Abstract

In an attempt to obtain a cost-effective and a simpler way of growing Dendrobium seedling in vitro, this study was conducted to investigate effects of basal media and organic additives on in vitro growth of Dendrobium seedlings.  The basal media used were that of half strength Murashige and Skoog (½ MS medium) and that containing foliar fertililizer 32:10:10 and trace elements (FT medium). Treatments were combinations of basal media (1/2 MS medium and FT medium) and different kinds of organic additive (tomato, potato, and mungbean sprout juice and banana homogenate). The experiment was conducted in a completely randomized design with three replicates, 5 culture vessels per replicate and 4 seedlings ca. 1.5 cm per vessel.  The experiment showed that FT medium resulted in significantly higher growth than ½ MS medium as indicated by seedling fresh weight.  The media containing tomato juice led to highest growth of seedling as shown by seedling fresh weight and seedling height. Since FT medium caused better growth than ½ MS medium, the FT medium could be used as a substitute for ½ MS medium for growing Dendrobium hybrid seedlings. The seedlings were successfully acclimatized in a shaded green house environment with 100% survival rate.

Abstract

Hybrid moth orchid (genus Phalaenopsis) is one of the most popular ornamentals in Indonesia. It has beautiful and long-lasting flowers, but cultivating this orchid is still become a challenging issues due to the need of specific condition to grow and long time period to re-blooming. Plant growth regulators (PGR) (i.e. Benzyladenine (BA)) has been widely documented as a flower-inducing substance in several orchids. However, the optimal concentration and its mechanism in inducing flower-stalk bud and re-blooming is still unclear. This research aimed to study the effects of BA application in the form of lanolin paste on hybrid Phalaenopsis flower-stalk buds. We conducted this study using completely randomized design with four replications at the greenhouse laboratory Faculty of Agriculture University of Lampung on August to December 2018. We divided the orchid into 5 group of BA concentration (0, 1000, 1500, 3000, or 6000 ppm). The sheath of the fourth or fifth buds of the flowers were carefully opened, then it smeared with BA. The percentage of bud break into flower spike or keiki, length of shoots or spike and number of open flowers were recorded until 10 weeks of observation. The results showed that, neither of the buds under the control treatment (without BA), 1000 ppm nor 1500 ppm BA broke and grew into keiki or spike. On the other hand, application of BA at 3000 ppm or 6000 ppm successfully induced 100% flower spikes on the buds treated. No keiki was formed in all buds treated. In addition, treatment of the buds with 6000 ppm BA produced longer flower spikes as well as more open flowers. We conclude that the application of BA (minimal 3000 ppm) successfully induced flower spike of  hybrid Phalaenopsis.

Abstract

Diuron herbicide has been used in the pineapple plantation in Lampung, Indonesia, for more than 35 years. It has been realized that the use of herbicides with the same mode of action intensively can speed up the evolution of resistant weeds over a long period of time. This study aimed to determine whether Dactyloctenium aegyptium from pineapple plantation has evolved resistance to diuron and to examine whether the resistance correlates with the weed physiological activities. The study was conducted at the University of Lampung, from September 2018 to March 2019. The study consisted of two stages, i.e. Stage 1: Weed resistance test and Stage 2: Physiological activity test on resistant weed. The study used a split-plot design. The main plot was the origins of weeds (exposed and unexposed to diuron) and thhe supplots was the diuron dose. The result showed that D. aegyptium exposed has high-level resistance to diuron. The physiological activities of D. aegyptium which has a high level of diuron resistance exhibited higher carbon assimilation, stomatal conductance, and transpiration rates than the sensitive D. aegyptium.

Abstract

Ubi kayu (Manihot esculenta Crantz) merupakan salah satu komoditas pangan yang memiliki nilai ekonomis.  Produktivitas ubi kayu masih tergolong rendah. Upaya yang dapat dilakukan untuk meningkatkan produksi ubi kayu adalah aplikasi auksin untuk memacu pembentukan akar karena hal yang paling penting dari ubi kayu adalah umbi yang akan di hasilkan.  Tujuan dilakukannya penelitian ini adalah mengetahui pengaruh NAA dan konsentrasi yang terbaik dalam meningkatkan jumlah akar produktif dan pertumbuhan tunas setek singkong. Penelitian ini menggunakan Rancangan Acak Kelompok (RAK) dengan perlakuan sebanyak 6 perlakuan yang diulang sebanyak 3 kali.  Perlakuan pada percobaan ini terdiri dari: A0=0 ppm NAA, A1=200 ppm NAA, A2=400 ppm NAA, A3=600 ppm NAA, A4=800 ppm NAA, dan A5=1000 ppm NAA.  Hasil penelitian yang didapatkan adalah NAA berpengaruh terhadap pertumbuhan tunas dan pengakaran setek singkong.  Konsentrasi 200-1000 ppm NAA meningkatkan pengakaran dan pertumbuhan tunas setek singkong, dimana konsentrasi terbaik adalah 1000 ppm NAA menghasilkan jumlah akar total, jumlah akar produktif, panjang akar, dan bobot segar akar tertinggi.

Abstract

Increasing cassava production is required to maintain supply sustainability and meet seed demand. The use of splice grafting and the application of auxin IBA are expected to accelerate rooting and enhance the productivity of cassava. The objective of this research is to study the effect of IBA application on the growth of cuttings and grafted seedlings of cassava. The research was conducted from October 2024 to February 2025 at the Integrated Field Laboratory of the Faculty of Agriculture, University of Lampung, Natar District, South Lampung. The experiment was designed according to randomized block design (RBD) with factorial treatments arrangement (3×2). The first factor was IBA concentration consisted of three levels, namely without IBA (A0), 1000 ppm (A1), and 2000 ppm IBA (A2). The second factor consisted of 2 types of plant materials, namely cuttings seedlings (B1) and slanted graft seedlings (B2). The results show that the application of IBA significantly increases the growth and production of cassava compared to without IBA application. IBA at 2000 ppm is the best concentration for the observed variables. The use of planting materials, whether cuttings or slanted grafts, both enhance the yield of cassava.

Abstract

Vanilla (Vanilla planifolia Andrews) is a high economic value plant within the Orchidaceae family (orchid family). Efforts to increase vanilla growth and production can be made through the application of auxins to stimulate root formation. The development of vanilla requires high-quality seedlings, as one of the indicators of successful propagation through cuttings is the formation of roots and the growth of shoots. The purpose of this research is to study the effect of various concentrations of IBA on the rooting of two-node vanilla cuttings. This study uses a completely randomized design (CRD) with 6 treatments, each repeated 3 times. The treatments in this experiment consist of: control (without IBA), 500 ppm IBA, 1000 ppm IBA, 1500 ppm IBA, 2000 ppm IBA, and 2500 ppm IBA. The observed variables include shoot length, number of opened leaves, number of primary roots, number of root tips, longest root length, fresh root weight, and dry root weight. The results showed that a concentration of 2000 ppm IBA was able to increase the number of primary roots, number of root tips, longest root length, fresh root weight, and dry root weight.

Abstract

Benzyladenine (BA) and thidiazuron (TDZ) are widely recognized as plant growth regulators that can stimulate bud activation in sugarcane. However, information regarding the synergistic effects of BA and TDZ, particularly in field-level sett treatment, remains limited. This study aimed to evaluate the effect of benzyladenine (BA) and its combination with thidiazuron (BA+TDZ) on bud emergence and seedling uniformity of sugarcane (Saccharum officinarum L.) setts. The experiment was arranged in a randomized complete block design with three replications, involving four sugarcane varieties (GP 99-8009, GP 05-17, GP 08-132, and TC 09) and three treatments: control (0 ppm), BA 50 ppm, and BA 50 ppm + TDZ 5 mg/L. Each experimental unit consisted of 20 buds from 10 two-budded setts. The results showed that the combination of BA 50 ppm + TDZ 5 mg/L significantly enhanced bud emerging percentage, plant height, and fresh and dry stem weights compared to BA alone and control. Variety had no significant effect, and no interaction was observed between variety and hormonal treatment. It can be concluded that the combination of BA and TDZ effectively stimulates bud emergence and early growth of sugarcane, indicating its potential use in seedling establishment.

Abstract

Vanda Douglas orchid is the one of tropical famous plant especially in Indonesia. The beauty of Vanda Douglas orchid is attract people’s attention as an artifial plant, cutting flower, garden decoration and public facility. The aim of research to learn more about : KNO3 fertilization, cytokinin ZPT application which is using Benzyladenine (BA) with or without Thidiazuron (TDZ), detect if there is an interaction or no interaction between application of KNO3 and ZPT cytokinin, also shoot decapitation on the flowering of Vanda Douglas orchids. Research was held in Sanggar Bunga Douglas Kelurahan Sumur Putri, Kecamatan Teluk Betung Utara, Kota Bandar Lampung started from November 2021 to February 2022. First attempt did using a Group Random Design (RAK) with three repetation. The treatment was arranged using factorial 2x5 and the first factor was KNO3 which was K0 (no fertilizer) and K1 (10 g/l of KNO3), and second factor were variant concentrate of BA and TDZ (mg/l) : no ZPT as a control, 200 BA, 400 BA, 200BA+15 TDZ and 400 BA+15 TDZ. Each treatment combination was repeating for three times and every single research consisted of 10 Vanda Douglas orchids and the number of whole plants reach 300 of Vanda Douglas orchids. Second attempt was a factorial research with Group Random Design (RAK) consisted of 2 factor. The first factor was no shoot decapitation (P0) and with shoot decapitation (P1). Second factor were variant concentrate of BA : 0, 200, 400, 200 mg/l + 15 mg/l TDZ, and 400 mg/l + 15 mg/l TDZ. Each treatment combination was repeating for three times and every single research consisted of 10 Vanda Douglas orchids and the number of whole plants reach 300 of Vanda Douglas orchids. Data analyze did by using variant analyze and if showed the significancy, it would continued by BNT test on 5% level. Observation of first attempt started on second week after KNO3 and ZPT sprayed. Observation of second attempt started in week 24 ( or week 12 after shoot decapitation). Variable that observed were : percentage of flowery plant (%) grain length (cm) and number of bud. The result of research showed that combination between KNO3 fertilization and ZPT gave the tiny effect to Vanda Douglas orchid flowering. Application of ZPT (BA or BA+TDZ), shoot decapitation and interaction between both factors was effecting to flowery plant percentage. Only for average of grain length variable and number of bud, the variant analyze did for single factor which was ZPT application. It was caused by no shoot decapitation treatment the whole plants (120 plants) wasn’t flowery. The variant analyze showed that ZPT application had real effect to grain length and bud.
