Abstract

In vitro culture method can be used to obtain superior chrysanthemum seedlings. The objective of this research is to determine the origin of the cuttings and the best type of planting media for the growth of chrysanthemum seedlings at acclimatization stage. Data collection for this research was carried out in March-May 2024 at Lampung State Polytechnic Ornamental Plant Net House. This research used a Randomized Block Design (RAK) with two factors, namely origin of cuttings seedlings and type of planting medium with 6 treatment combinations which were repeated 4 times. The first factor are origin of the cutting seedlings (P1) top of the plant, and (P2) middle part of the plant. Meanwhile, the second factor are type of planting medium (M1) husk charcoal, (M2) goat pen compost, and (M3) cocopeat. The results of this research show that the use of shoot cuttings with cocopeat and charcoal husk planting media showed better results in all parameters.

Abstract

The growth regulators in media have the potential to significantly influence the process of in vitro plant regeneration. The response of explants in forming shoots also depends on the type and the combination of growth regulators. This research aimed to identify the most optimal type and concentration of cytokinin for the multiplication of in vitro porang shoots. Additionally, it determined whether the addition of cytokinins should be accompanied by auxin/IAA for porang shoot multiplication. To achieve these objectives, a completely randomized design with six replications was employed, followed by a 5% BNJ. The tested treatments consisted of various combinations of growth regulators. Several observations were recorded, including the speed of explants forming prospective shoots/shoots (days), the age of the explants forming prospective shoots/shoots, the number of prospective shoots and shoots produced per explant, and shoot height (cm). The results showed that 1) Tdz 2 and IAA 0.5 mg.l-1 formed the highest shoots candidates and shoots in porang multiplication in vitro, and 2) the addition of BAP and Tdz 2 mg.l-1 should be combined with IAA 0.5 mg.l-1, while BAP and Tdz 3 mg.l-1 did not require IAA.

Abstract

The purpose of this study was to determine the best basic medium type and TDZ concentration for the development of somatic embryos from Dendrobium orchid leaf explants. Three replications were arranged factorial (2x3) in a completely randomized design for this study. First, there were two types of basic media, ½ MS and MS. The second factor was the concentration of cytokinin thidiazuron 1, 2 and 3 mg/L. Each experimental unit consisted of 5 culture bottles, each contained 5 explants. The research results showed that 1) The use of both types of basic media was able to induce callus formation on Dendrobium 'Gradita 31' orchid from leaf explants. 2) The use of 3 mg/L thidiazuron which combined with ½ MS or MS media was able to form primary callus faster than other treatments. 3) The higher percentage for embryo somatic and shoots formation were also found in 3 mg/L thidiazuron.
