Abstract

Oxidative stress and inflammatory reactions are the pathological mechanisms for most degenerative diseases. The black cumin seed oil (BCSO) contains compounds that can act as antioxidants and immunomodulators. Consuming BCSO is thought to improve antioxidant and immunomodulatory parameters in obese people. This study investigated the effect of BCSO consumption on antioxidant and immunomodulatory activity in healthy volunteers. We conducted a quasi-experimental study on 12 healthy volunteers in Yogyakarta, Indonesia. We asked the volunteers to consume BCSO for twenty days. We measured blood pressure, body mass index (BMI), malondialdehyde (MDA) level, Serum glutamic oxaloacetic transaminase (SGOT), serum glutamic pyruvate transaminase (SGPT) activity, CD4Th, and IFN-γ expression before and after consuming BCSO. We carried out the average difference test of the parameters before and after consumption of BCSO by dependent t-test. The results showed that 3x1 BCSO preparation for 20 days reduced MDA levels and increased CDTh and IFN-γ. Consuming BCSO for 20 days potentially improve the adaptive cellular immune response parameters.

Abstract

Mefenamic acid is an analgesic non-steroidal anti-inflammatory drug (NSAID) that is categorized as Biopharmaceutical Classification System (BCS) class II drug. Cocrystallization of mefenamic acid (MFA) with nicotinamide (NIC) has been reported to enhance the solubility of MFA for about 2.56 times higher than pure MFA. As a part of MFA-NIC development in pharmaceutical dosage form, we reported the improvement of mechanical properties of MFA-NIC cocrystal based on the presence of slip planes in crystal structure. The research was initially by synthesizing and characterizing the cocrystal by using X-ray diffraction analysis, and was followed by the investigation of the mechanical properties of MFA and its cocrystal, i.e. tensile strength and deformation parameter. The correlation of crystal structure with mechanical properties was determined by crystal packing motif and slip plane analysis from single crystal data by using Mercury 3.6. The tensile strength of MFA was acceptance at 20.0 MPa, and dropped at more than 25.0 MPa. A better tableting performance was observed in MFA-NIC cocrystal which had the tensile strength 1.5 times higher than MFA at 12.5 MPa and gradually increased with an increasing pressure up to 2.5 times at 22.5 MPa. The flat slip plane of MFA-NIC cocrystal was predicted as the key factor in the improvement of its mechanical properties, compared to zigzag slip plane of MFA. In conclusion, MFA-NIC cocrystal had the better mechanical properties than pure MFA due to its flat slip plane and it becomes potential to be explored in further development of solid dosage form.

Abstract

Elevated oxidative stress and glycation give to the enhancement of degenerative condition such as diabetes and heart disease. Herbal extracts such as Apium graveolens, Centella asiatica, and Orthosiphon stamineus possess well-documented antioxidant and anti-glycation properties. However, their therapeutic effectiveness is measured by poor bioavailability, highlighting the need for innovative formulations. Nanotechnology presents a indicative of success approach by improving the solubility and distribution of active compounds. This study developed a nanopolymeric formulation by combining extracts of Apium graveolens, Centella asiatica, and Orthosiphon stamineus utilizing the ionic gelation method by chitosan, NaTPP, and Tween 80. The nanoparticles were characterized based on particle size, PDI, zeta capability, TEM, and FTIR. In vitro assays utilizing peripheral blood mononuclear cells (PBMCs) were conducted to analyze antioxidant activity through Nrf2 and GST expression, and anti-glycation capability by measuring AGEs. The nanopolyherbal particles had an average size of 186 ± 46.81 nm, by a PDI of 0.434 ± 0.04 and a zeta capability of -29.69 ± 1.54 mV. Both 1% and 3% nanopolyherbal treatments increased Nrf2 stages (66.77 ± 7.65 and 63.13 ± 2.75, respectively) and GST stages (87.97 ± 49.48 and 73.57 ± 4.61) compared to the control group (Nrf2: 54.1 ± 1.0; GST: 68.93 ± 2.28). The nanopolyherbal formulation also significantly decreased AGE itemion (1%: 1075.67 ± 107.51; 3%: 813.33 ± 117.05) compared to the control (1497.33 ± 161.58). These outcomes reveal that the nanopolyherbal formulation enhances antioxidant activity and inhibits glycation in PBMCs, suggesting its capability for managing oxidative stress-related condition.
