Abstract

Pulasan (Nephelium ramboutan-ake) is a unique tropical fruit, but its morphological variations have not been well characterized. This study was conducted to obtain data on the characteristics of pulasan landrace in West Java, Indonesia, and to formulate the ideotype of pulasan with superior characteristics, such as high fruit weight, sweet taste, and thick and easily peelable aril from the seed. Based on the descriptors of rambutan, 61 fruit variants were observed. The results showed that the average fruit weight was 46.0 g, with the highest at 103.5 g and the lowest at 23.1 g. The °Brix level variation of pulasan fruit was 16.8–29.6, and its vitamin C content was 14.0–24.0 mg/100 g. Five variants with superior fruit characters were identified among the sample collections used in the study

Abstract

Isolation of cDNA Fragment of Gene Encoding for Actin from Melastoma malabthricum. M. malabathricum grows well in acidic soil with high Al solubility, thereby it can be used as a model plant for tolerance to aluminum and acid stresses. Actin is housekeeping gene used as an internal control for gene expression analysis. The objective of this research was to isolate and clone the cDNA fragments of MmACT encoding for actin of M. malabathricum. Total RNA was isolated and used as the template for cDNA synthesis by reverse transcription. Four cDNA fragments of MmACT, called MmACT1, MmACT2, MmACT3, and MmACT4, had been isolated and inserted into pGEM-T Easy plasmid. Nucleotide sequence analysis showed that the size of MmACT1 and MmACT2 is 617 bp, whereas MmACT3 and MmACT4 is 735 bp. The similarity among these four MmACT is about 78%-99% based on nucleotide sequence and about 98%-100% based on amino acid sequence. Phylogenetic analysis based on amino acid sequence showed that at 1% dissimilarity, the MmACT1, MmACT2, MmACT3 and the ACT5 Populus trichocarpha are clustered in one group, while the MmACT4 is grouped with ACT9 P. trichocarpa and ACT1 Gossypium hirsutum, and these two groups are separated from actin group of monocotyledonous plants. The sequence of MmACT fragments were registered in GenBank/EMBL/DDBJ database with accession numbers AB500686, AB500687, AB500688, and AB500689.

Abstract

The classification  of  cultivated plants should meet two approaches namely the botanical (classifying based on essential systematic plant characters) and the practical approach (clustering based on the analysis of commercial properties). This study analyzed taxonomy of mango cultivars grown in Indonesia based on morphology and agronomy characters. It obtained 84 recognizable cultivars.They are grouped into eigth main cultivar-groups (e.g. Berem, Madu, Gedong, Golek, Bapang, Arumanis, Kepodang, and Kebo) and eigthteen cultivar-groups. The 'Lalijiwo' cultivars synonym with 'Thaber', 'Tabar', 'Gurih' whereas 'Arummanis' synonim with 'Gadung'. Meanwhile, the cultivar 'Kates277' which is a member of Golek cultivar main group is homonim with the cultivar 'Kates' in Arummanis cultivars main group. Furthermore, the cultivar Nanas93 (one of Madu main group cultivar) that different with  the cultivar 'Nanas71' which is categorized as member of Bapang main cultivar group.   Key words: Taxonomy of cultivated plant, Indonesian mango, morphology, agronomy characters

Abstract

Meningkatnya kesadaran di kalangan masyarakat tentang efek bahaya dari penggunaan zat pewarna sintetis membuat eksplorasi zat pewarna alami terus dilakukan. Banyak pewarna yang dihasilkan dari tumbuhan dan digunakan untuk pewarnaan tekstil. Penelitian dilakukan untuk mencari potensi dari daun harendong (Melastoma malabathricum) dalam menghasilkan pewarna alami untuk kain katun. Optimalisasi pewarnaan kain dilakukan dengan penambahan mordan berupa tawas [KAl(SO4)2.I2H2O], kapur sirih (Ca(OH)2), dan tunjung (FeSO4) saat proses fiksasi pada kain katun. Kain katun yang telah diwarnai diidentifikasi warnanya menggunakan RGB (Red Green Blue) Color Chart Reader. Kain katun yang diwarnai dengan ekstrak daun harendong menghasilkan warna Banana (#E3CF57). Pemberian mordan saat fiksasi berpengaruh terhadap warna kain. Kain yang difiksasi menggunakan tawas berwarna Khaki 1 (#FFF68F), sedangkan yang difiksasi menggunakan kapur sirih berwarna Lightgoldenrod 1 (#FFEC8B) dan kain yang difiksasi menggunakan tunjung berwarna Sgi Gray 36 (#5B5B5B). Penambahan mordan juga memberikan pengaruh baik terhadap ketahanan luntur warna pada kain katun akibat pengaruh pencucian dan paparan sinar matahari. Daun harendong berpotensi baik dalam mewarnai kain katun.
