Abstract

Purple roselle has ingredients such as organic acids, anthocyanins, polysaccharides, and flavonoids. The antioxidant content in purple roselle has benefits in preventing cell damage due to free radicals. This study was conducted to discoverthe effect of supplementation yogurt fortified with a graded concentration of purple roselle extract on MDA levels and histology of the kidney organs of mice. Laboratory experimental study using 16 male Balb-C mice divided into four groups. The control (C) group was not given any treatment. The treatment groups 1,2, and 3 (T1, T2, T3) were given fortified purple roselle yogurt with concentrations of 5%, 10%, and 15%, respectively. The treatment was carried out for 14 days with a volume of 0.4 ml/day. Kidney organs were collected for histological analysis. Kidney malondialdehydelevels were assayed using the TBARS test and statistically analyzed with a 95% confidence level by one-way ANOVA analysis. Histology score analyzed using the Kruskal-Wallis test followed by the Mann- Whitney test. This study showedthat at concentrations of 10% and 15%, MDA levels of the mice kidneys reduce significantly compared to the control group. The kidney histology score results show some changes in treatment groups compares to control group. The average score decreasing from different treatment group, and the lowest score was group given yogurt fortified roselle extract 15%. The administration of fortified yogurt with purple roselle extract at a concentration of 15% can reduce MDA levels in the kidneys of mice, and the higher the concentration can reduce kidney damage in mice.

Abstract

Dioxins are highly toxic and hurt multiple organs and systems, including the reproductive organs. This study determined the supplementation of purple roselle (Hibiscus sabdariffa var. altissima) extract into yogurt to prevent testicular deterioration induced by 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). The fortification of purple roselle extract into yogurt (RY) was used as a supplement because bioactive components are known to be efficacious in preventing cell damage. Five groups of male Wistar rats were group as follows: 1) negative control (C), 2) TCDD (T), 3) TCDD and 0.5% RY (P1), 4) TCDD and 1% RY (P2), 5) TCDD and 1.5% RY (P3). All treatments were carried out orally for 12 days. Testicular Malondialdehyde (MDA) level and histopathological changes were described quantitatively by one-way ANOVA. The study's results showed that treatment groups P1 (0.5% RY) and P2 (1% RY) had significantly (P<0,05)  lower MDA levels than the T group, while group P3 had no significant decrease in MDA level. It proved that supplementation of purple roselle yogurt reduced the formation of MDA in testicular tissue. Histopathology changes show that all groups exposed to TCDD have depleted spermatogenic, Sertoli, and Leydig cells compared to the C group. Suggests that dioxin exposure can have adverse effects on male reproductive organs. This research concludes that supplementing yogurt fortified with purple roselle extract can prevent increased testicular MDA levels. The decrease in testicular cells due to TCDD toxicity was inevitable. However, antioxidant activity from roselle yogurt supplementation can prevent further damage in rat testicular cells. Abstract

The development of yoghurt variations is increasingly diverse nowadays. The addition of black rice bran flour into yoghurt processing can increase the functional value of yoghurt, including increasing taste, antioxidant content, and dietary fiber. This research was to determine the effect of storage time in goat's milk yoghurt with the addition of black rice bran flour at a cold temperature of 4c on acidity and alcohol levels. The research used with 5 treatments, 4 repetitions. The treatment includes storage periods  i.e., 0, 1, 2, 3 and 4 weeks. The acidity level will be calculated for each treatment group using the acid-base and alcohol titration method using the pycnometer method. The data obtained were analyzed using the one way ANOVA test followed by the Duncan Multiple Range Test. The results show that storage time has a very significant effect (p˂0.01) on acidity and alcohol levels. The highest acidity level was in week 2 (0.47%), then decreased continuously in weeks 3 and 4 of storage time. The alcohol content increased in line with storage time, reaching 0.0043% in week 4. If  we look at the acidity and alcohol levels, the recommendation for consuming goat's milk yoghurt with the addition of black rice bran flour is no more than 1 weeks of storage time.

Abstract

Kolitis merupakan sub tipe inflamatory bowel disease yang ditandai adanya segmen inflamasi dan ulserasi pada usus. Gejala khas yang ditimbulkan adalah diare berdarah. Ayam broiler sebagai hewan model kolitis dengan induksi Dextran Sodium Sulfate (DSS). Tujuan dari penelitian ini adalah mengetahui pengaruh DSS terhadap performa ayam broiler meliputi pertambahan bobot badan harian, FCR, dan indeks performa serta jumlah total BAL dalam sekum ayam broiler. Ayam berjumlah 32 ekor umur 15 hari dibagi menjadi delapan ekor per kelompok yaitu kontrol negatif (P0), dan kelompok perlakuan yang diinduksi DSS dengan konsentrasi 0,75%(P1); 0,5%(P2); 2,25%(P3) per hari selama enam hari dengan dicampurkan air minum. Bobot badan ayam ditimbang setiap tiga hari sekali dan pakan ditimbang setiap hari untuk perhitungan FCR. Ayam dieuthanasia umur 21 hari dan swab sekum dikultur pada media MRSA serta jumlah koloni dihitung dengan metode TPC. Analisis data menggunakan uji One Way ANOVA. Uji lanjutan post hoc tukey dengan tingkat kepercayaan 95%. Perhitungan Bakteri Asam Laktat menunjukkan adanya perbedaan signfikan (p<0,05) terhadap kontrol dan kelompok perlakuan mulai dari konsentrasi 0,75%. Induksi DSS dengan konsentrasi 2,25% berpengaruh terhadap PBBH secara signifikan (p<0,05), dan menurunkan indeks performa ayam broiler. Induksi DSS tidak berpengaruh signifikan (p>0,05) terhadap nilai FCR ayam broiler. Dapat disimpulkan bahwa induksi DSS pada berbagai tingkat dosis dapat menurunkan jumlah bakteri asam laktat dan induksi DSS konsentrasi 2,25% dapat menurunkan performa ayam broiler.

Abstract

Rodhamine B and Saccharin are often misused by food producers as food additive, which, if their use is not monitored, can produce free radicals that cause oxidative stress conditions in the body, leading to dangerous damage to the body. The addition of purple rosella in yoghurt can increase the functional value of the yoghurt, namely increasing the antioxidant content. The purpose of this study was to determine the benefits of giving purple rosella yoghurt in preventing damage to the duodenum and colon of (Rattus norvegicus) caused by Rodhamine B and Saccharin. This study used Completely Randomized Design (CRD) with 28 male R. novergicus, Wistar strain in aged 8-12 weeks. Group K was not given any treatment; R was given Rodhamine B, S was given Saccharin, KRS was given Rodhamine B and Saccharin, YR was given Rodhamine B and yoghurt, YS was given Saccharin and yogurt, YRS was given Rodhamine B, Saccharin and yogurt. The dose of Rodhamine B was 22.5 mg.kg-1, Saccharin was 157.77 mg.kg-1, and the volume of yoghurt was 1 mL.head-1. They were administered orally with a gastric tube for 14 days. The concentration of purple rosella extract added to yoghurt was 15% (v/v). Parameters were observed in histopathology of the duodenum and colon with the scoring method. Data analysis was done with the Kruskal-Wallis test followed by the Mann-Whitney test. The KRS group showed the heaviest damage, namely necrosis, villi erosion, and inflammatory cell infiltration of >75% of the total visual field in the duodenum and colon. The YR, YS, and YRS groups showed significant changes compared to the R, S, and KRS groups, namely necrosis, villous erosion, and inflammatory cell infiltration, which appeared to be 25-50% of the total visual field. However, this did not match the condition of group K, which appeared to be <25% of the total visual field. The provision of purple rosella yoghurt can prevent 50% damage to the duodenum and colon of Rattus novergicus due to Rodhamine B and Saccharin. Abstract

Chronic kidney disease (CKD) is a decrease or loss of the kidney's ability to excrete unused products, concentrate urine and convert electrolytes. A female Domestic short hair cat with clinical findings of Polyuria/polydipsia, decreased muscle mass, anorexia, dehydration, hypersalivation. Defecation was found a solid, dry, black-brownish yellow consistency (Score 2) and urine was dark yellow. The physical examination revealed a drastic decrease in body temperature and tachycardia. Hematological examination showed leukocytosis and hypochromic macrocytic anaemia. The result of blood smear tests showed the presence of neutrophils and other lymphocytes in large numbers. Blood chemistry analysis showed an increase BUN and creatinine (Azotemia) and also increase SGOT. Urinalysis examination revealed proteinuria, ketonuria, urobilinogenuria and hypersthenuria. Fecal examination revealed positive helminthiasis in the form of Toxocara cati eggs which was confirmed by the finding of adult worms in the duodenum during necropsy. The therapy given to the case cat included 50 ml/kgBW Ringer's lactate, 0,5 ml/kgBW Amoxicillin-Clavulanate, 2 mg/kgBW Ranitidine, 0,2 mg/kgBW Ipakitin® and 0,5 ml/kgBW Imboost®. A week after therapy, the cat's condition deteriorated further, with an undetectable temperature, seizures, critical condition and not being able to survive, so a necropsy was carried out. Macroscopic results of the kidneys during the necropsy procedure showed that the korteks of kidneys were pale with no change in size. The histophatology finding in the kidneys are necrosis and fibrosis tubulointerstitial. Microscopy finding in duodenum is inflammation and adult worm Toxocara cati in the lumen duodenum. The conclusion is this cat with diagnose Chronic kidney disease with histophatologycal finding is glomerulonephritis and duodenitis.

Abstract

Feline Leukemia Virus (FeLV) is one of the major causes of death in cats. A 4-year-old domestic cat weighing 2 kg presented with decreased appetite, weakness, sneezing, and nasal discharge. Physical examination revealed gingivitis accompanied by ulcers in the mouth and pale mucous membranes. Further diagnostic tests included hematology examination, blood chemistry examination, and the FeLV test kit. The results of hematology and blood chemistry examinations reported that the animal had leukocytosis, neutrophilia, thrombocytopenia, hypochromic macrocytic anemia, hyperproteinemia, hyperglobulinemia, decreased A/G ratio, hyperamylasemia, hyperbilirubinemia, and azotemia. Rapid test (kit) examination showed a positive result for FeLV. Based on the diagnostic results, the initial diagnosis was suspected Feline leukemia virus (FeLV) with an infausta prognosis. The treatment included antibiotics Amoxicillin at 18 mg/kgBW, Diphenhydramine HCl 1 mg/kgBW, Ornipural® 0.2 ml/kgBW, Kenalog® 1 mg/kgBW, Hematovit® dose 0.4 ml/kgBW, and supplements in the form of Vitamin C 0.5 ml/day and Fufang® 1 ml/kgBW. Despite the treatment, the cat’s condition did not improve and it succumbed on the sixth day of treatment.
