Abstract

CpG DNA is a kind of DNA synthetic immunostimulant containing CpG motifs consisting of short sequences containing one or more CpG motifs which act as a danger signal to the natural and innate immune systems. This study aimed to analyze the ability of CpG DNA to increase Interleukin 1β (IL1β) gene expression in tiger grouper so that it could be used as immunostimulant. Analysis of the expression level was carried out by extracting RNA- from the cephalic kidney which was treated with CpG DNA or with PBS as a control. Complementary DNA (cDNA) synthesis was carried out using the Ready To Go You Prime First Strand Beads (GE Healthcare) kit using the RT-PCR technique. The PCR product was analized by measuring the photostimulated luminescence value using UN SCAN IT software based on the thickness of the DNA fragments. The gene expression level and photostimulated luminescence value of the IL1β gene were higher than that of the PBS treatment used as a control. The luminescence value of the photostimulated IL1β gene in CpG DNA treatment was 113684 while the PBS treatment as a control was only 95610.

Abstract

Previous studies have succeeded in isolating, and the basic sequence of mRNA encoding FAMeT Scylla olivacea. Several studies have noted that the FAMeT encoding mRNA sequence has differences in crustacean species. The aim of this study was to determine the relationship between S. olivacea and other crustaceans based on the FAMeT encoding mRNA sequence. The similarity of the FAMeT encoding mRNA sequences showed that several crustaceans had similarities to the FAMeT mRNA sequences of S. olivacea. The phylogeny tree analysis showed the existence of 2 clusters of the relationship between the mRNA encoding FAMeT of S. olivacea on the same branch as 2 other crab species: S. serrata and S. paramamosain. It was also found in Portunus trituberculatus and P. pelagicus and river prawn Macrobrachium rosenbergii. These findings indicated that the S. olivacea crab was monophyletic with 2 other mangrove crabs, namely the crab and the river prawn M. rosenbergii. So that describes a very close kinship relationship genetically. Meanwhile, the second branch consists of crustaceans such as Homarus americanus, Cherax quadricarinatus, Fenneropenaeus merguiensis, Metapenaeus ensis and Litopenaeus vannamei.

Abstract

Fish is a source of high-quality protein. Protein in fish has a complete composition and amount of essential amino acids. However, fish is a type of food that is susceptible to biological damage and is susceptible to microbial contamination so that it quickly decays. One of the most important microorganisms in spoilage of fish meat is Escherichia coli bacteria. This study aims to determine the presence of Escherichia coli bacteria in scad, tuna and skipjack tuna. The research was carried out from July to September 2021. The research method used was a sample survey method, sampling was carried out at 3 market locations, namely the TPI market and the tasiu market. Furthermore, the samples were analyzed at the Laboratory of Fish Quarantine and Quality Control Center (BKIPM) Mamuju, West Sulawesi. The results showed that the largest E.coli content was found in tuna 1 in the TPI market, which was 20 MPN/g, and the smallest E.coli content was < 3 MPN/g, namely scad fish 1 in the tasiu and TPI markets, tuna 1 and 2 in the TPI market, tuna 2 in TPI market.

Abstract

CpG DNA merupakan imunostimulan DNA sintetik yang mengandung CpG motif, terdiri dari sekuen pendek satu atau lebih motif CpG dan berperan sebagai danger signal terhadap sistem kekebalan imun natural maupun bawaan. Penelitian bertujuan menganalisa kemampuan CpG DNA untuk meningkatkan ekspresi gen Interleukin 1β (IL1β) pada ikan kerapu macan agar dapat dijadikan sebagai imunostimulan. Analisis tingkat ekspresi gen dilakukan dengan cara mengekstrak RNA organ kepala ginjal yang diberi perlakuan CpG DNA dan perlakuan PBS sebagai kontrol.Sintesis DNA komplementer (cDNA) dilakukan menggunakan kit Ready To Go You Prime FirstStrand Beads (GE Healthcare) dengan teknik RT-PCR. Produk PCR selanjutnya dianalisa dengan UN SCAN IT berdasarkan ketebalan fragmen DNA. Tingkat ekspresi gen serta nilai luminescence photostimulated dari gen IL1β lebih tinggi dibandingkan dengan perlakuan PBS sebagai kontrol. Nilai luminescence photostimulated gen IL1β pada perlakuan CpG DNA yaitu 113684 sementarapada perlakuan PBS sebagai kontrol hanya sebesar 95610.

Abstract

Penelitian sebelumnya telah berhasil mengisolasi, dan sekuens basa nukleotida mRNA penyandi FAMeT Scylla olivacea. Beberapa penelitian menuliskan bahwa urutan mRNA penyandi FAMeT memiliki perbedaan pada spesies krustasea. Tujuan penelitian untuk mengetahui jarak kekerabatan S. olivacea dengan krustasea lainnya berdasarkan sekuen mRNA penyandi FAMeT. Kesamaan sekuen mRNA penyandi FAMeT menunjukkan beberapa krustasea yang memiliki kemiripan dengan sekuen mRNA FAMeT S. olivacea. Analisis pohon filogeni menunjukkan adanya 2 klasterkekerabatan mRNA penyandi FAMeT S. olivacea pada cabang yang sama dengan 2 spesies kepiting lainnya: S. serrata dan S. paramamosain. Selain pada genus Scylla, ditemukan pula pada Portunus trituberculatus dan P. pelagicus serta udang sungai Macrobrachium rosenbergii. Temuan tersebut menunjukkan bahwa kepiting S. olivacea monofiletik dengan 2 kepiting bakau lainnya, yaitu rajungan serta udang sungai M. rosenbergii. Sehingga menggambarkan hubungan kekerabatan yang sangat dekat secara genetik. Sementara pada cabang ke 2 terdiri atas krustasea jenis udang-udangan seperti Homarus americanus, Cherax quadricarinatus, Fenneropenaeus merguiensis, Metapenaeus ensis dan Litopenaeus vannamei.

Abstract

Udang vaname (Litopenaeus vannamei) adalah salah satu sepesies udang yang bernilai ekonomis tinggi, menjadi salah satu produk perikanan yang dapat menghasilkan devisa bagi negara. Salah satu faktor yang mempengaruhi tingkat keberhasilan penggelondongan udang vaname adalah kapadatan penebaran. Padat tebar yang tinggi akan meningkatkan kandungan bahan organik akibat penumpukan sisa pakan. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh padat tebar dan padat tebar yang optimum terhadap pertumbuhan dan kelangsungan hidup post larva udang vaname. Penelitian ini didesain degan meggunakan rancangan acak lengkap (RAL) yang tediri dari empat perlakuan dan 3 ulangan, perlakuan tersebut adalah 10 ekor/l; 15 ekor/l; 20 ekor/l dan 25 ekor/l. Bedasarkan hasil analisis ragam (anova) berbagai perlakuan dalam penelitian ini tidak memberikan pegaruh yang nyata (α>0,05) terhadap pertumbuhan dan kelangsungan hidup post larva udang vaname. Perlakuan terbaik dalam penelitian ini adalah padat tebar 25 ekor/liter.

Abstract

Keberhasilan suatu kegiatan budidaya terutama pada skala intensif salah satunya ditentukan oleh pakan buatan. Karenanya, pakan buatan menghabiskan porsi terbesar biaya operasional dalam suatu kegiatan budidaya. Penelitian tentang penggunaan bahan pakan alternatif banyak dilakukan untuk menekan besarnya biaya pakan ikan. Salah satunya adalah pemanfaatan limbah kulit pisang. Penelitian ini bertujuan untuk melihat untuk melihat pertumbuhan dan respon fisiologis ikan nila yabf diberi pakan mengandung tepung kulit pisang yang difermentasi Saccharomycess cerevisiae sehingga mendapatkan informasi yang lebih dalam mengenai potensi tepung kulit pisang terfermentasi untuk dijadikan bahan baku pakan menggantikan bahan baku pakan import yang memiliki harga tinggi seperti tepung ikan dan kedelai. Kulit pisang yang digunakan adalah jenis pisang kepok matang yang dihaluskan dan difermentasi denga ragi roti S. cerevisiae sebanyak 2%. Dosis yang digunakan adalah 0%, 10%, 15% dan 20%. Parameter yang diujikan adalah pertumbuhan bobot mutlak, TKO, gula darah dan IHS. pertumbuhan bobot mutlak, TKO, gula darah dan IHS terbaik terlihat pada perlakuan 15% kemudian diususl oleh perlakuan 20%, 10% dan terakhir adalah kontrol 0%. Dari hasil yang diperoleh dapat disimpulkan bahwa tepung kulit pisang terfermentasi dapat dijadikan sebagai bahan baku pakan untuk mengurangi penggunaan bahan baku pakan import seperti tepung ikan dan tepung kedelai.

Abstract

The purpose of this community service is to improve the knowledge and skills of partners in making economical, highly nutritious fish feed using BSF mango as the basic ingredient. The method of implementing this service uses training and mentoring which is evaluated using interview instruments and analyzed descriptively. The results of this service activity show that training in making economical fish feed provides benefits and positive impacts for fish farmers, where farmers can make fish feed that is relatively cheaper and has a high nutritional content compared to manufactured feed sold on the market.

Abstract

The biggest challenge in cultivation activities is the availability of sufficient quantities of feed with optimal nutritional content. Bioslurry is a local material from biogas production waste with good nutritional content. This research aims to determine the amount of solid bioslurry in feed composition that effectively increases fish growth and feed efficiency. This research used a complete design of 4 treatments and 3 replications (12 experimental units). The treatments tested were solid bioslurry as a substitute for corn flour, pollard flour, and fine bran with 0% (control) percentages, 5%, 10%, and 15%. The parameters observed were proximate bioslurry, specific growth rate, survival, feed efficiency, and feed conversion ratio. Data were analyzed using analysis of variance (ANOVA), followed by the W-Tuckey test. The proximate analysis of solid bioslurry shows crude protein was 5.54%, crude fiber 15.36%, BETN 7.19%, ash 34.13%, crude fat 0.53%, and water 37.25%. The best treatment is feed C with 5% bioslurry content produced growth of 31.5 g, the best feed efficiency (116.67%), and an FCR of 0.87%. In this research, information regarding feed formulations containing bioslurry can help make feed on tilapia fish raised in floating net cages for sustainable fisheries.
