Abstract

The objectives of this study were to reveal appropriate growth models describing early growth of Kejobong goat based on Growth Hormone (GH) gene sequence analysis. A total of 35 DNA samples and 1.960 records of quantitative traits of Kejobong goat were collected. The exon 3 of GH gene was amplified and was sequenced to determine the SNP. Body weight and body measurements of the goats were taken at 0-14 weeks of age. Four non-linear growth models were applied for analysis of growth to compare growth performance of different genotypes by Non-Linear Mixed Model. A non-synonymous mutation (g1170AG) genotyped into GG, AG and AA was significantly associated with growth traits. Animals with heterozygous genotype AG showed higher growth traits than animals with homozygous genotype AA. Nonetheless, animals with homozygous genotype GG had the same growth traits with those animals with heterozygous genotype AG and homozygous genotype AA. The most fitted model for describing body weight was Von Bertalanffy model, while for describing wither height and hip height was Brody model. SNP at exon 3 of the GH gene can be used as genetic marker for improvement of growth traits of Kejobong goats.

Abstract

The aim of the current research was to analyze the population structure and expression of KISS1 gene associated with reproductive traits through Follicle Stimulating Hormone (FSH) level and DNA sequencing analysis based on KISS1 gene. Genotypes of 23 goat does consist of Kacang goats (n=7), Kejobong goats (n=8) and Senduro goats (n=8) were investigated using DNA sequencing, 16 out of 23 samples were synchronized to examine their FSH level using ELISA method. The data were analyzed using MEGA X software for DNA sequences and General Linier Model (GLM) for FSH plasma level. The phylogenetic tree showed the high homology between Indonesian native goats with other species showing a gene conservatism. A significantly higher FSH plasma levels were obtained from Kacang and Kejobong than Senduro goat (P = 0.002), litter size (LS) 3 than LS 1 (P = 0.0175), further CAATGCGCAACGCT haplotype and GA genotype at g.2459 G>A have a higher FSH plasma than other haplotypes and genotypes (P = 0.0027; P<0.0001) and are associated with high LS (3.0±0.18). Neither sample collection times nor parities have different significantly. The current trial indicated that CAATGCGCAACGCT haplotype and GA genotype at g.2459 G>A were correlated with reproductive traits.

Abstract

             Apoptosis merupakan kematian sel yang terjadi saat pengkonversian daging yang berkaitan erat dengan kualitas daging. Salah satu parameter studi apoptosis adalah perubahan pH. Analisis pH dilakukan dengan menggunakan pH meter. Penelitian ini bertujuan untuk menganalisis nilai pH pada daging itik dan ayam yang disimpan selama 8 jam pada suhu ruang. Berdasarkan penelitian ini dapat diketahui bahwa nilai pH pada daging itik secara berturut-turut sebesar 6,70±0,21; 6,66±0,13; 6,64±0,31; 6,51±0,25 dan 6,50±0,19 sedangkan pada daging ayam sebesar 5,66±0,16; 5,59±0,11; 5,56±0,25; 5,52±0,16 dan 5,45±0,16. Penyimpanan daging pada suhu ruang menyebabkan penurunan nilai pH daging itik sebesar 0,20% dan daging ayam sebesar 0,21%.         Apoptosis is a death cell that occurs when meat conversion is closely related to meat quality. One of the parameters of study apoptosis is pH change. pH analysis was performed using pH meter. This study aims to analyze the pH value of duck and chicken meat stored for 8 hours at room temperature. Based on this research it can be seen that the pH value in duck meat in a row is 6.70 ± 0.21; 6.66 ± 0.13; 6.64 ± 0.31; 6.51 ± 0.25 and 6.50 ± 0.19 whereas chicken meat was 5.66 ± 0.16; 5.59 ± 0.11; 5.56 ± 0.25; 5.52 ± 0.16 and 5.45 ± 0.16. Storage of meat at room temperature caused a decrease in the value of pH of duck meat by 0.20% and chicken meat by 0.21%.

Abstract

Objective: The aim of this study was to identified genetic diversity of muscovy duck in Central Java through blood protein polymorphisms by using electrophoresis method.Methods: Blood sample was collected from a total of 60 muscovy ducks from the districts of Demak, Magelang and Pekalongan, 20 samples each. The Electrophoresis Polyacrylamide Gel of Electrophoresis–Thin Layer Electrophoresis (PAGE-TLE) was used in this study. Parameters observed were protein of albumin (Alb), ceruloplasmin (Cp), transferrin (Tf) and amylase-I (Am-I). Gene frequency, heterozygosity (Ho) and genetic distance were analyzed by using DISPAN program.Results: The results showed that the Alb, Cp, Tf and Am-I of muscovy duck showing polymorphic characters. Gene frequency of AlbB (0.61) was higher compared to AlbA (0.39). Gene frequency of CpA (0.62) was higher than the CpB (0.38). Furthermore, gene frequency of TfA and TfB were 0.32 and 0.68, respectively as well as gene frequency of Am-IA (0.82) was higher than Am-IB (0.18). Muscovy duck population of Demak showed higher Ho value for Alb and Am-I, whereas Magelang showed higher value for Cp and Tf. Population of muscovy duck of Demak showed closer genetic distance to Pekalongan than Magelang.Conclusions: The protein blood locus of three population of muscovy duck in Central Jawa showed polymorphic characters.

Abstract

Objective: The objective of this study was to investigate the genetic diversity of Sragen Black Cattle based on D-loop sequences analysis.Methods: A total of 25 blood samples belonged to Sragen Black Cattle that had no genetic relationship within sample. The DNA genome was extracted based on the manufacturer’s standard protocol using gSYNC DNA Mini Kit (Geneaid Biotech Ltd.). D-loop gene was amplified using specific primer forward: 5’- TAGTGCTAATACCAACGGCC-3’ and reverse: 5’- AGGCATTTTGAGTGCCTTGC-3’ and then was sequenced. The sequencing result was aligned and analyzed by Molecular Evolutionary Genetics Analysis (MEGA) version 6.0 to reveal genetic distance and phylogenetic tree. Genetic diversity and haplotype were analysed by DNA Sequence Polymorphism (DnaSp) v6.12.03.Results: The results revealed that there were 11 haplotypes with Pi = 0.00675±0.00201 and Hd = 0.767±0.086. Sragen Black Cattle was divided by two cluster in phylogenetic tree with average of genetic distance was 0.0032.Conclusions: In conclusion, all of Sragen Black Cattle are on the same cluster and have closer genetic relationship to Bos indicus rather than Bos taurus with similarity level 85.76 % based on BLAST program.
