Abstract

BACKGROUND: The acute Hepatitis B virus (HBV) infection usually ceases before six months, but chronic infection that lasts for more than six months might develop into liver cirrhosis and hepatocellular carcinoma (HCC). Viral particle load, HBV genotypes and association to the HBV x (HBx) gene mutations are the probable factors related to HCC occurrence. The mutation which leads to HBx T118N was found as the second most common HBx mutation in Indonesia, as compared to the known cancer-related HBx K130M/V131I mutant. However, the effect of T118N mutation and its combination with K130M/V131I on human hepatoma cells has not been elucidated well. Hence, this study was conducted to dissect the role of HBx T118N and its mutant combination in colony formation, as compared to the wild type HBx and cancer-related HBx K130M/V131I.METHODS: In this study, the genes encoding wild type HBx, HBx T118N, and HBx K130M/V131I mutations were obtained as synthetic gene. Meanwhile, the gene encoding HBx T118N/K130M/V131I mutations was successfully generated using site-directed mutagenesis. The optimum condition for colony formation assays was determined through Zeocin sensitivity test of HepG2 cells.RESULTS: Selection of HepG2 cells using Zeocin was determined at 200 µg/mL. Colony formation assays performed upon expression of HBx T118N and HBx T118N/K130M/V131I mutant proteins showed reduced colony numbers as compared to the expression of wild type HBx, similar to the effect from HBx K130M/V131I mutant expression.CONCLUSION: The HBx T118N and HBx T118N/K130M/V131I mutation caused less colony formation of HepG2 cells, similar to the K130/M131I mutation. This indicates a possible role of the T118N mutation in liver cancer development.KEYWORDS: colony formation assay, hepatitis B virus, HBx, T118N, K130M/V131I

Abstract

The role of women in supporting the family economy is what is interesting. Women's roles are generally limited to domestic work, including caring for their husbands and children, cooking, and performing other household chores. Several factors encourage women to contribute to their families' economy. First, women who are left behind by divorce or the death of their husbands (widows). They need to work hard to meet the living expenses that are usually met by their husbands, especially for those with children. Given this condition, there are many programs that support women in developing their abilities, namely through women's empowerment programs. Women's empowerment programs that aim to increase women's awareness and potential to achieve independence in supporting the family economy, namely, through the empowerment program for women as heads of families using traditional medicines. This community service will last for five (5) weeks and will take place in Menampu Village, Gumukmas District. In this activity, participants will gain knowledge covering traditional medicine production and management, as well as the marketing of traditional medicine products. Community service activities through the empowerment of women heads of families (PEKKA) through traditional medicine have been implemented well. However, until this module was compiled, several stages had not been implemented, namely, continuous monitoring and assistance with traditional medicine product marketing strategies. Therefore, this activity needs to be continued so that it produces a more perfect output, namely, the independence and welfare of the Empowered Pekka Development Group.Keywords: Female heads of households, Empowerment, Traditional medicine

Abstract

The development of pharmaceutical raw materials as local Indonesian probiotic concentrates requires attention to strain identity accuracy, safety, and health potential. This is due to the high potential of probiotics in preventing and addressing various health disorders from infancy to old age. Previous research successfully obtained lactic acid bacteria (LAB) isolates as probiotic candidates from tempeh produced in Jember, and preclinical testing showed that administering a consortium of five LAB isolates at a dose of 10^8 CFU/ml to BALB/c mice was an effective treatment for further application. This study aimed to perform molecular identification of the consortium of five LAB isolates from tempeh in Jember, East Java, based on 16S rRNA gene sequences. The consortium includes TA1, TB1, TK1, TK2, and TK4 isolates. This study used both qualitative descriptive and quantitative methods. Identification based on the 16S rRNA gene to determine LAB species strains involved bacterial genomic DNA isolation, amplification of the bacterial 16S rRNA gene, PCR product analysis, and phylogenetic identification and analysis. DNA amplification results showed that the consortium of five LAB isolates produced DNA fragments with bands of approximately 1300 bp. BLAST-N and phylogenetic analysis of the 16S rRNA gene showed that TB1 had 99.05% similarity with Kosakonia cowanii JCM 10956, TA1 had 99.46% similarity with Lactiplantibacillus plantarum JCM 1149, TK1 had 99.38% similarity with Lactiplantibacillus pentosus 124-2, TK2 had 96.86% similarity with Pseudomonas fluvialis ASS-1, and TK4 had 99.38% similarity with Lactiplantibacillus plantarum NBRC 15891. TA1, TB1, and TK4 were identified as non-pathogenic LAB strains, while TK1 and TK2 were not classified as LAB.

Abstract

This study aims to understand the activity and effective dose of N-4t-butylbenzoyl Acetate compound as a pain reliever. The analgesic activity test conducted was divided into 5 treatment groups. The analgesic effect test was performed using male mice from the Balb-C strain. In each test, each group consisted of 5 experimental animals. The first group (negative control) was given 1% Na-CMC suspension; the second group (positive control) received 400 mg/kgBW ibuprofen; while the third, fourth, and fifth groups were successively given 200, 400, and 600 mg/kgBW of N-4t-butylbenzoyl Acetate compound, respectively. After the treatment was given to each test animal, an analgesic test was conducted with a 30-minute interval. After that, it was injected with 1% acetic acid and observed for 180 minutes; every 10 minutes, the movement was observed for all treatments. The analysis was carried out by comparing the number of movements (writing reflex). Data analysis was performed statistically using ANOVA (analysis of variance). The number of mouse activities produced by male mice of the Balb-C strain showed that the N-4t-butylbenzoyl acetate compound at a dose of 1.56 mg had the highest activity, % protection, and % analgesic effectiveness compared to the other two doses. Based on statistical analysis, there was no significant difference in all treatments of the N-4t-butylbenzoyl acetate compound when compared to ibuprofen (positive control)
