Abstract

Legume nodulating bacteria (LNB), known also as rhizobia, are soil bacteria, which are able to form rootnodules and fi x nitrogen in the leguminous plants. The LNB availability in the soil depends on the type ofagroecosystem, where plant grows. In this study, we isolated LNB from the shrub agroecosystem in Sumatera,Indonesia, and obtained four selected bacterial strains. Among them, the isolate UGM48a formed root nodulein Macroptilium atropurpureum and showed highest number of nitrogenase activity. UGM48a also contains nifHand nodA genes. An analysis of the PCR-amplifi ed 16S rDNA and BLASTn analysis showed that UGM48adisplayed 96% similarity with Achromobacter xylosoxidans. In addition, UGM48a were successfully nodulatedGlycine max (L.) merr var. wilis. Abstract

SARS-CoV-2 telah menyebabkan 133,023 jiwa meninggal di Indonesia hingga Bulan Agustus 2021, dan 4,089,801 terkonfirmasi COVID-19. Penyebaran yang masif di masyarakat ini harus diimbangi dengan penegakan diagnosa yang cepat dan akurat. Metode RT-LAMP (Reverse-Transcriptase Loop Mediated Isothermal Amplification) memberikan alternatif deteksi yang mudah, murah, sederhana dan memungkinkan untuk dapat diakses oleh semua fasilitas kesehatan di Indonesia. Metode ini hanya membutuhkan waktu kurang dari 60 menit untuk mendeteksi patogen maupun virus. Tujuan dari penelitian ini adalah mengetahui perbandingan uji RT-LAMP berbasis pocket PCR dengan pemanfaatan water bath dalam mendeteksi virus SARS-Cov-2, penyebab penyakit COVID-19 yang menjadi pandemi saat ini. Optimasi RT-LAMP dilakukan menggunakan dua alat, yaitu pocket PCR dan water bath. Parameter optimasi meliputi suhu dan waktu optimum untuk mendeteksi SARS-Cov-2. Sampel optimasi menggunakan kontrol positif, berupa plasmid yang telah terinsersi gen SARS-Cov-2, dan kontrol negatif. Hasil optimasi RT-LAMP pocket PCR menunjukkan bahwa suhu 650C dengan waktu 30 menit telah mampu menghasilkan pendaran pada sampel plasmid kontrol positif. Variasi konsentrasi plasmid kontrol positif yang digunakan, yaitu 1 ng, 0,1 ng dan 0,01 ng, di mana semua pendar muncul di ketiga konsentrasi tersebut. Meskipun demikian, hasil optimasi suhu dan waktu RT-LAMP tersebut apabila diaplikasikan menggunakan water bath belum menunjukkan hasil pendaran, sebagaimana pada pocket PCR. Oleh karena itu, tahapan proses optimasi RT-LAMP menggunakan water bath masih perlu dilanjutkan untuk menemukan suhu dan waktu optimum sehingga diperoleh hasil deteksi dengan sensitifitas yang tinggi.

Abstract

The COVID-19 pandemic that has occurred throughout the world, including Indonesia, has affected all areas of people's lives. Family food security is one of the areas affected due to restrictions on activities in the community. One of the efforts that can be made to parse and improve family food security is to implement mindfulness which is carried out independently. This can be a solution for BSA members of Treko Village, Magelang Regency who are affected by the COVID-19 pandemic because they are able to help meet family food availability. The purpose of this service activity is to provide education and training to BSA members so they are able to strengthen family food security during a pandemic. The method of implementation is carried out in a blended manner, including (1) licensing and consolidation, (2) education and demonstrations, and (3) monitoring and consulting services. Kale plants and catfish are the choices cultivated by BSA members of Treko Village through Budikdamber. Partners get online education through social media groups about the benefits of budikdamber. In addition, offline training is conducted to provide direct education regarding the preparation of buckets for cultivation, planting media and how to care for budikdamber. Based on the results of the business analysis, members of the BSA Desa Treko who apply the budikdamber can save Rp. 448,000 per year on family expenses. This can help the family economy during the COVID-19 pandemic.

Abstract

Dampak pandemi COVID-19 telah mempengaruhi semua sektor kehidupan. Sektor ekonomi dan ketahanan pangan merupakan beberapa sektor yang terdampak COVID-19. Pendapatan dan daya beli masyarakat mengalami penurunan yang berimbas ke sektor ketahanan pangan. Masyarakat harus memenuhi gizi keluarga di tengah keterbatasan pendapatan. Dengan demikian, alternatif sumber pangan yang murah dan terjangkau diperlukan oleh masyarakat, termasuk juga oleh anggota Balai Sakinah ‘Aisyiyah (BSA) Desa Treko, dalam pemenuhan gizi keluarga. Budidaya ikan dan tanaman sayur dalam ember (budikdamber) dapat menjadi sebuah solusi dalam rangka pemenuhan sumber pangan dan gizi di masa pandemi. Tujuan dari kegiatan pengabdian ini adalah memberikan edukasi mengenai sistem pertanian budikdamber kepada BSA Desa Treko. Peserta dalam kegiatan pengabdian ini sebanyak 11 orang yang terdiri dari ibu rumah tangga. Pelaksanaan pengabdian dilakukan secara luring dengan memberikan edukasi dan pelatihan budikdamber. Tahapan pelaksanaan kegiatan, meliputi tahap persiapan media aquaponik tanaman sayur, tahap persiapan budidaya ikan budikdamber, tahap pemeliharaan, dan tahap pemanenan. Tanaman sayur dipilih tanaman kangkung, sedangkan ikan yang dibudidayakan berupa ikan lele. Kegiatan pengabdian terlaksana dengan baik, dan mitra memberikan respon positif selama pelaksanaan. Anggota BSA sangat antusias mencoba budikdamber di rumah masing-masing. Sebanyak 72,73% anggota BSA masih melakukan pemeliharaan hingga masa panen tanaman kangkung dan ikan lele. Tanaman sayur dan ikan lele hasil budikdamber diharapkan mampu menjadi sumber pangan dan gizi tambahan keluarga dalam rangka menjaga kesehatan tubuh dan menjaga daya tahan tubuh selama menghadapi COVID-19.

Abstract

Salak (Salacca zalacca) is a fruit that is widely developed in Sleman, Yogyakarta Special Region, and as an icon of the Ledoknongko Tourism Village. The Covid-19 pandemic has reduced the number of tourists visiting the Ledoknongko Tourism Village. Covid-19 is very influential on residents' income and, of course, the use of salak fruit. One alternative to using salak fruit is to process it into natural yeast made from salak fruit (Salak Yeast Water). Natural yeast or wild yeast is the yeast that is usually always present in some foods, such as fruits, vegetables, and cereals. Furthermore, the yeast can be used for making bread. The training was conducted for a group of women from the Ledoknongko Tourism Village, which included a pre-test, giving materials, the practice of making salak yeast and fried bread, and a post-test. The pre-test and post-test results showed the participant's understanding of salak yeast; in the pre-test, the correct answers were 30%, and in the post-test, 93%. In addition, according to a survey conducted, particpants have a high desire to continue to train skills and develop products from salak yeast water so that salak yeast water has the potential to continue to be developed and commercialized in Ledokonongko Tourism Village

Abstract

Dendrobium sp. orchids, especially hybrid varieties, are one of the species that can tolerate direct sunlight well. Fertilizing these orchids involves using foliar fertilizers, made from organic or chemical materials, applied through the leaves' stomata. This method involves spraying the fertilizer to provide additional nutrients beyond what the roots absorb. This research aimed to determine the effects of fertilizers on leaf count and plant height. The experiment included two treatments and two replications, with methods such as media sterilization, sample preparation, watering, transplanting, fertilizing, and plant observation. Data were analyzed using one-way ANOVA with a p-value <0.05. Foliar fertilizer was applied to the underside of the leaves between 8-9 AM or 3-4 PM when stomata were open. Spraying was avoided during the rainy season to prevent fertilizer runoff while stomata were closed. Observations revealed that using Grow Quick Leaf Booster (LB) fertilizer at a concentration of 2 ml (diluted with ½ water) promoted longer growth than Gandasil fertilizer at 1 ml (diluted with ½ L of water and 1 tablespoon), which also produced good results and accelerated orchid growth. Gandasil increased plant height, leaf count, leaf diameter, and the number of seedlings, while LB fertilizer primarily enhanced plant height. These findings suggest that while both fertilizers improve growth, Gandasil has a broader range of benefits, including vegetative growth and leaf development, whereas LB is more focused on increasing plant height.

Abstract

Gliocladium sp. It is known to have an antagonistic mechanism against other organisms by means of hyper parasitism, antibiosis and lysis, as well as competition. This study aims to determine the effectiveness of Gliocladium sp. in inhibiting several types of plant pathogenic fungi and their exoenzyme activity. Inhibition testing was carried out by the Dual Culture Assay method on several plant pathogenic fungi including Colletotrichum gloeosporioides, Rhizoctonia solani, Fusarium sp., and Phytophthora sp. The exoenzyme activity tested includes cellulolytic tests, amylolytic tests, and chitinolytic tests. Antagonist testing showed that Gliocladium sp. able to inhibit the growth of all pathogenic fungi tested with varying percentage of inhibition. High percentage of inhibition was shown in the pathogens Colletotrichum gloeosporioides and Rhizoctonia solani with percentages of 78.75% and 75%, respectively. Then, the antagonist activity against Fusarium sp. has a moderate resistance percentage of 56%. Meanwhile, a low percentage of inhibition is shown in Gliocladium sp. against Phytophthora sp. with a figure of 25%. From the characterization of exoenzyme activity in Gliocladium sp., negative results were obtained in liquid and solid Carboxymethyl Cellulose, amylum, and chitin media. Meanwhile, in the amylolytic test, positive growth was marked by the appearance of mycelia on the surface of the amylum liquid medium, but in the solid media negative results were obtained against the amylase enzyme. Gliocladium sp fungus has the potential to be a biocontrol agent against various plant pathogenic fungi

Abstract

Latar belakang: Isu pangan masih menjadi tantangan untuk memenuhi kebutuhan primer manusia. Hal ini disebabkan kurangnya aspek ketersediaan dan keterjangkauan pangan di masyarakat. Kebun gizi menjadi salah satu alternatif untuk menyediakan bahan pangan yang murah dan terjangkau dalam rangka pemenuhan gizi. Tujuan: Pengabdian ini bertujuan untuk memberikan edukasi dan pelatihan budidaya ikan dan sayuran melalui sistem pertanian Budikdamber sebagai sarana kebun gizi di sekolah. Metode: Kegiatan pengabdian terdiri dari beberapa tahapan, meliputi kegiatan konsolidasi, persiapan, pelaksanaan kegiatan, pemeliharaan, pemanenan, monitoring dan evaluasi. Pada tahap pelaksanaan kegiatan dilaksanakan dalam bentuk sosialisasi dan pelatihan pembuatan budikdamber. Hasil: Mitra pengabdian mengikuti keseluruhan rangkaian dengan antusias. Mitra terlibat aktif dalam pelatihan hingga pemanenan budikdamber. Tanaman kangkung dipanen sebanyak 2 kali selama kegiatan pengabdian, sedangkan ikan lele dipanen diakhir pemeliharaan. Berdasarkan hasil kuesioner, tingkat pengetahuan mitra mengenai budikdamber meningkat sebesar 30% setelah mengikuti kegiatan. Kesimpulan: Setelah mengikuti kegiatan ini, mitra pengabdian diharapkan dapat melakukan transfer ilmu dan ketrampilan di lingkungan keluarga dalam rangka menyediakan bahan pangan yanhg terjangkau dan murah.

Abstract

Di zaman yang semakin modern banyak macam pemicu atau stresor yang membuat individu cepat mengalami stres, sering kali individu yang mempunyai kecerdasan di atas rata-rata tetapi untuk penyelesaian problem atau permasalahan yang dihadapi di lingkungan kurang bisa menyelesaikan sehingga menimbulkan stres. Ketika individu tidak mampu mengendalikan stres yang dialami akan menimbulkan gejala-gejala yang merugikan dirinya sendiri sehigga mengganggu aktivitas di lingkunganya.Horticulture therapy atau terapi kebun atau ekoterapi merupakan terapi yang menggunakan media tanaman dan aktivitas berkebun. Horticultural Therapy Association (AHTA) terapi kebun telah terbukti efektif dalam mengurangi tingkat stres, kecemasan, dan depresi serta dapat meningkatkan kecerdasan emosional. Dalam penerapan terapi berbasis masyarakat ini salah satunya menggunakan terapi kebun Smart eco farming melalui aplikasi akuaponik dengan kombinasi budidaya ikan dalam ember (budikdamber). Adapun tujuan dari program terapi berbasis masyarakat ini ialah memberi peluang kepada siswa untuk ikut terlibat dalam melakukan aktivitas terapi kebun dan untuk mengembangkan keterampilan sosial, emosional, dan motorik, sekaligus meningkatkan kecerdasan emosional dan menurunkan stress pada siswa. Kata Kunci: budikdamber, terapi kebun, kecerdasan emosi, stress, sekolah dasar ABSTRACT In increasingly modern times, there are many kinds of triggers or stressors that cause individuals to experience stress quickly, often individuals who have above average intelligence but are unable to solve problems or problems faced in the environment, which causes stress. When individuals are unable to control the stress they experience, it will cause symptoms that are detrimental to themselves and thus disrupt activities in their environment. Horticulture therapy or garden therapy or ecotherapy is a therapy that uses plant media and gardening activities. Horticultural Therapy Association (AHTA) garden therapy has been proven to be effective in reducing levels of stress, anxiety and depression and can increase emotional intelligence. In implementing this community-based therapy, one of them is using Smart eco farming garden therapy through the aquaponics application with a combination of cultivating fish in buckets (budikdamber). The aim of this community-based therapy program is to provide opportunities for students to be involved in garden therapy activities and to develop social, emotional and motor skills, as well as increasing emotional intelligence and reducing stress in students.
