Abstract

Abstract

Abstract

2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) is a chemical compound resulting from the burning effect that is very dangerous for humans, plants, or animals which can cause skin damage, immunotoxic, hepatotoxic, carcinogenic, teratogenic, effects on reproduction, growth processes, neurobehavioral, and endocrine effects that are specific In addition, this compound can also reduce embryo weight and increase embryo mortality. This study aimed to determine the teratogenic of TCDD administration on eye diameters and body weight of chicken embryo. In this study there were 2 treatment groups, i.e: 1) P1 as a normal group without the addition of TCDD, and 2) P2 as a group with the addition of TCDD of 5 ng / egg with dissolved in 5 µL corn oil. The treatment was carried out for 7 days. The measurement data of eye diameters and body weight of chicken embryo were analyzed by the unpaired sample T test. The analysis showed that the teratogenic of TCDD had a significant effect on eye diameters and body weight of chicken embryos, where the teratogenic of TCDD caused a decrease in eye diameters and body weight loss in chicken embryos.

Abstract

Mammary gland tumours are frequently reported in adult female mice. It broadly affects the productivity and breeding performance of mice in breeding centres. The tumours might occur spontaneously due to internal factors such as genetic mutation and hormonal disturbance, however, external causes such as Mouse Mammary Tumour Virus (MMTV) infection was also reported. This study aimed to prove that age and number of parturition associate with mammary gland tumour case rate in female mice in Malang Raya. Chi-square tests were employed to analyse the association between risk factors and mammary gland tumour case rate in this study. Histopathologic analysis described three subtypes of mammary gland tumours in mice which were adenoma, adenocarcinoma type A and adenocarcinoma type B. Chi-square tests results of tumour case rate toward age and number of parturitions were consecutively 0,004 and 0,0025 which indicated that age and number of parturitions were associated with mammary gland tumours case rate in mice in Malang Raya. The mapping of tumour case distribution visually showed a trend that higher disease proportions were spotted in higher altitude of sampling location. This might relate to the geographical breeding characteristics or temperature which needs further study to explore the association of spatial factors with the mammary tumour incidence in mice.

Abstract

Purple roselle has ingredients such as organic acids, anthocyanins, polysaccharides, and flavonoids. The antioxidant content in purple roselle has benefits in preventing cell damage due to free radicals. This study was conducted to discoverthe effect of supplementation yogurt fortified with a graded concentration of purple roselle extract on MDA levels and histology of the kidney organs of mice. Laboratory experimental study using 16 male Balb-C mice divided into four groups. The control (C) group was not given any treatment. The treatment groups 1,2, and 3 (T1, T2, T3) were given fortified purple roselle yogurt with concentrations of 5%, 10%, and 15%, respectively. The treatment was carried out for 14 days with a volume of 0.4 ml/day. Kidney organs were collected for histological analysis. Kidney malondialdehydelevels were assayed using the TBARS test and statistically analyzed with a 95% confidence level by one-way ANOVA analysis. Histology score analyzed using the Kruskal-Wallis test followed by the Mann- Whitney test. This study showedthat at concentrations of 10% and 15%, MDA levels of the mice kidneys reduce significantly compared to the control group. The kidney histology score results show some changes in treatment groups compares to control group. The average score decreasing from different treatment group, and the lowest score was group given yogurt fortified roselle extract 15%. The administration of fortified yogurt with purple roselle extract at a concentration of 15% can reduce MDA levels in the kidneys of mice, and the higher the concentration can reduce kidney damage in mice.

Abstract

Dioxins are highly toxic and hurt multiple organs and systems, including the reproductive organs. This study determined the supplementation of purple roselle (Hibiscus sabdariffa var. altissima) extract into yogurt to prevent testicular deterioration induced by 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). The fortification of purple roselle extract into yogurt (RY) was used as a supplement because bioactive components are known to be efficacious in preventing cell damage. Five groups of male Wistar rats were group as follows: 1) negative control (C), 2) TCDD (T), 3) TCDD and 0.5% RY (P1), 4) TCDD and 1% RY (P2), 5) TCDD and 1.5% RY (P3). All treatments were carried out orally for 12 days. Testicular Malondialdehyde (MDA) level and histopathological changes were described quantitatively by one-way ANOVA. The study's results showed that treatment groups P1 (0.5% RY) and P2 (1% RY) had significantly (P<0,05)  lower MDA levels than the T group, while group P3 had no significant decrease in MDA level. It proved that supplementation of purple roselle yogurt reduced the formation of MDA in testicular tissue. Histopathology changes show that all groups exposed to TCDD have depleted spermatogenic, Sertoli, and Leydig cells compared to the C group. Suggests that dioxin exposure can have adverse effects on male reproductive organs. This research concludes that supplementing yogurt fortified with purple roselle extract can prevent increased testicular MDA levels. The decrease in testicular cells due to TCDD toxicity was inevitable. However, antioxidant activity from roselle yogurt supplementation can prevent further damage in rat testicular cells.
