Abstract

Oocyte vitrification is a major challenge in assisted reproductive technology. Oocyte vitrification with cumulus cells provide benefits in the process of maturation and fertilization. Vitrification leads to rapid temperature changes, therefore the decreasing in temperature could damage the cells even when the morphology was normal. Vitrification of mature oocytes is common because of its low sensitiveness towards low temperatures than immature oocytes. The aim of the research was to compare the effect of vitrification before and after in vitro maturation to the expression of hyaluronan. Maturation was operated in medium TC 50 ?L in CO2 incubators for 24 hours. Vitrification started with washing oocyte in PBS basic medium supplemented with 20% serum for 1-2 minutes, then in equilibration medium PBS + 20% serum + 10% ethylene glycol for 10-14 minutes, then transferred to 20% serum + PBS + 0.5 M sucrose + 15% ethylene glycol + PROH 15% for 25-30 seconds. Thawing was processed by submerging the oocytes in the media: 1). PBS + 20% serum + 0.5 M sucrose (K1); 2) PBS + 20% serum + 0.25 M sucrose (K2); and 3).PBS + 20% serum + 0.1 M sucrose (K3). Immunocytochemical stain was performed to evaluate the hyaluronan expression. Remmele scale index (Immunoreactive score, IRS) was used to read the result. There was no differences of hyaluronan expression in oocyte and cumulus group of K1, K2 and K3 at p< 0.05, statistically. We concluded that there was no difference of hyaluronan expression on oocyte and cumulus between vitrified oocyte of pre and post in vitro maturation which indicated that oocyte could be vitrified in the immature and mature state.

Abstract

Oocyte vitrification today became a hope to preserve fertility. Its was a major challenge because of oocyte characteristic in every phase. Immature oocytes were more sensitive to osmotic stress and the membrane wes less stable while mature oocyte have spindles that were very susceptible to temperature decrease. The study aim to compare the effect of vitrification before and after in vitro maturation to the expression TGF beta and GDF9. Oocyte of ewes divided into control groups (K0), K1 maturation prior vitrification, K2 vitrification prior maturation. Vitrification begins with washing oocytes in PBS supplemented of 20%serum for 1-2 minutes, followed by equilibration medium PBS + 20% serum + 10% ethylene glycol for 10-14 minutes, then transferred to 20% serum + PBS + 0.5 M sucrose + 15% ethylene glycol + PROH 15% for 25-30 seconds. Thawing was processed by in the media: 1). PBS + 20% serum + 0.5 M sucrose, 2).PBS + 20% serum + 0.25 M sucrose, and 3).PBS + 20% serum + 0.1 M sucrose. TGF beta expression both in oocyte and cummulus of K0 and K1 was significant statistically difference (p<0.05) compare with K2. GDF9 expression both in oocyte and cummulus of K0 and K1 was significant statistically difference (p<0.05) compare with K2. We concluded that immature oocyte give better expression of TGF â and GDF9 than mature oocyte.

Bax expression on oocyte in control group differed significantly with treatment group. Bax expression on cumulus in control group differed significantly with treatment group. Bcl-2/Bax expression ratio on oocyte and cumulus did not differ significantly in all groupConclusion: No difference Bcl-2/Bax expression ratio on oocyte and cumulus between vitrified oocyte pre and post in vitro maturation.

Abstract

The aim of this study was to analyze human sperm motility, viability, and morphology before and after cryopreservation. This true laboratory experimental study had pre and post randomized one group design. The study was conducted at the Embryology, Andrology, and Genetics Laboratory, Department of Medical Biology, Faculty of Medicine, Universitas Airlangga from August to November 2017. The eighteen samples of fresh semen were collected from male volunteers who agreed and signed the informed consent of the study. Samples were analyzed their motility, viability, and morphology before and after cryopreservation. Results of this study indicated differentiation between motility before and after cryopreservation. Cryopreservation process decreased progressive motility (42.22 + 9.46%; 17.83 + 6.24%; p< 0.0001) and increased the number of immotile spermatozoa (35.44 + 10.15%; 60.11 + 12.53%; p< 0.0001). Cryopreservation also decreased human sperm viability (73.78 + 8.91%; 40.83 + 12.89%; p< 0.0001) and morphology (10.94 + 4.96%; 7.39 + 3.90%; p< 0.0001). Cryopreservation of human spermatozoa caused the decreased of motility, viability, and morphology.

Abstract

Introduction: Allethrin is an active ingredient used in anti-mosquito drugs. Allethrin enters the body through inhalation, allegedly can disrupt the quality of spermatozoa by decreasing its amount, motility, viability and morphology. Many studies suggested that with sufficient intake of vitamin C and E would be able to minimize free radicals caused by allethrin. This study aims to investigate the effect of giving antioxidant vitamin C and E on spermatozoa quality of white rat (Rattus norvegicus) strain Wistar male with allethrin exposure.Methods: This research was a post-test design randomized control group design using Rattus novergicus male strains Wistar exposed to allethrin (LPB IPB veterinary stem cell). A total of 16 white rat strains Wistar male divided into 4 groups, one control group (without any treatment) and 3 treatment groups (given antioxidant vitamin C, vitamin E and both). The exposure of mosquito coils was performed 8 hours per day for 45 days and on the 46th day, spermatozoa was observed. The data were analyzed using ANOVA one way and BNT.Results: The results showed that treatment with vitamins affected the amount, motility and viability of rat spermatozoa exposed to allethrin. The mean amount of control group spermatozoa and treatment was 14,93, the mean spermatozoa motility control and treatment groups were 45,13 and the mean spermatozoa viability of control and treatment groups was 58,88. This is shown by the real difference between the control group and the treatment (p <0,05). Similarly, in the control group found morphological abnormalities.Conclusion: Exposure of mosquito coils 8 hours/day for 45 days decreased the spermatozoa quality. Overall, the admission of antioxidant vitamin C and E are able to maintain the quality of spermatozoa instead of one type of vitamin.

Abstract

Introduction: Chronic hepatitis B raises serious concern due to its high morbidity, such as cirrhosis and hepatocellular carcinoma, and mortality from 267 per 100,000 person-years. Up to now, medical therapy that successfully eradicates the hepatitis B virus is not available. Therapy is given in the long term for suppressing viral replication and disease progression. Nucleoside analog (NA) is a medication that is consumed orally once a day for years. Previous studies showed that patients who were treated with NA had a different quality of life (QoL) compared to naïve patients. This research aimed to analyze the QoL of chronic hepatitis B patients who consumed NA by comparing it with naïve patients at Dr. Soetomo General Academic Hospital Surabaya. Methods: Subjects were recruited consecutively from chronic hepatitis B patients at Dr. Soetomo General Academic Hospital Surabaya during 9 February – 31 May 2021. Data about sociodemographic characteristics, hepatitis B therapy history, and QoL were gained by using the SF-36 questionnaire and medical records. Subjects were divided into NA and naïve groups which responses underwent an analytical comparison. Results: NA group had significantly higher QoL in a physical component score (PCS) and mental component score (MCS), with physical functioning (PF), role limitations due to physical health (RP), role limitations due to emotional problems (RM), energy/fatigue (VT), emotional well-being (MH), social functioning (SF), and general health perception (GH) subscales having p<0.05Conclusion: The QoL of patients who were treated with NA was significantly higher than that of naïve patients in terms of physical and mental components.

Abstract

Background: Cigarrete smoke has been ascociated with many health-related problems especially in long time use. One of many health problems that are affected is the reproductive system that give negative impact in fertility. However, there are limited studies about this in acute exposure of cigarette smoke. Objective: The aim of this study is to investigate the effect of acute exposure of cigarette smoke on endometrial thickness and number of ovarian follicles in rats. Methods: Thirty adult female rats were randomly divided into 3 groups: P0, P1 and P2. The P1 and P2 groups were exposed to cigarette smoke for 15 days with dose of 2 and 4 cigarettes daily, respectively. Vaginal swab was done twice, before and after the last treatment, to determine the proestrus phase of the animals. Histological slide of uterus and ovarium using Haematoxylin Eosin staining were observed under light microscop. Number of ovarian follicles were measured, and the thickness of endometrium was meassured using CellSens software version 2.3 (Olympus Life Science). Statistical analysis of data was performed using MANOVA in SPSS software version 23 for Windows.   Results: There were no significant differences in both endometrial thickness and number of ovarian follicles between 3 groups (p>0.05). However, we found that there were a downward trend of some variables (endometrial thickness, number of primary follicles, de Graff follicles, and corpus luteum) from P0 to P2. Conclusion: We concluded that acute exposure of cigarette smoke not yet influenced the female reproductive organ, based on histological appearance of endemotrium and ovarium.

Abstract

Latar belakang : Hubungan antara indeks massa tubuh dengan kualitas semen masih kontroversial. Tujuan penelitian ini adalah menilai hubungan antara indeks massa tubuh dengan kualitas sperma di RSUD Dr. Soetomo, Surabaya, Indonesia. Metode : Penelitian ini adalah penelitian observasional yang menggunakan desain penelitian retrospektif-cross sectional. Sampel penelitian yang digunakan adalah 502 pria yang dibagi kedalam 4 kelompok berdasarkan indeks massa tubuhnya, yaitu kurus (<18.5), normal (18.5-25), kelebihan berat badan (25.1-27), and obesitas (>27). Data dianalisis menggunakan Fisher’s exact test. Hasil : Tidak ada perbedaan signifikan di antara tiap kelompok berdasarkan seluruh parameter analisis semen yang meliputi konsentrasi, motilitas, dan morfologi sperma. Kesimpulan : Tidak ada perbedaan profil analisis semen yang signifikan di antara setiap kelompok indeks massa tubuh.
