Abstract

Coffee contains bioactive compounds such as chlorogenic acid and caffeine. Chlorogenic acid is recognized as a natural antioxidant, while caffeine is widely used to reduce fatigue and drowsiness. This study aims to simultaneously determine the content of chlorogenic acid and caffeine in robusta coffee leaf powder (Coffea canephora) using Thin Layer Chromatography (TLC) densitometry. The study identified an optimal eluent composition of ethyl acetate:formic acid:distilled water (18:1:1), with maximum wavelengths of 330 nm for chlorogenic acid and 274 nm for caffeine. High linearity was demonstrated with correlation coefficients of 0.993 for chlorogenic acid and 0.994 for caffeine. Sensitivity was validated with LOD values of 100.664 ng for chlorogenic acid and 297.232 ng for caffeine, and LOQ values of 301.994 ng for chlorogenic acid and 891.697 ng for caffeine. Selectivity was confirmed as the method effectively separated chlorogenic acid, caffeine, and other compounds with a resolution (Rs) ≥ 1.5, while specificity was evidenced through purity and identity values exceeding 0.99. Precision was proven through repeatability (%RSD for chlorogenic acid: 2.94%; caffeine: 2.27%) and intermediate precision (%RSD for chlorogenic acid: 2.26%; caffeine: 2.18%). Accuracy was achieved with mean recovery rates of 102.22% for chlorogenic acid and 99.73% for caffeine. The determination of chlorogenic acid and caffeine concentrations in robusta coffee leaf powder revealed variations between samples: PTPN XII Renteng yielded 2.278% ± 0.209% for chlorogenic acid and 1.171% ± 0.821% for caffeine, while Puslitkoka Indonesia samples showed 1.238% ± 0.206% for chlorogenic acid and 0.917% ± 0.823% for caffeine. Based on validation parameter assessments, the method is deemed valid as it meets all acceptance criteria for each parameter. This research provides an efficient and accurate analytical approach for simultaneous determination of bioactive compounds in robusta coffee leaves.

Abstract

Antioxidants are compounds that can inhibit an oxidation reaction, by binding to free radicals that play a role in the emergence of several degenerative diseases. Arabica coffee (Coffea arabica) and cinnamon (Cinnamomum burmannii) are plants that have antioxidant activity so that when combined they have the potential to produce a synergistic effect. The purpose of this study was to test the antioxidant effects of the combination of methanol extract of Arabica coffee leaves and cinnamon bark in both the water and ether fractions. Antioxidant activity was determined using the DPPH method, as a positive control vitamin C was used, and antioxidant activity was expressed in the form of IC50. The results showed a synergistic effect of the two extracts where the antioxidant activity of both the water fraction and the ether fraction in the combined form was greater than the single form. The highest antioxidant activity was shown by the water fraction of the combination of methanol extract of Arabica coffee leaves and cinnamon bark in a ratio (1:2) with an IC50 value of 8.759 ± 0.050 g/mL. This antioxidant activity is lower than the antioxidant activity of vitamin C with an IC50 value of 3.267 ± 0.007 g/mL

Abstract

Perilaku Bullying dan kecanduan Gadget pada anak merupakan dua masalah serius yang memerlukan perhatian khusus dari guru disekolah, orangtua bahkan masyarakat. Bullying adalah bentuk perundungan yang umumnya sering ditemukan di lingkungan sekolah. Pelaku Bullying  bervariasi, Bullying bisa dilakukan oleh siswa kepada siswa lain atau bahkan bulyying bisa dilakukan oleh guru atau staf sekolah kepada siswa. Kecanduan gadget adalah sebuah kondisi yang dialami oleh pengguna gadjet gadget secara berlebih sehingga menimbulkan dampak buruk bagi kesehatan. Penggunaan gadjet ini biasanya digunakan secara berlebihan oleh anak dan berdampak buruk terhadap perilaku anak. Kegiatan Parenting tentang “Stop Bullying” dan “Kontroling anak dalam penggunaan gadjet agar tidak kecanduan Bermain Gadget” di TK Khodijah 112 Dadapan bertujuan untuk meningkatkan kesadaran orangtua untuk berperan penting dalam mengontrol penggunaan gajdet sehingga anaknya tidak kecanduan bermain gadjet sehingga berakibat mempengaruhi perilaku anak. Parenting ini juga bertujuan untuk pemahaman orangtua mengenai dampak negatif Bullying. Kecanduan bermain gadget dapat di cegah dengan pemanfaatan fiture “kontrol orangtua” pada gadjet. Peran aktif guru dan orangtua dalam mendukung dan mengawasi anak-anak mereka juga sangat penting untuk keberhasilan program ini.
