentration of plant growth regulator for organ growth from female flower explant of oil palm. This research has already done from June 2014 to May 2015 at Laboratory of Plant Physiology and Tissue Culture Department of Biology Faculty of Mathematics and Science University of North Sumatera. This research used Nonfactorial Completely Random Design. Explant was treated with five concentrations of 2,4-Dichlorophenoxy acetic acid (2,4-D; 99, 110, 120, 132, and 140 mg/L) for callus induction on Y3 medium (Eeuwens 1976). The result of this research showed that organ was formed from this treatment (basal segment of female flower explant) was root organ. 2,4-D plant growth regulator positively affected to growing of the root. The best result for time of callus induction, time of root growth, the highest percentage of explants that formed the root, fresh weight and dry weight of callus that has become the root generation was resulted from 99 mg/L 2,4-D.   Key words: Elaeis guineensis Jacq., female flower, plant growth regulator 2,4-D, organogenesis

Abstract

The research on induction of banana barangan shoot (Musa acuminata L.) from North Nias through tissue culture by giving 2,4-D and kinetin was conducted in the Laboratory of Tissue Culture  University of North Sumatera from May until October 2014. The main objective of this research was to know the explants of banana weevil in apical and basal position with growth regulator 2,4-D with kinetin which are able to induce shoots from callus initials. On this research, the treatment tested for induction of shoots was growth regulators 2,4-D in the concentration 0 mg/L, 1 mg/L, 1,5 mg/L, 2 mg/L and 2,5 mg/L with growth regulators kinetin in the concentration 0 mg/L, 5 mg/L, 6 mg/L, 7 mg/L and 8 mg/L. The research was designed using Completely Randomized Design (CRD) two factorial with repetition of experiment. The results of research showed that the interaction between concentration of 2,5 mg/L 2,4-D with 5 mg/L kinetin was fastest (79 days) forming the shoot derived from explant basal part. The concentration of 2,5 mg/L 2,4-D with 5 mg/L kinetin produced the average number of shoots formed for 3.00 and average number of highest shoots for 1.50 cm. Abstract

The research or barangan banana (Musa acuminata L.) shoot induction by application of NAA and BAP based on the source of explans basal that had been carried out in the system Culture Laboratory of the University of North Sumatra. This study aims to discover the source of explants forming shoots by application of NAA and BAP and the number of shoots formed by the application of NAA and BAP based sources of banana explants. The study uses a completely randomized design with two factorial, i.e. the concentration factor NAA (0 mg/l, 1.5 mg/l, 2.0 mg/l, 2.5 mg/l, and 3.0 mg/l) and concentration factor BAP (0 mg/l, 4 mg/l, 5 mg/l, 6 mg/l, and 7 mg/l) with the source explants apical and basal parts. The best position that created the shoots explants contained in explant basal part by the number of shoot formed is 83. In the treatment of 1.5 mg/l NAA and 5 mg/l BAP average number of shoots most 3,75 and 2,30 cm tall shoots, the formation of the shoots is in the 80th day. The application of NAA and BAP on basal explants have a significant influence on the formation of the shoots.   Key words: Musa acuminata L., NAA, BAP,explants basal, shoot.

Abstract

HUBUNGAN ANTARA KUALITAS MAKANAN DENGAN KEPUASAN PELANGGAN PENGGUNA LAYANAN PESAN ANTAR OJEK ONLINE (STUDI KASUS DI KANTIN BUNDA, KARAWANG) ABSTRAK Penelitian ini bertujuan untuk mengetahui hubungan antara kualitas makanan dengan kepuasan pelanggan pengguna layanan ojek online. Jenis penelitian ini adalah penelitian survey untuk mencari hubungan kausal antara variabe melalui pengujian hipotesis. Penelitian ini dilakukan dengan jumlah responden sebanyak 67 orang pelanggan yang melakukan pembelian lebih dari 3 kali di Kantin Bunda Karawang. Penganmilan sampel menggunakan teknik sampel randoom dengan menyebarkan kuesioner menggunakan google ddocs. Metode analisis data yang digunakan dalam penelitian ini adalah menggunakan uji korelasi, sedangkan untuk menguji hipotesis menggunakan uji korelasi spearman. Berdasarkan penelitian taraf kesalahan yang digunakan adalah 0,05. Hasil model analisis korelasi menunjukan terdapat huungan antara kualitas makanan dengan kepuasan pelanggan pengguna layanan pesan antar ojek online dengan nilai rho hitung 0,57 lebih besar dari rho tabel 0,20. Berdasarkan hasil perhitungan karena rho hitung lebih besar dari rho tabel maka dapat disimpulkan bahwa terdapat hubungan antara kualitas makanan dengan kepuasan pelanggan pengguna layanan pean antar ojek online. Kata kunci : Kualitas Makanan, Kepuasan Pelanggan, Layanan Pesan Antar Online RELATIONSHIP BETWEEN FOOD QUALITY AND CUSTOMER SATISFACTION ON ONLINE OJEK MESSAGE SERVICE (CASE STUDY IN KANTIN BUNDA, KARAWANG) ABSTRACT This study aims to determine the relationship between food quality and customer satisfaction of users of online motorcycle taxi services. This type of research is survey research to find causal relationships between variables through hypothesis testing. This study conducted with a total of 67 respondents who made purchases more than three times at the Kunda Bunda Karawang. Sampling using the Random sample technique by distributing questionnaires using Google Docs. Data analysis method used in this study is to use a correlation test, while to test the hypothesis using the Spearman correlation test. Based on the research the error level used was 0.05. The results of the correlation analysis model show that there is a relationship between the quality of food and customer satisfaction of users of messaging services between online motorbike taxi drivers with assisted rho values ​​of 0.57 higher than rho table 0.20. Based on the results of the calculation because rho counts are more significant than rho table, it can be concluded that there is a relationship between food quality and customer satisfaction among service users among online motorbikes. Abstract

This research aimed to describe the factors that cause the low usage of long term contraception methods by women of child bearing age in Gunung Terang. The method used in this research was descriptive methods. The data were analyzed by one way table percentage. The population in this research were 911 women of child bearing age who use short term contraception methods and 90 women as samples. The samples were obtained by proportional random sampling technique. The results showed that (1) Most of women have good knowledge about contraception. (2) Counseling about family planning has been held very well. (3) Long term contraception in family planning service place has been available. (4) The cost of family planning service was affordable. (5) Almost of all women of child bearing ages husband was provide support for use contraception.Penelitian ini bertujuan untuk mendeskripsikan faktor-faktor penyebab rendahnya penggunaan metode kontrasepsi jangka panjang oleh wanita usia subur di Gunung Terang. Metode penelitian yang digunakan adalah deskriptif. Teknik analisis data menggunakan tabel persentase satu arah. Populasi pada penelitian ini adalah 911 wanita usia subur yang menggunakan metode kontrasepsi jangka pendek dengan sampel sebanyak 90 wanita. Sampel diperoleh dengan teknik proportional random sampling. Hasil penelitian menunjukkan (1) Sebagian besar wanita memiliki pengetahuan baik mengenai alat kontrasepsi. (2) Penyuluhan tentang keluarga berencana telah berlangsung dengan baik. (3) Pelayanan metode kontrasepsi jangka panjang di tempat pelayanan keluarga berencana telah tersedia. (4) Biaya pelayanan keluarga berencana cukup terjangkau. (5) Sebagian besar suami memberikan dukungan terhadap penggunaan alat kontrasepsi.Kata kunci: keluarga berencana, metode kontrasepsi jangka panjang, wanita usia subur

Abstract

Sains sebagai proses inkuiri menerapkan metode ilmiah dalam memecahkan permasalahan dan menemukan konsep baru. Dalam proses inkuiri atau penemuan diperlukan kemampuan khusus dalam berfikir maupun bertindak. Salah satu kemampuan yang perlu dikembangkan adalah kemampuan siswa untuk berfikir kreatif. Tujuan penelitian ini adalah mengkaji pengaruh project based learning (PjBL) berpendekatan kontekstual terhadap kreativitas peserta didik. Penelitian ini menggunakan post test control group design. Penelitian ini menggunakan cluster random sampling untuk menentukan dua kelompok penelitian. Kelompok eksperimen diberikan pembelajaran dengan model PjBL berpendekatan kontekstual dan kelompok kontrol diberikan pembelajaran secara konvensional. Variabel bebas dalam penelitian ini adalah macam metode pembelajaran dan variabel terikatnya adalah kreativitas peserta didik. Rata- rata skor kreativitas peserta didik pada kelompok eksperimen adalah 7,52 dan 6,78 untuk kelompok kontrol. Hasil uji-t menunjukkan terdapat perbedaan signifikans pada kreativitas peserta didik antara kelompok eksperimen dan kelompok kontrol. Penggunaan PjBL berpendekatan science edutainment memberikan pengaruh yang signifikans terhadap kreativitas peserta didik.

Abstract

An experiment on gossypol content hasbeen conducted on callus derived from Gossypium hirsutum L. subcultures 5, 6, and 7 after elicitation with Verticillium dahliae Kleb and Rhizoctonia solani Kuhn. Callus was cultured on solid Linsmaier and Skoog  (LS)  medium  supplemented  with 10 –5 M NAA and 10 –6 M 2,4-D, and subcultured for 5, 6, and 7 times. Callus was then elicited with 40  μg dry weight /ml homogenate of fungi derived from V. dahliae and R. solani, and harvested 0, 2, 4, and 6 days after elicitation. The gossypol was analyzed qualitatively and quantitatively using High Performance Liquid Chromatography (HPLC). The maximum gossypol content elicited with R. solani was obtained on callus subculture 5, which was harvested on 4 days after elicitation, i.e. 177,995 ±  0,248 μg/g dry weight,  whilst that with V. dahliae  was obtained on subculture 5, harvested four days after elicitation, i.e.108,021± 0,507 μg/g dry weight. The gossypol content of control callus on subculture 5, 6, and 7 was 37,885 ± 0,779 μg/g dry weight  23,170 ±  0,003 μg/g dry weight, 12,284 ± 0,221 μg/g dry weight, respectively. The subculture level, elicitor type, and harvesting time gave significant effect on gossypol content of G. hirsutum callus culture.

Abstract

The study of essential oils detection of patchouli leaves (Pogostemon cablin Benth) with combination of 2,4-Dichlorofenoxyacetate and coconut water which produced calli had been done at Departement of Food Crops and Horticulture, Tissue Culture Laboratory, North Sumatra and the Organic Chemistry Laboratory of the Faculty of Mathematics and Natural Sciences, University of Sumatera Utara. The aim of the study was to determine the concentration of the combination of 2.4-D and coconut water which added in MS media for detection essential oils in patchouli calli in vitro. The experiment used factorial Completely Randomized Design (CRD) with two factors. The first factor was coconut water with concentrations of 0, 10 and 20%. The second factor is 2.4-D with a concentrations of 0, 0.5 and 1 ppm.The results showed that the optimum combination of 2,4-D and coconut water was capable to induce calli initiation after 13 DAP (days after planting), 80% of calli production, green color, nonfriable, and calli weight of 0,28 gram. The TLC (Thin Layer Chromatography) result showed that the treatment of 0.5 ppm 2,4-D and 10% coconut water produced the same Rf value as Rf standard (patchouli oil) which was 0.87

Abstract

Based on observations implementation of learning computer and basic network indicated only limited by using print module media, with lecture methods and guided practice and have not used learning media with android technology. This research purpose to obtain validity of learning media android-based on computer and basic network subjects. Interactive learning media based on android is designed to improve students' understanding so as to improve learning outcomes. This research is a Research and Development (R and D) with 4-D method development procedures (Define, Design, Develop, Desseminate). Data analysis techniques used descriptive analysis techniques to describe the validity of interactive learning media android-based. The results obtained from this development research the validity of Android-based interactive learning media is declared valid on the media aspects and material aspects. Based on the result of this reserch concluded that the interactive learning media android- based was declared valid to be used as teaching materials in computer and basic network.
