Abstract

The aim of this study is to explore the impact of interactive-conceptual-instruction with visualization on logic thinking, conceptual understanding and spiritual aspect embedding. The sampling technique used in this research was convenience sampling. This research used mixed methods with embedded experiment design. The data of logic thinking and conceptual understanding was collected through pre-test and post-test. The questionnaires were used to collect data on spiritual attitudes and were analyzed qualitatively. The results showed that logic thinking-ability improves from no logic thinking to inductive-deductive logic thinking. The improvement of logic thinking abilities was in the medium category. Understanding the concept shifted from incompetent to conceptual understanding with improvement, in the medium category. The questionnaires analysis showed that the students got spiritual values related to the awareness of God believing behind the natural phenomena that impact on their spiritual attitudes. This study also tried to apply the results of pure science research to support learning activities. Based on the result of the research, it can be concluded that the learning process using interactive conceptual with visualization can improve logic thinking, conceptual comprehension, and spiritual attitude embeding.

Abstract

This study aimed to isolate and explore the actinomycetes of CIFOR forest origin as an antimicrobial and antifungal agent, to produce an extracellular xylanase, and to identify isolates based on 16S rRNA gene sequences. Actinomycetes were isolated using Humic-acid Vitamin-B agar (HV) media. Actinomycetes colonies that grow on the medium HV was subsequently purified by growing them on yeast malt agar (YMA) media), then an antagonistic test of selected bacteria against Bacillus sp., Escherichia coli, Fusarium oxysporum, and Sclerotium sp was performed. Xylanase activity test was detected by observing a clear zone, followed by identification. Total of 35 isolates of actinomycetes isolated based on their colony morphology characteristics and diverse types of spore chains showed Streptomyces spp. of isolates CFR-06, CFR-15, CFR-17, CFR-18, and CFR-19 were able to inhibit the growth of Bacillus sp.. The highest inhibition zone has a diameter of 10.1 mm (isolate CFR-17). Isolates CFR-01 and CFR-15 were able to inhibit the growth of E. coli with the highest inhibition zone diameter of 5.1 mm (isolate CFR-15). Isolates CFR-29 and CFR-12 were able to inhibit the growth of F. oxysporum while isolate CFR-35 were able to inhibit the growth of Sclerotium sp.. Xylanase activity test showed that isolates CFR-12, CFR-20, CFR-22, CFR-24, CFR-25, CFR-30, CFR-33, CFR-34 have an ability to produce extracellular xylanase enzyme. Abstract

Most all of earth and space science concepts are unobservable and require reasoning. Unobservable concepts have made prospective physics teachers found it hard to understand the concepts correctly. It needs visualization to help understand the concepts easily. The aim of this study was to enhance the mastery concept of earth science topic especially the earth atmosphere using interactive conceptual with visualizations and authentic data analysis with GrADS (Grid Analysis Display System). There were 23 prospective physics teachers in Bengkulu, Indonesia participated in this study. A convenience sampling technique was employed in this study. This study used pre-experimental, one group pretest-posttest design. The data were collected using pretest and posttest, questionnaires, and observation sheets. The pretest and posttest were analyzed quantitatively, while the questionnaires were analyzed qualitatively. The results showed that the prospective physics teachers’ concept mastery improved from no mastery to compete with the average normalized gain of 0.64 (medium category). 30.44% of the prospective physics teachers obtained the N-gain score in the high category and 69.56% obtained the N-gain score in the moderate category. This compilation gave them experience about the real condition of solar radiation in Indonesian atmosphere during their learning activity. It concluded that the earth science learning using interactive conceptual instruction supported by visualization and GrADS could enhance the students’ concept mastery.

Abstract

First report of Begomovirus infection on papaya in Bengkulu, Indonesia. A field survey was conducted during 2019, wefound a severe systemic yellow mosaic, striped green mosaic on leaves petiole, green spots on the fruit of papaya, leafmalformation, and stunting symptoms on three papaya cultivation area in Rejang Lebong, Kepahiang, Bengkulu Tengah, andSeluma, Bengkulu Province, Indonesia. A begomo-like virus was inferred to be the possible cause of the virus-disease-likesymptoms. The study aimed to identify the causal of those typical symptoms on papaya. PCR using universal primer fortranscriptional activator protein (TrAp) and replication-associated protein (Rep) gene of Begomovirus successfully amplifiedthe DNA fragments of 900 bp in all 10 detected samples, except for samples with leaf malformation and stunting symptoms. Itis indicating that those typical symptoms on papaya is associated with Begomovirus infection, while the causal of leafmalformation and stunting is unknown yet. This work is the first report of Begomovirus infected papaya in Indonesia. Severedisease incidence caused by this pathogen was observed on papaya plants in Bengkulu Province that was in the range of 42–100%. This finding is a precious information to be used for identification, and characterization the species of the virus,determining control strategies against the disease.

Abstract

Makrofungi merupakan fungi yang dapat dilihat dengan mata secara langsung, sebagian makrofungi hidup ditanah dan sebagian hidup sebagai safrofit pada tanaman dan ada juga yang hidup bersimbiosis dengan akar tanaman. Fungi Basidiomycota adalah fungi yang memiliki basidiokarp atau tubuh buah yang besar yang dapat dilihat dengan mata secara langsung  dan memiliki ciri-ciri diantarannya mempunyai basidium dan koneksi penjepit. Desa Barumanis terletak di Kecamatan Bermani Ulu Kabupaten Rejang Lebong,  Provinsi Bengkulu. Kawasan di sekitar hutan Desa Barumanis memiliki suhu yang dingin sehingga berpotensi memiliki keanekaragaman makrofungi khususnya filum  Basidiomycota.  Karakteristik kawasan lingkungan Desa Barumani terletak di kawasan kaki Bukit Daun dimana sebagian besar wilayahnya merupakan  perkebunan dan pertanian. Tujuan penelitian ini yaitu untuk mendapatkan jenis makrofungi filum Basidiomycota yang tumbuh di Desa Barumanis dan kawasan hutan sekitarnya Kecamatan Bermani Ulu, Kabupaten Rejang Lebong. Metode yang digunakan dalam penelitian ini yaitu metode jelajah (Cruise Method) yaitu dengan menjelajahi Desa Barumanis dan kawasan hutan sekitarnya, Kecamatan Bermani Ulu, Kabupaten Rejang Lebong. kemudian data yang diperoleh diolah secara analisis deskripsi kualitatif. Dari hasil penelitian yang telah dilakukan ditemukan spesies makrofungi yaitu Homidicutis marginata, Lycoperdon pyorforme, Fomitopsis rosea, Tramella fuciformis, Ganoderma lobatum, Auricuaria americana, Mycena fumosa, Baespoora myosura, Baespoora myosura, Galerina autumnalis, Phelinus gilvus, Hyplohoma acutum, Laccaria amethystina, Cantharellus cinnabarinus, Coprinus variegatus, Mycena leptothepalla, Agrocybe cylindracea, Stereum ostrea, Pleorotus sp. Makrofungi yang diperoleh dari penelitian ini diharapkan menjadi informasi keanekaragaman jenis makrofungi dari filum Basidiomycota di Desa Barumanis dan kawasan hutan sekitarnya Kecamatan Bermani Ulu, Kabupaten Rejang Lebong dan melengkapi data makrofungi yang terdapat di Provinsi Bengkulu.

Abstract

Infectious diseases caused by microbes are increasing; the growth of microbes can be inhibited by using antibiotics. Continuous and inappropriate use of antibiotics causes microbes to become resistant. The resistance can be reduced by finding new sources of antibiotics, one of which is from the bird nest fern (Asplenium nidus). This study aimed to determine the potency and concentration of A. Nidus extract in inhibiting the growth of Staphylococcus aureus ATCC 6538, Escherichia coli ATCC 8739, Pseudomonas aeruginosa ATCC 15442, and Bacillus subtilis ATCC 19659. The extract was prepared using the maceration method with methanol as the solvent. Phytochemical tests include alkaloids, flavonoids, steroids, terpenoids, saponins, phenolics, tannins, and quinones. The minimum inhibitory concentration test was carried out on 12 treatments with three replications; the antimicrobial effectiveness test was carried out on seven treatments with five replications using the disc diffusion method. Phytochemical test results showed A. Nidus contains flavonoids, tannins, saponins, and phenolics. A. nidus extract has the potential to inhibit the growth of the testing pathogenic bacteria, with the most effective concentration on P. aeruginosa of 45% with an inhibitory zone was about 14,16 mm, on E. Coli started of 55% with an inhibitory zone was about 13,68 mm, on B. Subtilis of 65% with an inhibitory zone was about 14,80 mm and S. aureus was 75% with an inhibitory zone of 11,96 mm. Abstract

Chitinolytic bacteria are bacteria that have chitinolytic activity, which is able to hydrolysis the composition of chitin which composes many fungal cell walls. Chitinolytic bacteria are currently more widely used because of their ability as a biological control agent to the pathogenic fungi especially in horticultural and plantation crops. This research was conducted with the aim of obtaining isolates of chitinolytic bacteria that were able to inhibit the growth of plant pathogenic fungi in Vitro on chitin agar media. Fusarium oxysporum, Sclerotium rolfsii, and Rhizoctonia solanii are used in the inhibition test of chitinolytic bacteria. Bacteria were isolated and screened from the soil of Bukit Dua Belas National Park and Oil Palm Plantations in Jambi using 0.3% chitin agar media. The results showed that two of 10 bacterial isolates were able to produce inhibition zones to the growth of hyphae of pathogenic fungi on potato dextrose agar (PDA) media. TB04-13 isolate was able to produce the largest inhibition in F. oxysporum and R. solanii about 42% and 42.05% respectively, while TB04-15 isolate produced the biggest inhibition in S. Rolfsii ranged to 25.50%. Based on the chitinolytic index (CI) values, isolates TB04-13 and TB04-15 produced CI values ​​of 1.60 and 0.63, respectively. The morphological characteristics and Gram staining of both TB04-13 and TB04-15 chitinolytic isolates are included in rod-shaped and Gram-positive bacteria. Both of these isolates can be used as antifungal-producing candidates for plant pathogenic fungi in Indonesia.

Abstract

Coliform is a group of microbes that are used as indicators of water quality. Water pollution is generally caused by pathogenic microbes from feces, household waste, and industrial activity waste. This study aimed to estimate the total number of coliform contamination in several rivers in Kepahiang Regency, namely Tebat Monok (TM), Sempiyang (SPY), Penanjung Panjang (PP), Embong Ijok (EI) Air Langkap(ALK), and Air Belimbing (ABB).  Total coliform and Fecal coliform tests were carried out using the Most Probable Number (MPN) method on Lactose Broth, Brillian Green Lactose Bile Broth and pour plates on Eosin Methylene Blue Agar media. Measurement of abiotic factors was on temperature and pH parameters. The test results of total coliform showed that 6 rivers contained total coliform under the Class II river water quality standards with a range of 1210/100 mL– 4310/100 mL and 2 rivers that were contaminated with Fecal coliform, TM and ALK, have the content of 1500/100 mL and 1700 / 100 mL. The results of the measurement of the abiotic factor, the river pH range was 7.4 - 8.2. The lowest temperature was 25oC in SPY river and the highest temperature was 26 oC on the TM, PP, EI, ALK, and ABB rivers.

Abstract

Kabau (Archidendron bubalinum) merupakan tumbuhan yang termasuk famili Leguminosae berbentuk pohon dan menghasilkan biji berwarna hijau. Masyarakat Bengkulu memanfaatkan buah kabau sebagai lalapan. Bagian kulit buah tumbuhan kabau berpotensi sebagai antifungi Candida albicans karena mengandung senyawa fitokimia. C. albicans merupakan salah satu mikroorganisme patogen penyebab infeksi seperti kandidiasis, sariawan, lesi pada kulit, dan vulvovaginitis. Penelitian ini bertujuan untuk mengetahui potensi ekstrak kulit buah kabau sebagai antifungi serta mengetahui konsentrasi ekstrak yang optimal dalam menghambat pertumbuhan fungi. Ekstrak kulit buah kabau diuji pada fungi C. albicans dengan metode difusi menggunakan kertas cakram pada media Sabouraud Dextrose Agar (SDA). Ekstrak kulit buah kabau dilarutkan dengan pelarut aquades dan dimetil sulfoksida 2% pada konsentrasi ekstrak 5%, 10%, 20%, 40% dan 80%. Hasil penelitian menunjukkan konsentrasi 40% dengan pelarut aquades yang paling efektif dalam menghambat pertumbuhan fungi C. albicans dengan diameter daya hambat 4,05 mm. Dari data hasil penelitian tersebut, maka dapat disimpulkan bahwa ekstrak kulit buah kabau berpotensi sebagai antifungi C. albicans.
