Abstract

The Polymerase Chain Reaction (PCR) technique is extensively employed in molecular biology to precisely detect Mycobacterium tuberculosis. Prior to conducting PCR, extracting of high-quality genomic Deoxyribonucleic Acid (DNA) is crucial to ensure accurate and reliable results. The primary objective of this study is to conduct a comparative analysis of the purity and concentration of M. tuberculosis DNA acquired through the utilization of the boiling method and the spin column extraction methods. A descriptive comparative research design was employed, utilizing a sample of 16 sputum specimens that had previously been confirmed as positive for M. tuberculosis through Acid-Fast Bacteria (AFB) examination and Molecular Rapid Test (MRT). The extraction of DNA was carried out using the boiling method and the spin column method. Subsequently, the concentration and purity of the extracted DNA were assessed using the NanoDrop Spectrophotometer, and the results were compared. The obtained yield of M. tuberculosis DNA isolates through the boiling method ranged from 9.6 ng/µL to 1258.7 ng/µL, with an average purity value of 1.23. Conversely, for the spin column method, the concentration of M. tuberculosis DNA isolates ranged from 8.7 ng/µL to 207.8 ng/µL, with an average purity value of 1.83. In conclusion, there is a significant difference between the purity and concentration of M. tuberculosis DNA extraction results using the boiling method and spin column methods.

Abstract

The legacy of the Majapahit Kingdom is a very important legacy for the development of culture and civilization in Indonesia. The founding of this kingdom is estimated to be in the thirteenth to sixteenth centuries, until it was known as the largest and most powerful kingdom in the archipelago. This article examines various aspects of Majapahit heritage, including architecture, art, literature, and government systems. Through analysis of artifacts such as temple remains, inscriptions and literary works, as well as their influence on local society and culture, this article was created to provide deeper knowledge about Majapahit's contribution to shaping the identity of the Indonesian nation. Apart from that, discussing the challenges of preserving these heritages in the modern era is also an important focus, considering the high historical value and tourism potential that can be developed. Thus, it is hoped that this article can increase awareness of the importance of preserving Majapahit's historical heritage for future generations.

Abstract

Artikel ini membahas implementasi toleransi antar agama dalam pembangunan infrastruktur di Desa Gajah Mati, Kabupaten Bengkulu Tengah, dan menilai dampaknya terhadap kerukunan sosial dan pembangunan berkelanjutan. Penelitian ini menggunakan pendekatan studi kasus untuk mengeksplorasi bagaimana infrastruktur yang inklusif dapat mempengaruhi hubungan antar kelompok agama dan mendukung pembangunan desa. Melalui wawancara mendalam, diskusi kelompok terarah, dan observasi langsung, penelitian ini mengidentifikasi peran penting tokoh agama dan pemerintah desa dalam perencanaan dan pelaksanaan infrastruktur yang merangkul keberagaman agama. Hasil penelitian menunjukkan bahwa infrastruktur yang dirancang dengan prinsip-prinsip toleransi berkontribusi signifikan terhadap pengurangan konflik sosial, peningkatan interaksi positif antar kelompok, dan pengembangan potensi sosial serta ekonomi desa. Namun, tantangan seperti resistensi dari beberapa kelompok, keterbatasan anggaran, dan kurangnya komunikasi efektif juga diidentifikasi. Implikasi jangka panjang dari hasil penelitian ini menunjukkan bahwa model keberhasilan Desa Gajah Mati dapat menjadi contoh bagi desa lain dalam mengintegrasikan nilai-nilai toleransi dalam pembangunan, dengan potensi untuk memperkuat persatuan dan kesatuan di masyarakat multikultural.

Abstract

This study aims to address global issues in technology that are challenges in social studies learning in order to improve students' understanding or appeal and skills in social studies learning. The method used is a literature study by reviewing relevant literature on inquiry-based learning models, problem-based learning, expository, and contextual. The results of the study indicate that the application of active learning strategies, such as inquiry and problem-based learning, can encourage students to think critically, creatively, and improve data analysis and communication skills. In addition, social studies learning that is integrated with real experiences is considered effective in shaping students' social attitudes and empathy. This study recommends that teachers optimize variations in learning strategies according to student characteristics and materials, in order to achieve social studies education goals holistically.

Abstract

This research examines how the use of social media platforms, especially TikTok and Twitter, can influence Generation Z's interest in history. Generation Z, accustomed to digital technology, is often less interested in historical subjects that are considered irrelevant. However, by adapting existing content formats on TikTok and Twitter that are more dynamic and easily accessible to Gen-Z, history can be presented in a more interesting way. TikTok, with its short videos, provides an opportunity to present historical facts visually and creatively, while Twitter allows the exchange of ideas and discussions based on trending historical topics. This research method uses a qualitative approach with content analysis from various accounts that discuss the history of the two platforms. The findings of this research show that with a more relaxed and interactive approach, Generation Z's interest in history can increase, especially if the content presented is able to connect historical events with contemporary issues that are relevant to them. Therefore, TikTok and Twitter not only have the potential to convey historical information, but also to create more interesting learning, and are appropriate to the digital lifestyle of Generation Z
