Abstract

The efficacy of povidone iodine as a surface disinfectant was assessed in white shrimp (Litopenaeus vannamei) eggs and larvae. Eggs and nauplii were exposed to povidone iodine 20 ppm for 20 seconds. Culture-based method and Terminal Restriction Fragment Length Polymorphism (T-RFLP) were used to monitor the total number of bacteria and diversity of the bacterial community associated with shrimp eggs and larva at each developmental stage. Povidone iodine reduced the total culturable bacteria, especially Vibrio, on eggs and nauplii, as well as the total bacteria in the whole community as estimated by T-RFLP results. Povidone iodine also reduced the diversity of bacterial community and altered the evenness of phylotypes distribution suggesting that the use of povidone iodine as a surface disinfectant in shrimp aquaculture should be reconsidered.

Abstract

Abstract

This research aimed at analyzing the phytase from fungal isolates SB1, SB2, BS, and WF produced in cornstarch with glucose medium (CS+Glu) as carbon sources and Potatoes dextrose broth (PDB). The activity of phytase from isolates SB1, SB2, BS, and WF produced in CS+Glu medium was 2.97 UmL-1, 2.87 UmL-1, 3.18 UmL-1, and 4.37 UmL-1, respectively, while the activity of phytases was 2.07 UmL-1, 2.17 UmL-1, 2.22 UmL-1, and 2.78 UmL-1 respectively in PDB medium. The optimal temperature of SB1 and WF phytase was 40°C, while SB2 and BS were 50°C and 60°C, respectively. The optimal pH of SB1 and WF phytase was 5.0, while SB2 and BS phytase were 6.0, and 4.0 respectively. 18S rRNA gene analysis revealed that SB1 was 99% identical to Aspergillus niger ANTS (KY825168.1), SB2, BS, and WF were 99% identical to A. niger Moriga leaf (MG889596.1). Multiple sequences and phylogenetic analysis of phytase gene showed that phyA_SB1 and phyA_SB2 were 98% homology with A. ficuum (AAB26466), 97% with A. niger (ADP05107) while phyA_WF was 99% with A. ochraceoroseus (PLB29348), 98% with A. niger (ADP05105). The deduced proteins contain conserved motifs RHGARYPTD at N-terminal while lacking HD motif at C-terminal. These phytases were in the same cluster with Aspergillus sp. phytase A indicating that they belong to Histidine Acid Phosphatases (HAP) family.

Abstract

The avocado peels and seeds are potentially produced into vegetable oil through the soxhletation method. The soxhlet extracted oil characteristics are influenced by extraction time. The proper extraction time generated the oil standard characteristics. This research aimed to determine the effect of extraction time on yield, physicochemical characteristics, and antioxidant potential of avocado waste oil. The research was conducted through the extraction and characterization of avocado waste oil. This study used a Completely Randomized Design with one factor (extraction time: 4, 5, and 6 hours). An increase in extraction time caused an increase in yield and specific gravity, but the hydrolytic-oxidative stability and antioxidant potential were decreased. The 4 hours of extraction time was selected as the best treatment. The characteristics of avocado peel oil extracted for 4 hours were 6.25% of yield, 0.961 g/mL of specific gravity, 0.27% of free fatty acids (FFA), 0.44 mgKOH/g of acid value (AV), 0.98 mEq/kg of peroxide value (PV), 4.63 mEq/kg of p-Anisidine value (PAV), 8.86 mEq/kg of total oxidation (TOTOX), 68.786% of radical scavenging activity, and 523.96 mgTr/g of reducing power. The characteristics of avocado seed oil extracted for 4 hours were 4.59% of yield, 0.973 g/mL of specific gravity, 0.23% of FFA, 0.33 mEq/kg of AV, 0.48 mEq/kg of PV, 3.77 mEq/kg of PAV, 5.05 mEq/kg of TOTOX, 53.95% of radical scavenging activity, and 438.05 mgTr/g of reducing power. Altogether, avocado waste oil characteristics in each extraction time met the vegetable oil requirements and potentially developed in food industries.

Abstract

Abstract. Azolla microphylla is included Azollaceae family that very rich of protein, essential amino acid, vitamin and mineral. Azolla plant can be a potential source of nutrient for fish. The study was aimed to determine the utilization of potential of A. microphylla as a feed for Anguilla bicolor bicolor. This study was used a completely randomized design (CRD), with five treatments and three replications. About 15 of A. bicolor bicolor with an average body length of 20-25 cm and a weight of ± 10 g were divided into five groups, into each aquarium containing three fish and they have been given treatment test by substitution of A. microphylla (0%, 10%, 20%, 30%, 40%) for 30 days. The parameters of this study were absolute weight gain, specific growth rate, feed convertion ratio and survival rate of A. bicolor. The ANOVA analysis were continued by the Tukey test in a confidence level of 95%, has shown difference results significantly (P 0.05) for control treatment (0% A. microphylla) which were absolute weight gain: 0,13±0,07 g, SGR: 0,55±0,11(%/day), FCR: 13,25±2,62 and SR: 100%. All treatment that substituted by A. microphylla haven’t shown positive results. Based results of the research, concentrations of A. microphylla which has been substituted (10%, 20%, 30%, 40%) for 30 days, it hasn’t shown potential feed for A. bicolor bicolorKeywords: Azolla microphylla, Anguilla bicolor bicolor, feed, substitution, potentialAbstrak. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL), dengan lima perlakuan dan tiga kali ulangan. Sekitar 15 ekor A. bicolor bicolor dengan rata-rata panjang tubuh 20-25 cm dan berat ± 10 g dibagi menjadi lima kelompok, ke dalam akuarium masing-masing berisi tiga ikan untuk diberi pakan uji dengan substitusi A. microphylla (0%, 10%, 20%, 30%, 40%) selama 30 hari. Parameter penelitian ini adalah pertambahan berat mutlak, laju pertumbuhan spesifik, rasio konversi pakan dan tingkat kelangsungan hidup A. bicolor bicolor. Hasil analisis ANOVA dilanjutkan uji Tukey dengan taraf kepercayaan 95%, menunjukkan hasil beda nyata (P0,05) pada perlakuan kontrol (0% A. microphylla) dengan pertambahan berat mutlak: 0,13±0,07 g, SGR: 0,55±0,11(%/hari), FCR: 13,25±2,62 dan SR: 100%. Seluruh perlakuan dengan substitusi A. microphylla belum menunjukkan hasil positif untuk ikan sidat. Abstract

Indole acetic acid (IAA) is an auxin hormone that can regulate plant growth and development. Bacteria produce IAA through L-tryptophan metabolism. The purpose of this study was to determine the ability of endophytic bacteria isolated from Daun Dewa plants to produce IAA to increase the germination and growth of tomato seedlings. Isolation of endophytic bacteria was carried out by sterilizing plant surfaces. The production of IAA is done by adding L-tryptophan precursors. The results showed that there were six endophytic bacteria, and three isolates of them have the potential to produce IAA, namely: AD1.1, UD1.1, and UD2.1. The highest IAA concentration was produced by isolate AD1.1. Growth in sprouts is significantly influenced by the concentration of IAA; the higher the concentration of IAA, the longer the shoots and the roots. Based on the 16S rRNA gene sequence, these isolates belong to the genus Klebsiella.

Abstract

Background: Antiapoptotic Bcl-2 proteins are overexpressed in cancer cells, leading to inhibition of apoptosis and the development of therapeutic resistance. Targeting only one type of antiapoptotic protein may have limited efficacy in cancer therapy. Anticancer drugs capable of inhibiting Bcl-2, Bcl-XL and Mcl-1 simultaneously are necessary to be explored. Penicillium sp. produces various bioactive compounds with anticancer, antibacterial, and antiviral activities. The aim of this research was to determine the best bioactive compound candidates for inhibiting Bcl-2, Bcl-XL, and Mcl-1 proteins.Materials and methods: Molecular docking analysis was conducted to estimate the binding affinity of Penicillium sp. bioactive compounds with Bcl-2, Bcl-XL, and Mcl-1 proteins. Compounds with the lowest binding energies were visualized using PyMol and Ligplot+ and further subjected to drug-likeness testing based on Lipinski's rule of five.Results: Bioactive compounds with the highest binding affinities were verruculogen and wortmannin. Wortmannin complied with Lipinski's rule of five. Meanwhile, verruculogen violated one out of the five rules by having a molecular weight >500 Da. Both compounds could be used as oral drugs.Conclusion: Verruculogen and wortmannin from Penicillium sp. show significant potential as oral anticancer drug candidates.Keywords: Bcl-2, Bcl-XL, Mcl-1, Penicillium sp., in silico

Abstract

Their nanometer size, broad spectrum, and antibacterial mechanism give silver nanoparticles (NPAg) the potential to be used to inhibit the growth and spread of methicillin‐resistant Staphylococcus aureus (MSRA) in medical devices. Synthesis of AgNPs from Stachytarpheta jamaicensis leaf extract is considered more environmentally friendly and has low production costs. The objective of this study was to investigate the properties and antibacterial potential of AgNPs by utilizing S. jamaicensis leaf extract at concentrations of 0.5%, 1.0%, and 1.5% in a 1 mM AgNO3 precursor. Nanoparticle characterization was performed on the AgNP supernatant obtained by centrifuging the synthesis solution at 100 rpm for 5 min. Characterization of the NPAg size was confirmed by surface plasmon resonance (SPR) analysis in UV‐Vis spectrophotometer, while the size distribution was measured by a particle size analyzer. Surface morphology was performed using scanning electron microscopy (SEM), and antibacterial activity was evaluated by the disc diffusion method. The results showed that AgNPs had the best nanoparticle characteristics in an extract concentration of 0.5%, the synthesis indicated by SPR at a wavelength of 434 nm and an average size of 80.67 nm. SEM analysis showed the formation of clusters at variations of 1.0% and 1.5%. The antibacterial activity of AgNPs against MRSA was the highest at 0.5%, with an inhibition zone diameter of 0.77 mm. The higher concentration of S. jamaicencis leaf extract increases the risk of cluster formation, which enlarges the AgNPs, while antibacterial activity was reduced.
