Abstract

An investigation has been conducted on the antimicrobial activities of extract teh hijau leaves (Camellia sinensis) and jati belanda leaves (Guazuma ulmifolia Lamk.) using TLC-Bioautography. The aom of this research is to test the antimicrobial activity of extract against some pathogen microbes. The preliminary research conducted screening test using several tipe of microbes including Bacillus subtilis, Escherichia coli, Pseudomonas aeruginosa, Salmonella typhi, Shigella dysentriae, Staphylococcus aureus, Staphylococcus epidermidis, Strepthococcus mutans, and Vibrio cholera. The screening results of extract inhibited almost all of pathogen microbies, except Bacillus subtilis and Staphylococcus aureus for extract teh hijau leaves and Vibrio cholera, Staphylococcus aureus, and Escherichia coli for extract jati belanda leaves. The research was continued with the antimicrobial activity against severeral microbes at different Rf and the result showed that the higher inhibited at Staphylococcus epidermidis Rf 0,02 and Shigella dysentriae Rf 0,49, 0,73 and 0,8 for teh hijau leaves, Pseudomonas aeruginosa Rf 0,02 dan 0,49 for jati belanda leaves. That extracts have antimicrobial activity to several pathogen microbes, so that are can use as one of alternative antimicrobial.

Abstract

Preliminary research permot leaf (Passiflora foetida L.) used Acetobacter xylinum bacterial culture and Sacharomyces cerevisiae yeast fermentation in determined the alcohol content of kombucha with the purpose of determined the alcohol content of kombucha products permot leaf (Passiflora foetida L.) from Makassar in South Sulawesi gas chromatography. Based on result of research fermentation of kombucha product from permot leaf by add glucose 100 gram and inoculum kombucha 10 % obtaired content by gas chromatography as big as 0,0627292 %.                                                                                     Keyword : Permot leaf, kombucha, alcohol, gas chromatography

Abstract

A research has been done about profit of permot leaves (Passiflora foetida L.) by using culture of kombucha with the aim to know the use of permot leaves (Passiflora foetida L.) as preparation of kombucha in a fermentation. Permot dried leaves boiled in 1000 ml water until boiling then got extract of permot. Extract of permot leaves added with 100 gram glucose and 10% of inoculum kombucha culture then fermented for 5 – 12 days. The results of kombucha preparation from permot leaves then testing with organoleptic, measuring pH and total acid test. Based of the results in preparation kombucha permot leaves (Passiflora foetida L.) showed that the  color-test organoleptic with hedonic method was obtained values 2.5, flavor 1.9, and taste 2.4. pH measurement before fermentation is 4 and after that is 3 with total acid of 0.66%. Key Word : Permot Leaves (Passiflora foetida L.), Fermentation, Kombucha of Preparation

Abstract

Utilization of wild plants around us needs to be done to expand the chances of wild plants being used as traditional medicine. These plants have the main content of the active compounds in the form of steroids, terpenoids, flavonoids, tannins and is reported to have antibacterial activity. This research aims to determine the concentration of endophytic fungi isolates IDGG 02, IDGG 03 and IDGG 06 from the leaves of galing-galing (Cayratia trifolia L). The first stage purification isolates of endophytic fungi in order to obtain single isolates. Then fermented using a shaker for 21 days at room temperature. Fermented extracted with ethyl acetate solvent using a liquid-liquid extraction methods to obtain a dry extract. Testing the minimum inhibitory concentration (MIC) with concentrations of 2048, 1024, 512, 256, 128, 64, 32, 16, 8 and 4 µg / mL and testing for antibacterial activity using agar diffusion methods against bacteria Staphylococcus aureus, Staphlococcus epidermidis, Escherichia coli and Vibrio cholerae.The results obtained MIC 2048 MIC ug / mL to extract isolates IDGG 02 and IDGG 06, while the extract obtained 03 isolates IDGG 1024 MIC ug / ml against all bacteria. Results of testing the antibacterial activity of endophytic fungi isolated extract IDGG 02, 03 IDGG produce diameter zone of inhibition at a concentration of 2048 mg / mL against Staphylococcus aureus, Staphlococcus epidermidis, Escherichia coli and Vibrio cholerae.

Abstract

Daun patikan kebo (Euphorbia hirta L) merupakan salah satu tumbuhan tradisional yang memiliki aktivitas antibakteri. Patikan kebo memiliki kandungan senyawa antibakteri seperti flavonoid, alkaloid, tanin, terpenoid dan polifenol. Tujuan penelitian ini adalah untuk mengetahui aktivitas antibakteri ekstrak etanol daun patikan kebo (Euphorbia hirta L). Penelitian pendahuluan dengan uji skrining menggunakan bakteri Propionibacterium acnes, Pseudomonas aeruginosa, Staphylococcus aureus, Staphylococcus epidermidis. Metode uji aktivitas antibakteri yang digunakan dalam penelitian ini adalah difusi agar dan KLT-Bioautografi. Hasil penelitian diperoleh untuk difusi agar pada konsentrasi terbesar yaitu 8%, pada bakteri Propionibacterium acnes dengan diameter zona hambat 15,83 mm, Pseudomonas aeruginosa dengan diameter zona hambat 18,20 mm, Staphylococcus aureus dengan diameter zona hambat 16,56 mm dan Staphylococcus epidermidis dengan diameter zona hambat 19,54 mm. Sedangkan untuk hasil uji KLT-Bioautografi dengan menggunakan eluen Etil asetat:Etanol (6:1) yang menunjukkan hasil dengan nilai Rf = 0,94 dan 0,80 yang dapat menghambat pertumbuhan bakteri Propionibacterium acnes dan Staphylococcus aureus. Pada nilai Rf = 0,94 dan 0,78 dapat menghambat pertumbuhan bakteri Pseudomonas aeruginosa dan Staphylococcus epidermidis.

Abstract

This study is aimed at testing the antibacterial activity of the compound Bis (2-Ethylhexyl) phthalate leaves fraction of Colocacia esculenta L against enteric gram negative bacteria. The search for bioactive compounds employs the vacuum liquid chromatography method used for fractionation and purification, spectroscopic analysis (infrared, proton and carbon core magnetic resonance, chromatography-mass spectroscopy) gas, Minimal Inhibitory Concentration (MIC), Minimal Bactericidal Concentration (MBC), Agar diffusion and TLC-Bioautography for activity testing against Enteric Gram Negative Bacteria. The results of the fractionation shows that based on agar diffusion, the B fraction obtains the MIC value is 0.1%, and MBC value is 1%. Spectroscopic analysis of the secondary metabolites of the leaves fraction of Colocacia esculenta L obtains the chemical structure of Bis (2-Ethylhexyl) phthalate with the molecular formula C24H38O4 and molecular weight 390.5. The test on the compound Bis (2-Ethylhexyl) phthalate from the leaves fraction of Colocacia esculenta L L by TLC-Bio autography obtains an active Rf value of 0.7 against bacteria Escherichia coli, Pseudomonas aeruginosa, Salmonella thypi, Streptococcus mutans, and Vibrio cholerae, by diffusion Agar method, the largest inhibition zone in Salmonella thypi bacteria. Based on the results, the identification of secondary metabolites from the leaves fraction of Colocacia esculenta L obtains the chemical compound Bis (2-Ethylhexyl) phthalate and based on the results of the antibacterial activity test of the chemical compound Bis (2-Ethylhexyl) phthalate leaves of Colocacia Esculenta L is potential for the treatment of enteric gram negative bacteria infections.
