Abstract

Research has been conducted on the study of the interaction between cyclic peptide ADTC2(Ac-CADTPPC-NH2) and E-cadherin protein using the molecular docking method. The aim of this study is to determine the position of the binding site and the energy of interaction between the ADTC2 peptide and the EC1-EC2 domain of E-kadherin. This research was divided into two parts, (1) Preliminary test using molecular dynamics (DM) method with Gromacs v4.5.4 software, (2) interaction of the peptide ADTC2 with EC1-EC2 using the molecular docking method (MD) with Autodock 4.2 software. Docking was performed with the blind dock method on EC1 and EC2 position. In the second step, the gridbox position was reduced based on the binding activity between E-cadherin and peptides. The strongest interaction and Van der Walls bonds were obtained in boxes B, C and D. The results showed that the ADTC2 peptide had a biological activity to inhibit the interaction of E-cadherin...E-cadherin by forming a complex with the EC1-EC2 domain. This inhibition occured by forming two binding sites in the EC1 domain (interaction energies are -23.309 kJ / mol and -26.234 kJ / mol, respectively) and one binding site in the EC2 domain (interaction energies are -22.677 kJ / mol). Based on preliminary tests, it can be proven that the native structure of ADTC2 is cyclic with optimization energy of -52504.78 kJ/mol and very stable from the beginning to the end of DM with an RMSD was <2 Å.

Abstract

Diabetes is a chronic disease characterized by high body sugar levels, but the body does not utilize them. Based on data from 2023, 14% of Indonesia's population has diabetes. The spread of diabetes also penetrates rural areas, posing a threat to rural areas and threatening the health of rural communities, and Saentis Village is no exception. The program "Healthy Movement Together: Blood Sugar Checking to Support the 9 Pillars of Health Transformation" is a community service initiative that aims to increase public health awareness. Some people in Saentis Village have a history of diabetes. This activity aims to provide education and conduct initial screening for diabetes. The service method is carried out by collaborating between medical personnel, local government, and the community. The activity began by providing education about the dangers and early symptoms of diabetes and continued by conducting an initial diabetes screening by checking blood sugar. Ninety-six people attended this activity. Based on the results of the examination, it was found that as many as 17 people had high blood sugar levels, and as many as 14 people experienced hyperglycemia. Based on the results of the screening that has been carried out, it is known that there are people who are not aware of the importance of maintaining a lifestyle, so there are still 14.6% of people who have pre-diabetes conditions, so it is necessary to check HbA1C.

Abstract

Abstract

The biodiversity owned by a country is a gift that must be appreciated. The potential of these natural resources can be used as research material to overcome existing problems. One of them is about how to answer the challenges of the effects caused by the use of chemicals that can harm the environment and surrounding living things. Therefore, it is necessary to modify the use of chemicals to be more optimal and of course safe both in use and effects. Modification of the particle size of a substance provides benefits because the size of a substance becomes smaller (nanoparticles) so that it will expand the surface of the contact substance. Several applications of the use of nanoparticles of a substance as a drug target carrier agent, sensor, increased bioavailability, and reduced effects of a substance have been carried out by several researchers. This study aims to evaluate the continuation of the potential of namnam leaves which have quite good bioactivity as antimicrobials. The method used is laboratory experimental. The research stages include the manufacture of namnam leaf extract, phytochemical test of the extract, and antimicrobial activity test. The extract was made by maceration using methanol for 3x24 hours and produced a yield of 11.2%. The results of the phytochemical test showed that the methanol extract of namnam leaves contains active compounds such as flavonoids, saponins, phenolics, steroids, and tannins. The content of these active substances has the potential as antimicrobials. The preliminary bioactivity test was carried out using the well method which produced inhibition zones of 15 mm, 18 mm, and 18 mm. The value of this inhibition zone is included in the sensitive category according to the CLSI data division.

Abstract

Carrots are one of the most popular plants in Indonesia. In addition to being a plant with a flavor favored, carrots also contain various bioactivities that can be utilized for health. This study aims to investigate the bioactivity of carrot extract, which includes antioxidant, antibacterial, and antifungal. The method used in this study is DPPH, which aims to determine antioxidant activity, and the disc diffusion method to determine antibacterial and antifungal activity. Carrot extract was obtained using the maceration method with a ratio of 1:10 using ethanol solvent 96% for the antioxidant test, while for the antibacterial and antifungal tests, acetone solvent was used. The extracted carrots yielded a yield of 10.1% (v/b). Based on the results of phytochemical screening, it is known that carrots contain flavonoids, saponins, tannins, and alkaloids, as well as functional groups indicating the presence of beta-carotene in carrots through FTIR. Carrots show weak antioxidant activity with an IC50 value of 125.944 ppm. The inhibitory activity of carrot extract during 24 hours of observation are concentration 40% (0.7 mm), 60% (0.77 mm) and 80% (0.85 mm). Meanwhile, in the antifungal test, carrot extract showed negative results at concentrations 60 and 80%, while at a concentration of 100% showed an inhibition zone of 0.835 mm.
