Abstract

Perkebunan jeruk di Desa Umbulrejo, Kecamatan Umbulsari, Kabupaten Jember menimbulkan permasalahan lingkungan. Jeruk yang telah dipanen umumnya akan disortir terlebih dahulu. Jeruk yang tidak masuk ke dalam kriteria akan dibuang dan dibiarkan begitu saja menjadi limbah. Oleh karena itu, kegiatan ini bertujuan untuk menyadarkan masyarakat akan pengelolaan limbah jeruk yang benar. Limbah jeruk, khususnya pada kulit jeruk dapat dimanfaatkan menjadi minyak atsiri dan lilin aromaterapi. Limbah kulit jeruk diekstraksi untuk mendapatkan minyaknya menggunakan metode ekstraksi padat-cair. Diharapkan kedepannya seluruh masyarakat di Desa Umbulrejo dapat memanfaatkan limbah jeruk agar menjadi produk yang memiliki nilai ekonomis yang lebih tinggi. Hasil dari program ini adalah meningkatnya pengetahuan masyarakat tentang pemanfaatan limbah kulit jeruk menjadi minyak atsiri dan lilin aromaterapi

Abstract

Indonesia has been recognized for its rich natural ingredients such as spices, herbs, and extracts for decades. Furthermore, these components also have been used as an herbal medicine for a long time ago. Meanwhile, the apparent capability comparison of several components on antimicrobial activity has not been updated yet. This study was conducting the comparison of antimicrobial activity for several materials such as noni (Morinda citrifolia), garlic (Allium sativum), celery (Apium graveolens), galangal (Alpinia galangal), ginger (Zingiber officinale), yellow turmeric (Curcuma longa), lime (Citrus aurantifolia), papaya (Carica papaya) leaf, betel (Piper betle) leaf, and cutcherry (Kaempferia galangal) using agar dilution method. Two types of bacteria are used for the test, namely gram-negative bacteria and gram-positive bacteria, with a total of seven bacteria. The media used were TSA (Trypticase Soy Agar) media for gram-negative bacteria and MRSA (Methicillin-Resistant Staphylococcus Aureus) media for gram-positive bacteria. This study had been conducted by using a fast screening method, which referred to a semi-qualitative method. Several components, such as noni, lime, and betel leaf, showed a significant result of antimicrobial activity. Otherwise, other compounds, surprisingly, could not suppress bacterial growth.

Abstract

Quantitative structure-activity relationship (QSAR) has been studied for ferulic acid derivatives to determine the QSAR model able to predict anticancer. As the subject of this research was a set of experimentally calculated IC50 value data of 6 ferulic acid derivatives against cervical cancer cells (HELA). QSAR analysis was based on multilinear regression calculation on fitting subset using log (1/IC50) as the dependent variable, and dipole moment, partition coefficient in the n-octanol/water, and atomic net charges of the aromatic carbons as independent variables. The values of the descriptors were obtained from semiempirical PM3 quantum mechanic calculation. The relationship between log (1/IC50) and the descriptors was described by the result in the QSAR model. The QSAR model for ferulic acid derivatives against HELA cell lineswas developed with the statistical parameters of R=0.998; R2=0.999; SE=0.00857; and F=394. The calculated log (1/IC50) using QSAR Hansch Model for ferulic acid derivatives have excellent agreement with experimental data of Log (1/IC50).

Abstract

Sargassum sp., part of brown algae, is traditionally used as animal feed. Nonetheless, Sargassum contains phenolic compounds that promise to be the raw material of natural antioxidants. The work is to extract phenolic compounds from Sargassum sp. in ethanol and to investigate the effect of the process on phenol yields. Several dried Sargassum sp. are milled in a certain size and extraction using ultrasound assisted extraction with ethanol solvent. Process variables are the extraction time (30-50 minutes) and the Ratio of Sargassum sp. to ethanol which is 5:100-15:100 (b/v), and power of 170-190 watts. Phenol compounds are quantified using the error acid method. The DPPH method is performed to check the antioxidant activity. To investigate the influence of the process, surface response methods based on central composite designs are applied in this work. 153.334 mgGAE/g in 30-minute extraction conditions, 170 watts of power, and a ratio between masses and solvents of 0.05. The antioxidant activity (IC50) of Sargassum sp. extract is 87.57 ppm.

Abstract

Labu siam (Sechium edule) merupakan tumbuhan dari famili Cucurbitaceae mengandung beberapa vitamin, seperti vitamin A, B dan C sehingga seringkali digunakan untuk mengobati penyakit. Berdasarkan analisis fitokimia ekstrak labu siam menunjukkan adanya senyawa bioaktif berupa fenolik. Senyawa fenolik merupakan metabolit sekunder dan dimanfaatkan sebagai bahan kosmetik dan obat-obatan karena mengurangi stress oksidatif. Penelitian ini bertujuan untuk mengetahui kandungan total fenolik dan aktivitas antioksidan ekstrak labu siam (Sechium edule) dengan metode spektrofotometri UV-Vis. Ekstrak labu siam diperoleh dengan metode Ultrasonic Assisted-Extraction (UAE) dengan pelarut etanol 96%. Variabel ekstraksi yang digunakan adalah waktu (15, 20, dan 25 menit), amplitudo (45, 65, dan 85 %), serta rasio pelarut (1:5, 1:10, dan 1:15). Penentuan kandungan total fenolik didasarkan pada metode Folin-Ciocalteau serta aktivitas antioksidan menggunakan DPPH. Penentuan parameter ekstraksi labu siam terhadap kandungan total fenolik ditentukan dengan metode Response Surface Methodology dengan tipe Box-Behnken. Kondisi optimal yang diperoleh untuk ekstraksi labu siam sebesar 1,327 mg GAE/gr sampel dan aktivitas antioksidan sebesar 29,45% pada kondisi waktu 20 menit, amplitudo 65%, dan rasio pelarut 1:10.

Abstract

Coffea sp. is one of the largest plantation commodities in Indonesia, especially coffee leaves containing phenolic compounds. This research was conducted to extract phenolic compounds using the Ultrasound-Assisted Extraction method with ethanol solvent which aims to determine the optimum operating conditions of extraction as well as the influence of variations in amplitude, time, and ratio of solvents produced. The experimental design was carried out using Design Expert 13 software with the response surface method box-Behnken design. The research variables used were amplitude variations (50%, 60%, and 70%), time (10, 20, and 30 minutes), and solvent ratios (0.1; 0.15; and 0.2 g/mL).Based on our study, these parameters affect the total phenolic content. The model equation for the total phenolic content of coffee leaves obtained is Y = 0.1349 – 0.0016 A – 0.0505 B + 0.0010 C + 0.0018 AB + 0.0043 AC – 0.0018 BC – 0.0004 A2 + 0.0178 B2 – 0.0014 C2 (R2 = 0.9758) with the optimum total phenolic content located in the 17th running of 0.209 mg GAE/g under conditions of 20 minutes, the ratio of material to solvent is 0.2 g/mL, and an amplitude of 50%.

Abstract

Anthocyanins are polyphenolic compounds whose existence is very abundant in nature with diversity in various types of plants. The purpose of this study was to optimize the anthocyanin extraction process from cocoa shell waste using the MAE method in order to obtain the optimum particle size, ingredient/solvent ratio, and extraction time to produce high anthocyanin concentrations. The optimized extraction process variables include the size of the particle (1,05; 1,25; 1,49 and 2,5 x 10-4 m), the ratio of substrate/solvent (4,5; 12,5; 62,5; 79,5 x 10-3 g/mL), time of extraction (2, 4, 10, 14 minutes) and microwave power (100; 275; 450; 625 watts). The Design Expert vs11 program with the Box-Behnken Design Response Surface Methodology (RSM) was used in the research and the selection of process conditions was carried out from a combination of factors that resulted in an optimal response. The relationship between variables on the modeled anthocyanin concentration response: Y=0,000486-9,98637E-07A+0,026734B0,000041C-7,58240E-07D-0,000102AB+2,48606E-07AC+2,62878E09AD-0,000539BC+0,000012BD+9,71853E08CD (A: particle size; B: cocoa shell: solvent ratio; C: extraction time; D: microwave power). The optimal response value for anthocyanin concentration was 11,85.10-4 M and the conditions of the extraction process are the particle size in the extraction process was 0,105 mm, the ratio of cocoa peel mass/ethanol was 0,03125 g/mL, the extraction time was 2 minutes, and the microwave power was 100 W.

Abstract

This study aims to utilize sweet lime leaves which are only waste that has no value, where sweet lime leaves are one of the ingredients that contain essential oils. Apart from the skin, it turns out that this sweet lime leaf can also be extracted using the Microwave Hydrodistillation (MHD) method. This study uses three variables namely; the F/S ratio used was 0.375 g/mL; 0.5g/mL; 0.65 g/mL, power 150 watts, 300 watts, and 450 watts, and time 30, 60, and 90 minutes. The extraction method is carried out using fresh ingredients with a solvent volume of 200 ml. The test carried out is the GC-MS test. The results of the test method and the purpose of this study were to determine the limonene content in the extraction of sweet lime leaves using the MHD method with variations in ratio, power, and time. The optimum yield of essential oils is at a power of 300 W, 90 minutes, and a ratio of 0.625 with a yield of 0.373%. The biggest components produced in the MHD method of extracting citrus leaves were Germacrene D 29.51%, Alpha-Copaene 18.47%, CIS-CAROPHYLNE 15.42%, 9-Eicosene (E) 8.58% and Limonene 5.51%.

Abstract

Turmeric leaves (Curcuma longa L.) contain phytochemical compounds that can be utilized as natural food preservatives or bioformalin due to its antibacterial properties. The extraction method used in this study is Ultrasound-Assisted Extraction (UAE), with the aim to determine the effect of variable sample-solvent ratio, time, and particle size on total flavonoid, tannin, and alkaloid compounds, as well as to determine the optimal shelf life of fresh tuna. This study used sample-solvent ratio variables of 1:10, 1:15, and 1:20; time variables of 10, 20, and 30 minutes; and particle size variables of 60, 80, and 100 mesh. The highest total flavonoid compound was obtained in the variable sample-solvent ratio of 1:20, particle size of 100 mesh, and time of 20 minutes at 98,076 mg/L. The highest total tannin compound was obtained in the variable sample-solvent ratio of 1:15, particle size of 60 mesh, and time of 10 minutes at 41,697 mg/L. The highest total alkaloid compound was obtained in the variable sample-solvent ratio of 1:10, particle size of 100 mesh, and time of 20 minutes at 10,092 mg/L. The optimum curing time for tuna is 36 hours at room temperature with variable sample-solvent ratio, time, and particle size of 1:20 g/mL, 20 minutes, and 100 mesh with 20% concentration. The running has the highest flavonoid compounds, so it can be concluded that flavonoid compounds have a major effect on the preservation process of tuna.

Abstract

Using cooking oil repeatedly will have an impact on health. Cooking oil repeatedly changes color to a darker color, produces an unpleasant odor, and has high levels of fatty acids. Cooking oil experiences changes due to the chemical properties of cooking oil, such as a reaction where the color changes to brown when cooking oil is used at high temperatures by reacting with air. Therefore, it is necessary to purify used cooking oil to be used as a material that is more beneficial to society. The utilization of cocoa shell waste into carbon that has been processed is used for the adsorption process of purification of used cooking oil. The process of purifying used cooking oil begins with making activated charcoal, chemical activation of activated charcoal with hydrochloric acid (HCl), assembling appropriate technology for oil purification, adsorption process of used cooking oil which goes through several stages of purification, namely zeolite stone, cotton, cocoa charcoal, cotton and filter cloth. , and the process of using purified used cooking oil into aromatherapy candles. The materials used in making candles are paraffin and used cooking oil in a 1:1 ratio. Candles are packaged attractively to provide consumers and add selling value. This activity attracted the enthusiasm of the community to utilize used cooking oil into aroma therapy candles. In addition, the Panti Village Tourism Awareness Group is interested in utilizing used cooking oil to make souvenir aroma therapy candles at Panti village tourist attractions.It is hoped that using used cooking oil that has been purified into aromatherapy candles will become a source of income for the people of Kedaton Plantation, reducing waste disposal so that the environment will be better protected.
