Abstract

Anther culture technique is able to accelerate plant breeding activities. The objectives of this research was to determine the agronomic traits that could be used for selection of the dihaploid rice line population through the calculation of heritability, genotypic correlation, path analysis and selecting dihaploid rice lines. The plant material used was 65 dihaploid rice lines DH 1 of F1 anther culture plants and Ciherang and Inpari 13 as check varieties. Agronomic characters that could be used as selection character are generative plant height, number of filled grain per panicle, the total number of grains per panicle, weight of 1,000 grains, and grain per hill. Twenty three dihaploid lines were selected based good agronomic characters with criteria generative plant height between 80-120 cm, number of filled grains per panicle > 100, number of grains per panicle > 120, weight of 1,000 grains > 20 g, and grain per hill > 25 g.Keywords: anther culture, heritability, agronomic characters

Abstract

Observation and identification of rice germplasm with advanced traits are important activities in the process of obtaining a variety of rice because every genotype will show unique characters. Genotype derived from breeding program as anther culture need to be characterized to identify the doubled haploid lines that are potential to be developed in red rice breeding. This research was aimed to obtain agronomic characters of doubled haploid lines of red rice. Materials used were 59 genotypes, i.e., 57 doubled haploid (DH0) lines with red grain and 2 check varieties (Inpari 24 and Aek Sibundong) as control. The experiment used a randomized complete block design with three replications. The results exhibited there was agronomic variation among DH0 of red rice lines. Variance analysis showed that genotype affected most characters, except for grain dry weight per hill. Plant height, number of productive tiller, harvesting age, panicle length, number of grains per panicle, and number of filled grain per panicle can be used as a reference for effective selection to develop high productivity of red rice variety.

Abstract

Doubled haploid rice formation through anther culture becomes one of the alternatives in the plant breeding. The lines need to be characterized, clustered, and selected. This study aimed at determining heritability, characters, and cluster of doubled haploid lines obtained from anther culture. This experiment was conducted at IPB Rice Field Experiment Station, Darmaga, Bogor from October 2016 until January 2017. The experiment was managed according to randomized complete block design (RCBD) with 3 replications and genotype as the single factor treatment. Genotypes used were 60 genotypes consisting of 56 doubled haploid lines and 4 control varieties. The controls were Ciherang, Inpara 5, Inpari 29, and Inpari 34 Salin Agritan. The results showed that all characters have significant effect and high heritability (above 60%), so they could be used as a selection character. The tested genotypes could be grouped into three main groups. The first group consisted of 11 genotypes, the second group consisted of 44 genotypes, and the third group consisted of 5 genotypes. The main group determinant characters were productive tillers, percentage of filled grain number, plant height, panicle length, and weight of 1,000 grains. The number of filled grain and productive tillers can be used as selection characters together with yield potential.

Abstract

Rainfed rice tolerant to drought and high yielding would be an alternative to rainfed lowland areas prone to drought stress. Selection in the early phase of plant growth will accelerate the effort to obtain rainfed varieties. The objective of this experiment was to identify characters that significantly affect the selection of drought tolerance using PEG 6000 concentration of 25% in the germination phase. The experiment was conducted in greenhouse of ICABIOGRAD, Bogor, using a factorial randomized complete block design with three replications. The first factor was 32 rice genotypes while the second factor was 2 levels of PEG 6000, i.e., concentrations of 0 and 25%. The results showed that the correlation analysis and principal component analysis obtain the important characters namely the germination percentage, seminal root length, and the dry weight of seminal roots. Based on discriminant analysis, the index matching value of 96.77% was suitable to distinguish tolerant and sensitive genotypes using PEG 6000 concentration of 25% in the germination phase.

Abstract

Development of doubled haploid lines of upland rice through anther culture is one way to obtain superior upland rice varieties tolerant to drought. The objectives of this research were to determine the response of doubled haploid lines to drought stress at seedling stage and to select tolerant doubled haploid lines. The research was conducted at a screenhouse of Muara Research Station, Indonesia Center of Rice Research (ICRR) from November to December 2017. Fourteen doubled haploid lines and 4 check varieties, namely Inpago 10 and Limboto, Salumpikit (drought tolerant check) and IR 20 (drought sensitive check) were used in this study. The experiment was arranged in a randomized complete block design with three replications. The observations were conducted on leaf rolling, leaf drought, and recovery ability. Based on Friedman nonparametric analysis, HR-5-7-1-1 and HR-7-15-2-1 had moderate response with estimated median values for leaf rolling of 3.44 and 3.00, respectively. Three doubled haploid lines namely HR-2-27-2-7, HR-2-34-1-3 and HR-7-15-2-1 had moderate response to drought with estimated median value of leaf drought of 3.56. Nine lines of doubled haploid had tolerant response with estimated median value of recovery ability of 1.00-1.56. Based on weighted selection index, nine doubled haploid lines with positive and high selection index were identified; however, only two lines (HR-7-15-2-1 and B3-2) had selection index higher than Inpago 10 and Limboto.Keywords: drought tolerance, leaf rolling, recovery, weighted selection index

Abstract

Rainfed rice breeding for high yield is an alternative to increase national rice production. The breeding can beaccelerated using anther culture technique. The selection of high productivity lines may be more effective if it also involvesthe traits contributing to or affecting the yield traits. This study aimed at determining suitable characters as selection criteriaand obtain a selection index model for high yielding doubled haploid rainfed rice lines. The experiment was conductedin a greenhouse of ICABOGRAD, Bogor and Sawah Baru Experimental Station, Bogor Agricultural University using arandomized complete block design (RCBD) with three replications. Thirty doubled haploid lines derived from anther cultureand two check varieties namely Ciherang and Inpari 18 were used. The results showed that plant height (TT), number of filledgrain per panicle (GI), and productivity (PRD) could be used as suitable selection characters. The determination of selectionindex obtained a selection model which was Selection index = 0.48 PRD + 0.31 JGI - 0.31 TT. This model can be used toselect high yielding doubled haploid rainfed rice lines.

Abstract

Padi hitam merupakan sumber karbohidrat yang bermanfaat untuk kesehatan. Pengujian daya hasil merupakan tahapan penting dalam menentukan keberhasilan pemuliaan padi beras hitam. Tujuan penelitian ini ialah untuk memperoleh informasi keragaan agronomi dan menyeleksi galur-galur dihaploid padi beras hitam yang memiliki penampilan agronomi baik dan berdaya hasil tinggi untuk pengujian multi lokasi. Penelitian ini dilakukan dari bulan November 2019 sampai Juli 2020 di Bogor, Jawa Barat dan Malang, Jawa Timur. Sebanyak 23 galur dihaploid padi hitam yang memiliki sifat agronomi baik dengan produktivitas tinggi dan 3 varietas pembanding yaitu Aek Sibundong, Jeliteng, dan Inpari 24 digunakan sebagai materi genetik. Studi ini menggunakan rancangan rancangan kelompok lengkap teracak dengan 3 ulangan pada setiap percobaan. Hasil penelitian menunjukkan bahwa interaksi genotipe dan lingkungan berpengaruh nyata terhadap semua karakter pengamatan kecuali pada karakter lama pengisian gabah berdasarkan analisis keragaman. Sebanyak 14 galur yang terseleksi berdasarkan indeks terboboti memiliki karakteristik jumlah anakan produktif per rumpun dari 15.8 hingga 25.9, umur panen genjah dari 115.2 to 121.9 hari setelah semai, dan produktivitas >5.4 ton ha-1. Keragaan agronomi dan daya hasil galur-galur dihaploid padi yang terseleksi pada penelitian ini memerlukan pengujian lebih lanjut melalui uji multi lokasi untuk memperoleh calon varietas unggul baru padi beras hitam. Abstract

Bacterial leaf blight (BLB), caused by Xanthomonas oryzae pv. oryzae (Xoo) is a significant disease attacking rice crops worldwide. This disease attacks at various stages of plant growth and causes significant yield loss. Breeding new varieties resistant to BLB is important to support sustainable agriculture in the future. This study aimed to identify new superior green super rice (GSR) lines resistant to BLB disease. The experiment evaluated the resistance of lowland rice lines obtained from anther culture using a factorial randomized complete block design. The 1st factor was genotype, consisting of 20 lines, 2 checks of commercial varieties (Inpari 42 Agritan GSR and Inpari 18), a resistant check (Conde), and a susceptible check (Taichung Native 1). The 2nd factor was BLB pathotypes (i.e., III, IV, and VIII). Quantitative data on disease severity and severity index were analyzed using analysis of variance and t-Dunnett test at 5% level. The results showed that the interaction between genotype and pathotype affected the disease severity and severity index in both growth phases. The tested lines exhibited varying resistance, from susceptible to resistant, to BLB. Four lines (SN 11, 13, 57, and 58) showed moderate to resistant criteria for BLB disease of 3 pathotypes in both growth phases. The selected lines can be used as a source of parents for breeders and candidates for new superior varieties with BLB resistance properties to support the reduction of synthetic chemical bactericide inputs and control BLB disease. However, further field evaluations are necessary to assess their performance.

Abstract

The improvement of eggplant yield through breeding is crucial. Doubled haploid technology has accelerated the development of varieties. This study utilized doubled haploid anther culture-derived lines (confirmed through flow cytometry and morphology) developed from embryos from a previous study to evaluate the morpho-agronomic performance and yield of doubled haploid eggplant lines. The experiment used a randomized complete block design (RCBD), three replications, and one factor, namely genotype (35 doubled haploid lines, three F1 varieties). Observation was made on plant height, dichotomous height, stem diameter, days to flowering and harvesting, fruit length, fruit diameter, weight per fruit, number of fruits, and fruit yield per plant. The data were analyzed using ANOVA, t-Dunnett, Tukey-Kramer at a 5% level, Pearson correlation coefficient, and selection index. The results showed significant variability in the population of doubled haploid lines. Fruit yield was positively correlated with plant height, dichotomous height, stem diameter, days to flowering, fruit length, and weight per fruit, but negatively correlated with the number of fruits per plant. These variables can be used as selection criteria because of their high heritability and genotypic coefficients of variation. The selection index revealed that the high-yielding doubled haploid lines with desirable morpho-agronomic traits were RS-P2, RS-P6, RS-P9, RS-P14, RS-P18, RS-H19, RS-H20, RS-H23, RS-H27, RS-H3, RS-M31, RS-M32, RS-M33, RS-M34, and RS-M37. The selected lines with high yield and good quality fruit, similar to the commercial hybrid parent, were Hitavi’s derived lines. All selected lines serve as the genetic basis for production improvements and long-term breeding programs for sustainable and productive eggplants that benefit farmers.

Abstract

Rice production is facing a decline due to climate change and land conversion. The development of high-yielding and multi-tolerant abiotic stress rice varieties (such as drought, salinity, and flooding) is needed for adaptation to a changing climate. This research aimed to identify potential doubled-haploid (DH) rice lines with good agronomic performance and high yield through index selection. The study was conducted in the rainy season of 2024, and used a randomized complete block design with a single genotype factor and three replications. The genetic material evaluated consisted of 56 DH rice lines and 5 check varieties, i.e., Inpari 18, Inpari 30, Inpari 34, Inpari 35, and Ciherang. Observation was conducted on plant height (PH), number of vegetative tillers (NVT), number of productive tillers (NPT), days to 50% flowering (DF), days to harvesting (DTH), number of filled grains (NFG), total grains per panicle (TG), panicle length (PL), and productivity. The results showed high heritability in all observed variables, indicating that the variables could be used as selection criteria. The weighted index selection involving PH, NPT, DTH, and productivity resulted in 30 DH rice lines with good agronomic traits such as medium PH, medium NPT, medium maturity, and productivity above the check varieties. Those selected DH lines can be evaluated further in more diverse environments to study the effects of genotype, environment, and G×E interactions.
