Abstract

Many of potential enzymes can be found in the ecoenzyme, one of them is amylase. Amylase is an enzyme that is able to hydrolize the glycoside bonds of starch or starch into dextrin, glucose and maltose which is widely used in various industries such as beverages. This study aims to isolate the ecoenzyme bacteria and test their ability to produce amylase. Isolation of ecoenzyme bacteria was carried out using serial dilution methods 10-1, 10-3, 10-5. A total of 0.1 ml of each dilution series was pipetted and spread on the Nutrient agar (NA) medium. The bacterial isolates that grew were then purified and identified by morphological observation, Gram staining and their enzymatic activity was tested using Starch Agar (SA) media qualitatively. The results of this study showed there were 39 bacterial isolates from ecoenzyme with different morphological characteristics. The amylase activity test were found that 34 isolates had a positive activity to hydrolize the starch in the SA media which was indicated by the formation of a clear zone around the bacterial colonies. Each bacterial isolate had a different hydrolysis index value, which ranged from 9.45 to 23.65. The highest clear zone diameter index value from starch hydrolysis was EJM 15 isolate.

Abstract

The methanol extract of sambang darah leaves (Excoecaria cochinchinensis L.) contains flavonoids, saponins, tannins and triterpenoids which have the potential as antibacterial compounds. The goal of this research is to produce liquid soap that preparations with four formulas by varying the methanol extract of sambang darah leaves as the active ingredient, characterizing, and understanding its antibacterial activity. Organoleptic tests, foam stability, pH, specific gravity, and viscosity were all used to characterize liquid soap. The best results were obtained with liquid soap formulation 3 (3 g extract), which had foam stability of 84.10%, a pH of 9.62, specific gravity of 1.03 g/mL, and viscosity of 492.933 cP. The organoleptic test revealed that the liquid soap was a homogeneous viscous liquid with a blackish-brown color and a characteristic extract odor. The inhibition zone for antibacterial activity against Staphylococcus aureus and Escherichia coli was 9.15 mm and 13.72 mm, respectively. In antiseptic tests, all soap formulas reduced bacteria on the thumb.

Abstract

Latar Belakang: Penderita diabetes melitus rentan terhadap komplikasi infeksi jamur. Salah satu infeksi yang paling sering mengenai penderita diabetes melitus adalah kandidiasis. Beberapa faktor yang memudahkan infeksi kandidiasis, yaitu kesehatan mulut yang buruk, penurunan sekresi dan pH saliva, peningkatan kadar glukosa darah, serta keadaan imunodefisiensi. Pasien DM dapat mengalami infeksi kandidiasis sebesar 36,7% yang disebabkan oleh jamur Candida albicans. Penelitian ini bertujuan untuk melihat hubungan antara lama menderita penyakit dan kadar glukosa darah terhadap kejadian kandidiasis oral. Metode: Penelitian ini menggunakan studi observatif analitik dengan desain penelitian cross-sectional dilakukan di RSUD Harapan dan Doa Kota Bengkulu pada bulan Mei 2019 dengan jumlah sampel 96 pasien diabetes mellitus tipe 2 melakukan kontrol di poli penyakit dalam diambil secara consecutive sampling. Analisis hubungan antara dua variabel menggunakan uji komparatif Chi Square. Analisis multivariat antar semua variabel menggunakan regresi logisitik. Hasil: Subjek penelitian kelompok lama menderita penyakit DM tipe 2 ≥5 tahun merupakan kelompok positif kandidiasis oral tertinggi sebanyak 32 orang (66,7%) dan kelompok kadar glukosa darah tidak terkontrol 43 orang (76,8%). Hasil uji statistik bivariat Chi Square untuk lama menderita DM tipe 2 diperoleh nilai p=0,024 (p < 0,05), untuk hasil uji statistik bivariat Chi Square kadar glukosa darah tipe 2 diperoleh nilai p<0,001 (p < 0,05), menunjukan terdapat hubungan yang bermakna antara lama menderita DM dan kadar glukosa darah dengan kejadian kandidiasis. Hasil analisis multivariat menggunakan regresi logisitik diperoleh nilai untuk kadar glukosa darah OR=11,393, nilai lama menderita DM dengan OR=1,735. Kesimpulan: Terdapat hubungan yang bermakna antara lama menderita penyakit dan kadar glukosa darah dengan kejadian kandidiasis oral. Abstract

Rajo Lelo Forest Park (TAHURA) is located in Tanjung Terdana Village, Pondok Kelapa District, Central Bengkulu Regency. This forest area is overgrown with various trees, including ectomycorrhizal host trees. A purposive sampling plot method was used on each found fungus to determine ectomycorrhizal species' diversity, abundance, and distribution. Five plots were used, each measuring 20 cm by 20 cm. The data collected from these plots revealed important information about the forest's ecological dynamics. We examined the samples to determine the types of ectomycorrhizal species and how they aided tree health and forest biodiversity. Fungi found in the plot were identified morphologically by comparing morphological characters to references. According to the results of the analysis, there are 118 individual fungi from nine species and seven families. The species diversity index is classified as moderate, at 1.61. Clitopilus sp. is 33.05% more abundant than any other ectomycorrhizal fungus. However, there are 0.85% fewer species of Scleroderma sp., Lactarius sp., and Macrolepiota sp. than any other species. The distribution index of ectomycorrhizal fungi in Rajo Lelo Forest Park is 1.34, placing them in the clustering category.

Abstract

Candidiasis is an infection caused by the yeast of Candida albicans. C. albicans infection often affects the vaginal area which is known as vulvovaginal candidiasis. Candidiasis can be treated using traditional medicine or herbal medicine derived from plants, one of which is senduduk bulu plant (Clidemia hirta (L.) D.Don). This study aims to determine the phytochemical screening of the ethanol extract of Senduduk Bulu (C. hirta) leaves and to determine the anti-candidiasis activity of the ethanol extract of C. hirta leaves in inhibiting the growth of the C. albicans at concentrations of 10%, 25%, 50 % and 75%. Activity test of ethanol extract of C. hirta against C. albicans was analyzed by SPSS 25 application with ANOVA test analysis. The extract was prepared by maceration method with 96% ethanol solvent. Phytochemical screening is observed by the precipitation reaction and the color change reaction. The anti-candidiasis activity test was carried out with 3 replications using the disc diffusion method. The end result of making ethanol extract of C. hirta is a dark green viscous extract, with a yield calculation of 17.96%. Phytochemical test results showed that C. hirta contained flavonoids, saponins, tannins and steroids. C. hirta extract inhibited the growth of C. albicans. The most effective concentration of C. hirta extract in inhibiting the growth of the C. albicans was 75% with an inhibition power of 28.73 mm (very strong category).
