Abstract

Mulberry leaf is a traditional herb and predicted has ecdysteronecompound which act as antihyperglicemid. Glybenclamide is a synthetic medicine used to cure diabetes mellitus type 2. The leaf reported as competitive inhibitor of CYP3A4 enzyme, which metabolizingglibenclamide. However, in many cases, combination of herbs with synthesis drugs causes interaction if used at the same time. This research aimed to see interaction of ethanol mulberry leaf extract with glibenclamide through CYP3A4 gene expression in HepG2 cell culture.Sample of mulberry extract, glibenclamide, and combination both sample were tested into cell HepG2 culture. Then RNA were isolated and purification using real time PCR to see the gene CYP3A4 expression. As a result, mulberry extract acts as inhibitor enzyme CYP3A4, while glibenclamide is enzyme substrate.The combination of mulberry and glibenclamide showed increased of expression of CYP3A4 gene, means greater enzyme produced, and lower medicine on blood plasma.

Abstract

The research was carried out to isolate and characterize of bioactive proteins from plant parts of Benincasa hispida (Thunb.) Cogn and to analyse of the toxicity and cytotoxicity of the proteins. The proteins were extracted with phosphate buffer saline, then they were precipitated using 80% saturated ammonium sulphate, continued with the dialysis  using pH 7 phosphate buffer. The dialysate was fractionated through gel filtration chromatography and characterized using SDS-PAGE. The toxicity of the proteins was analyzed through brine shrimp lethality test (BSLT), followed with cytotoxicity test using HeLa and K-562 cancer cell lines. Three bioactive protein fractions were isolated from the fruits, the seeds and roots. The lowest yield of proteins was 0.021% from the fruit, then 0.051% from the seed, while the highest was 0.54% from the root. All proteins were toxic on BSLT with LC50 within the range of 24-39 µg. Characterization of proteins using SDS-PAGE indicated the molecular mass of those proteins were approximately 17-29 kDa. The cytotoxicity test of the root protein showed that the protein could inhibit proliferation of HeLa cell up to 28.50% and K-562 cell up to 36.60% compared to that  of non treated cell.                   Key words: Benincasa hispida (Thunb.) Abstract

Science education in early childhood plays a crucial role in their cognitive and creative development. Science education in early childhood also has a vital role in supporting cognitive and creative development. This research aims to explore the benefits provided by science education in stimulating critical thinking, problem solving and innovation skills in children. Through qualitative and quantitative approaches, data is collected from various sources, including classroom observations, interviews with educators, and analysis of children's learning outcomes. The results showed that children who were involved in science activities from an early age showed significant improvements in cognitive skills such as memory, concentration and analytical abilities. Apart from that, science education has also been proven to encourage children's creativity by enriching their imagination and ability to think outside the box. This article concludes that the integration of science education in the PAUD curriculum not only supports children's intellectual development but also forms a strong foundation for creativity and innovation in the future. Therefore, it is important for educators and policy makers to recognize and prioritize science education as an essential component in early childhood education, science education must be a top priority in early childhood education strategies by educators and policy makers.

Abstract

Kemampuan berbahasa merupakan aspek penting dalam perkembangan anak yang berperan signifikan dalam proses belajar, komunikasi, dan sosialisasi mereka.Anak-anak dengan kemampuan berbahasa yang baik cenderung lebih mudah memahami pelajaran dan berinteraksi dengan teman sebaya.Namun, kemampuan berbahasa ini bisa sangat bervariasi antar individu, terutama antara anak-anak dengan kepribadian yang berbeda, seperti anak-anak introvert yang cenderung lebih pendiam dan mungkin menghadapi tantangan lebih besar dalam berkomunikasi. Alat Permainan Edukatif (APE) telah lama digunakan sebagai metode untuk merangsang perkembangan kognitif dan bahasa pada anak-anak. Alat Peraga Edukasi ini dibuat untuk menstimulasi anak-anak agar pembelajran jadi menarik melalui belajar sambil bermain, yang membantu anak-anak belajar dengan cara yang menyenangkan dan tidak menekan. Penelitian ini bertujuan untuk mengeksplorasi dan mengevaluasi efektivitas APE dalam meningkatkan kemampuan bahasa anak introvert.Metodologi penelitian ini melibatkan pengamatan langsung dan pengukuran kemampuan berbahasa anak-anak introvert sebelum dan sesudah penggunaan APE dalam lingkungan pendidikan. Dari penelitian yang dilakukan dapat menunjukkan bahwa penggunaan alat peraga ini mampu sangat baik dalam meningkatkan kemampuan berbahasa anak-anak introvert, terutama dalam hal ekspresi verbal dan pemahaman kosakata.Dengan demikian, penelitian ini memberikan bukti empiris bahwa APE efektif dalam mengembangkan kemampuan bahasa anak-anak introvert. Hasilnya peneliti mengharapkan mampu menambah pengalaman dan ilmu yang bermanfaat bagi guru, ayah bunda , dan peneliti dalam merancang strategi pembelajaran yang lebih inklusif dan efektif, serta mendorong pengembangan APE yang lebih tepat sasaran untuk memenuhi kebutuhan spesifik anak-anak introvert. Dalam penelitian ini menggunaka Deskriptif kualitatif.

Abstract

Guru yang baik akan menciptakan suasana belajar yang menyenangkan, mendorong anak untuk aktif belajar, serta menumbuhkan rasa percaya diri dalam diri anak. Penelitian ini bertujuan untuk menganalisis peran guru laki-laki dalam membentuk Sidoarjo. Penelitian ini menggunakan metode kualitatif deskriptif dengan pendekatan studi kasus, yang bertujuan untuk menggambarkan secara rinci kontribusi guru laki-laki dalam perkembangan motorik kasar dan prososial anak di taman kanak-kanak (TK) di Sidoarjo. Subjek penelitian terdiri dari guru laki-laki yang mengajar di TK serta anak-anak usia dini sebagai peserta didik. Data diperoleh melalui wawancara mendalam, observasi langsung, dan dokumentasi, dengan fokus pada kegiatan yang melibatkan keterampilan motorik kasar dan prososial anak. Hasil penelitian menunjukkan bahwa guru laki-laki memainkan peran yang signifikan dalam perkembangan motorik kasar anak, antara lain melalui kegiatan fisik seperti berlari, melompat, dan permainan yang melibatkan koordinasi tubuh. Selain itu, mereka juga berperan dalam mengajarkan keterampilan prososial, seperti empati, berbagi, dan kerja sama, melalui interaksi sosial dan permainan kelompok. Guru laki-laki memberikan teladan dalam pengembangan karakter sosial dan mendukung anak-anak dalam gabungan kata.

Abstract

This study aims to analyze the influence of gender stereotypes held by Early Childhood Education (PAUD) teachers on the physical, motoric and social emotional development of early childhood. Gender stereotypes can influence the way teachers interact with children, which in turn can affect their development in various aspects. The method used in this research is a quantitative method with a survey design. Data was collected through questionnaires distributed to PAUD teachers and direct observation of teacher interactions with children in class. The research results show that teachers' gender stereotypes significantly influence children's physical, motoric and social emotional development. Teachers who hold strong gender stereotypes tend to provide more support and opportunities to children according to traditional gender roles, resulting in an imbalance in the development of physical motor and social emotional skills between boys and girls. This research suggests the need for training for PAUD teachers to be aware of and overcome gender stereotypes, as well as the importance of more inclusive and gender-sensitive educational approaches to support the holistic development of early childhood.

Abstract

Protection of early childhood children from violence and exploitation is a crucial issue that requires serious attention from various parties, including parents, educators and policy makers. This article discusses the legal interventions needed to protect young children from violence and exploitation. In this modern era, children face various risks that can threaten their physical, mental and emotional development. This research examines the existing legal framework in Indonesia, as well as international policies and regulations that aim to provide protection for children. In addition, this article evaluates the effectiveness of law implementation in the field and identifies the challenges faced in implementing child protection laws. Through a qualitative approach, data is collected from various sources, including case studies, interviews with legal experts, and analysis of legal documents. The research results show that despite progress in legislation, there are still gaps in legal implementation that need to be addressed. This article suggests concrete steps to strengthen protection mechanisms, raise public awareness, and involve various stakeholders in collective efforts to protect young children from violence and exploitation. This research emphasizes that early childhood protection must be a priority in the national development agenda. It is hoped that the implementation of comprehensive and sustainable intervention programs will be able to create a healthy, safe and prosperous young generation. Practical recommendations for stakeholders are presented as a guide in increasing the effectiveness of protecting children from violence and exploitation.
