Abstract

ABSTRACT : Antioxidants are nutrient that can prevent the oxidative damage in the body. Arabica coffee and pandan is most common species of plant found in Indonesia. Arabica coffee and pandan known as plants with antioxidant activity. This study aimed to determine and compare antioxidant activity of water and ether fraction from methanolic extract of Arabica coffee leaves, pandan leaves and both combination by DPPH method. Vitamin C was used as positive control. The result show that Arabica coffee leaves have antioxidant activity better than pandan leaves and also gives big donation of antioxidant activity in their combinations. Statistical analysis showed that antioxidant activity from all sample were significanly different (p<0,05).

Abstract

Rhodamine B is one of the synthetic dyes that forbidden in cosmetics. The study was conducted to validate TLC-densitometry method for determination Rhodamin B in lipstick at Jember University area. Samples was taken by purposive sampling method. The analyte were dissolved with ethanol 70% and cromatographed on silica Gel GF 254 TLC plate using etil asetat : metanol : amoniak in the ratio (15:3:3) (v/v/v). Then, validation and determination sample can be done using TLC-densitometry with maximum wavelength 554 nm. Method was found linear over the concentration range of 6 – 32 ng with correlation coefficient of 0,9965. Spesificity showed calculation of purity and identity more than 0.99. The limits of detection and quantitation limits obtained 0.558 and 1.86 ng/spot. The precision test which includes precision repeatability and intermediate precision, amounting to RSD 6.57% and test accuracy , resulting % recovery and RSD value of 97.74 % ± 2.54% . Based on these results lipstick sample purposively sampled none containing rhodamine B and safe consumer use. Keywords: TLC - densitometry , lipstick , rhodamine B , the validation method of analysis

Abstract

Gelatin is widely used in some food products as gelling agent, including jelly soft candy. Most of commercial gelatin is derived from pork skin. Islam prohibits their followers to consume any food products containing pig derivates, including porcine gelatin. Therefore, it is necessary to develop some rapid and reliable methods for detection of porcine gelatin in jelly soft candy. FTIR method combined with chemometrics were chosen because it is rapid, easy and reliable. The purpose of this study was to classify of porcine gelatin in jelly soft candy using FTIR method combined with multivariate statistical methods (chemometrics). The classification method of chemometrics used to test porcine gelatin in commercial product of jelly soft candy was succesful and also in a good assessment with Xematest Pork. Abstract

Yam bean is one of tubers group that has many benefits because of its chemical constituents named inulin. Yam bean are produced in several regions in Indonesia such as Tegal, Bogor, Prembun, and Madura. In this research, inulin determination in yam bean tuber from Gresik East Java as one of yam bean production center, performed to obtain the plant source in Indonesia which is potential to produce inulin. Inulin determination was conducted using TLC Densitometry method that has been validated. Based on a standard curve that meets the requirements of linearity (r=0,997; Vxo=3,847%; Xp=355,407ng), the results showed that levels of inulin in yam bean tuber derived from Gresik was 12,322%±1,733% and mean recovery was 100,08% ± 1,142%. Abstract

Inulin is a polysaccharide , which is composed of fructose units. Inulin is generally found in plants family Compositae, Amirilidaceae, and Poaceae. Benefits inulin is as bifidogenic body, stimulate the immune system , relieve constipation , reduce the risk of osteoporosis . Inulin can be consumed alone as a prebiotic supplement. The purpose of this study was to develop a method for determination of inulin TLC-Densitometry in dahlia tuber extract samples (Dahlia spp L.) grown in soil media and polybag . Determination of inulin as a method of TLC densitometry using the stationary phase TLC plates Silica Gel F254, mobile phase = acetic acid glacial p.a: methanol p.a: aquabides sterille (v/v/v/v) (0,5:7,5:2) , solvent = Aquabides sterille: Ethanol 96 % p.a (3:1) v/v, apparition stains is mixed aniline in acetone 1% v/v:diphenylamine in acetone 10% b/v:phosphoric acid (5:5:1), and the wavelength () 380 nm . Validation of the method showed that the research procedures performed has had good linearity with a coefficient correlation 0.996 and Vx0 value 4.850%. This method also gives good accuracy and precision that is the percent recovery of inulin at 99,96%±0,39%. and the value of repeatability = 0,993% < 2,7% and intermediate precision RSD = 0,554% < 2,7% with limits of detection (LOD) and limit of quantitation (LOQ), respectively, 71,03 ng/spot and 236.76 ng/spot. The results showed levels of inulin in the dahlia tuber extract soil planted mediated by 86.256%±0.669% (%b/b±RSD%), and dahlia tubers grown polybag mediated by 76.146%±1.657% (%b/b±RSD%).

Abstract

Inulin is a polymer which have many benefits for health and used widely for food. The absence of research on determination of inulin content in dahlia tuber extract from different species, made this study very important to do. The purpose of this study was to determine the inulin content in dahlia tuber extract from different species. So it can be known the most effective type of dahlia to be the major source of inulin production. The method used in this study was TLC densitometry. The result showed that the inulin content in Dahlia variabilis tuber extract was 73.93 ± 1.209%, and the inulin content in Dahlia pinnata tuber extract was 66.76 ± 0.572%. Abstract

Flavonoids are a group of natural compounds with variety phenolic structures and are found in plants. The aim of this research was to study whether NIR and chemometric methods could be used to determine the flavonoid content. These methods were compared to UV-Vis spectrofotometry. Flavonoid was extracted from plant leaves by ultrasonic and maceration. NIR spectral data of selected leave extracts were correlated with flavonoid content using chemometric. In this study, the chemometric method that used for quantitative and qualitative analysis were Partial Least Square (PLS) and Linear Discriminant Analysis (LDA), respectively. The PLS R2 calibration was 0.9916499 and RMSEC was 2.1521897. In addition, the R2 of LOOCV and 2-Fold-Cross-Validation were 0.9986664 and 0.9823225, respectively. Furthermore, LDA gave accuracy of 100%. The significance of flavonoid content that have been measured by NIR and UV-Vis spectrofotometry was evaluated with paired samples t-test. In conclusion, flavonoid content that have been measured with both methods gave no significant difference. Keywords: flavonoid, chemometric, LDA, NIR, PLS

Abstract

A new compound from 5-fluorouracil (5-FU) derivatives, 1-(4-methoxybenzoyloxymethyl)-5-fluorouracil has been synthesized in a two steps reaction, alkylation (5-FU and formaldehyde to form 1-hydroxymethyl-5-fluorouracil) and esterification (1-hydroxymethyl-5-fluorouracil and 4-methoxybenzoylchloride to form 1-(4-methoxybenzoyloxymethyl)-5-fluorouracil). Reaction product purified by chromatography column, the product has been characteryzed for physical apperance is a yellowish white, crystal and melting range 168-169OC. Based on the FTIR and 1H-NMR spectra, it showed that the purified product does not contain a single compound. This fact is supported by Thin Layer Chromatography (TLC) result showing two spots for the pressure of 1-(4-methoxybenzoyloxymethyl)-5-fluorouracil and 4-methoxybenzoate acid.
