Abstract

Red fruit (Pandanus conoideus Lam.) belongs to genus Pandanus. Red fruit oil has the activity as an antibacterial. The research aimed to obtain endophytic fungi isolates on the red fruit peel and assay the antibacterial activity by TLC-Bioautography. The research was experimentally conducted including the process of isolation of endophytic fungi, purification, macroscopic examination, screening for antibacterial activity against Staphylococcus aureus and Salmonella typhi, fermentation for 7, 14 and 21 days, separation and antibacterial assay by TLC-Bioautography against Staphylococcus aureus and Salmonella typhi, the isolate fermentate of endophytic fungi for 7 days showed an Rf value of 0.49 against Staphylococcus aureus and an 0.47 against Salmonella typhi. The isolate fermentate of endophytic fungi for 14 days showed an Rf value of 0.58 against Staphylococcus aureus and an 0.69 against Salmonella typhi. The isolate fermentate of endophytic fungi for 21 days showed an Rf value of 0.69 against Staphylococcus aureus and an 0.67 against Salmonella typhi. In conclusion, red fruit peel had endophytic fungi isolate with the potency as an antibacterial.

Abstract

Walnut seeds (Canarium indicum L.) contain tannin, flavonoids, carotenoids which is potentially fungtioning as antioxidants. The research aimed to examine the antioxidant activity of fermentate extract of walnut seeds endophytic fungi by autography-TLC. The result of the endophytic fungi isolation in Walnut seeds (Canarium indicum L.) obtained 5 isolates with characteristics. The test result of screening free antiradical activity obtained that isolate 4th was active as an antioxidant indicated by the presence of brown spots with the addition of 1,1-diphenyl-2-picrylhydrazyl 0.004%. Isolate 4th was fermented using Maltosa Yeast Broth (MYB) medium to obtain fermentate supernatant and mycelia. The Fermentate of supernantant of isolate 4th was extracted to obtained supernantant of ethyl acetate extract. Based on the result of free antiradical activity test of ethyl acetate fermentate extract isolate 4th by autography-TLC using ethyl acetate: methanol: water eluent (6:2:1), obtained Rf 0.78 value active as free antiradical by spraying 1,1diphenyl-2-picrylhydrazil 0.004%.

Abstract

Acne is one skin problem commonly known as acne vulgaris, that is a state where the skin pores are clogged. Some of the bakteria causing the growth on acne are Propianibacterium acne and Staphylococcus epidermidis . Green tea (Camellia sinensis L.) is one of the plants that can be useful as antibacterial. This study aimed to obtained the antibacterial activity of ethanol extract leaves and sregs of green tea which has the  potential  to inhibit the growth of acne causing bacteria by the agar diffusion method. The simplisia of green tea leaves and dregs are macerated with ethanol 96% and obtained the weight of ethanol extract of green tea leaves 34.6 grams. In determination of Minimum Inhibitory Concentration (MIC) ethanol extract of the leaves and the dregs, the MIC value obtained is the same as the two test bacteria on the concentration of 0.1%; whereas, KBM value is on the concentration of 0.4%. The result of the antibacterial activity of ethanol extract of green tea leaves at the largest concentration of 8% has an inhibitory zone diameter of 18.11 mm to Propianibacterium acne and 18.05 mm to Staphylococcus epidermidis. For the antibacterial activity test of ethanol extract the green tea dregs at the largest concentration of 8% has the largest average inhibitory zone diameter of 17.45 mm against Propinibacterium acne and 15.68 mm against Staphylococcus epidermidis.

Abstract

Isolation of bacterial cellulose from fruits in traditional market of Makassar have been done. The objective of this research is to adding diversity of microorganism that can producing cellulose and observe it’s both optimum temperature and pH. First step is isolation of bacteria from Watermelon, Papaya, Cantaloupe, Mango, Dragon fruit, and Banana that found in traditional market in Makassar using Hestrin-Schramm (HS) agar. Each colony bacteria have been separated to making pure culture using HS agar then the colony was screening in HS broth to obtain colony bacteria that producing cellulose. Screening test found that 10 isolate that can produce cellulose which is MB-B02, MS-B01, MS-B03, NB-B03, PB-B01, PB-B02, PB-B03, PB-B04, PS-B01, and PS-B03. Then the colony that producing cellulose was optimize its temperature and pH. Cellulose obtained from optimization was measure it’s weight to determine optimum temperature and pH. Based from optimization found that 250C was optimum temperature for MS-B01, MS-B03, NB-B03, dan PB-B04, and 370 was optimum temperature for MB-B02, PB-B01, PB-B03, PS-B01, dan PS-B03, meanwhile for isolate PB-B02 optimum both in 250C dan 370C dan pH optimum for all isolate is pH 6

Abstract

Antibacterial activity of taro leaves ekxtract has been conducted. The Recearch aimed to determine the antibacterial activity and concentration of ethanol extract of taro leaves (Colocacia esculeta) inhibited the growth of either Salmonella thypi and Staphylococcus aureus by agar diffusion. The result of the antibacterial activity screening in concentration of 1% showed that the ethanol extract of taro leaves by solid dilution provided activity againts Salmonella thypii and Staphylococcus aureus. Based on the test results of antibacterial activity by agar diffusion using concentration of 1% , 2% and 4 % showed that the ethanol extract of taro has the largest inhibition zone diameter againts Staphylococcus aureus of 4% was 15 mm.

Abstract

The aim of the research to determine the antibacterial and antioxidant activity of Colocacia esculenta fraction leaves derive from Pare-Pare South Sulawesi in using TLC- Bioautography and diphenilpycril Hydroxil. Screening antibacterial activity test of the Colocacia esculenta fraction leaves exhibited in concentration 1% active on pathogenic bacteria Salmonella thypi. Based on either antibacterial and antioxidant activities by TLC-Bioautography showed that fraction at Rf 0.70 value active on bacteria Salmonella thypi and antioxidant active as well. Identification of active chemical compound showed flavonoid compound using Iodium-KI and antimo (III) clorida as reagent.
