Abstract

Red super dragon fruit peel (Hylocereus costaricensis) extract has capability to scavenge DPPH and the activity could be maintained by using maltodextrin as coating. The aims of the research are to determine ratio of red super dragon fruit peel ethanol extract toward maltodextrin producing microcapsule with the highest coating efficiency and DPPH scavenging content by microcapsule of red super dragon fruit peel extract. Microencapsulation was carried out using freeze drying technique. Comparison of red super dragon fruit peel extract and maltodextrin were 1: 8, 1: 10, 1: 12, and 1:14 (w / w). The results show that the highest efficiency ratio of coating extract of red super dragon fruit peel with maltodextrin was 1: 8 with percentage of 90,77%. Level of DPPH scavenging content of red super dragon fruit peel extract microcapsule with ratio of 1:8 on 5 minutes and 60 minutes each is 27.36% and 46.62%. Abstract

Gembili bulbs (Dioscorea esculenta L.) have a fairly high carbohydrate content, so they can potentially be a new source of glucomannan compounds. This study aims to determine the yield and glucomannan characteristics of Gembili bulbs at different extraction times and temperatures. Glucomannan was obtained by extraction with water at temperatures of 45, 60, 75, 90, and 105oC for 45, 60, 75, 90, and 105 minutes, and precipitated with isopropyl alcohol. The results showed that the highest yield was obtained at the extraction time of 90 minutes and a temperature of 105oC of 53.09%. The glucomannan of Gembili bulb has a molecular weight of 1,865 x 104 g/mol with ash and water content values of 0.866% and 10.45%, respectively. The results of identification of functional groups with an FTIR spectrophotometer showed that there was a functional group stretching vibration of glucomannan compounds, including the –OH group at a wavenumber of 3431 cm-1 and -C-O-C (glycosidic bond) at a wavenumber of 1020.34 cm-1 and bending vibration of -CH functional groups on wavenumber of 850.61 cm-1.

Abstract

Research on the analysis of fiber and protein on cereal based on coconut pulp and skipjack tuna (Katsuwonus pelamis) flour has been carried out. The purpose of this study was to determine the ratio of coconut pulp flour and skipjack tuna flour which produced the highest fiber content and protein content in cereal processed products and to know the preference level of panelists for cereal processed products. The ratio of coconut pulp and skipjack tuna flour used in this study, namely 25:75, 50:50, 75:25 (w/w), and without the addition of coconut pulp flour and skipjack tuna flour. The results showed that cereal with the highest fiber content was obtained at a ratio of 75:25 i.e. 10.52% and the lowest fiber content was obtained at the cereal without the addition of coconut pulp flour and skipjack tuna flour i.e. 1.19%, whereas the highest protein content of cereal was obtained at a ratio of 25:75 i.e. 26.02% and the lowest protein content was obtained at the cereal without the addition of coconut pulp flour and skipjack tuna flour i.e. 9.17%. The highest preference level (5.57) of the panelists for the cereal flour of coconut pulp and skipjack tuna flour is the 75:25 ratio. Abstract

Utilizing of lycopene extract in VCO has serve as antioxidant agent that protected VCO from oxidation process during storage. Belnding of VCO and tomato lycopene extract has been done that purpose to determine changes in peroxide number in the mixture of VCO-lycopene extract tomatoes based on comparison of ratios and length of storage. Peroxide contents from the mixture of tomato extract with VCO were determined using a spectrophotometric method at a wavelength of 500 nm. The results showed that the best results were obtained at a ratio of 40: 3 (v/w) with peroxide numbers 0.08 meq O2/ kg, 1.38 meq O2/ kg, 1.84 meq O2/ kg, 3.21 meq O2/ kg, 3.46 meq O2/ kg and 3.83 meq O2/ kg respectively. Peroxide numbers increased with increasing storage time, the highest increase occurred at week 3 at a ratio of 40: 1 (v/w) which was equal to 9.62 meq O2/ kg. The addition of lycopene was better at reducing the VCO oxidation process than without the addition of lycopene. Keywords: peroxide value, VCO, lycopene, duration of storage, ratio comparison

Abstract

Research on the antioxidant activity of the local onion roots of Palu (Allium cepa Var Aggregatum L.) with various polarity solvents has been carried out. This study aims to determine the potential of Palu local shallot root extract as an antioxidant and determine the IC50 value of the Palu local shallot root antioxidant activity. The method applied in this research is maceration using n-hexane, ethyl acetate, and ethanol. The extracts from each solvent were tested for their antioxidant activity by the DPPH method. The results of the study showed that the IC50 value in n-hexane extract, ethyl acetate extract, and ethanol extract were 579.98 ppm, 518.28 ppm 315.83 ppm and as a comparison used ascorbic acid obtained IC50 of 53.69 ppm. Based on the results obtained, the best antioxidant activity is in polar solvents (ethanol extract) compared to other solvents. Abstract

The quality of Dumbo Catfish (Clarias gariepinus) oil can be maintained during storage with the addition of crude lycopene in tomatoes as an antioxidant. The purpose of this study was to obtain the best ratio between crude lycopene in tomatoes and Dumbo catfish oil during storage. The study was conducted using a variable ratio of crude lycopene: Dumbo catfish oil with 4 treatment levels (6:40, 8:40, 10:40, and 12:40 (w/v)) and storage time variables of 1, 2, 3, and 4 weeks. Lycopene levels in Dumbo catfish oil were analyzed using a UV-Vis spectrophotometer, while the quality of Dumbo catfish oil during storage was determined through peroxide value analysis. The results showed that the best ratio of crude lycopene of tomatoes: Dumbo catfish oil was 12:40 at the third week of storage with lycopene content of 0.257 grams and the peroxide value of Dumbo catfish oil was 3.45 meq/kg or had met IFOS standards. Abstract

Research about the retention of red spinach leaves extract (Alternanthera amoena Voss) as a natural coloring and preservative in wet noodles has been done. The study aim is to determine the anthocyanin retention of red spinach leaves extract in the processing of wet noodles and determine the organoleptic quality of wet noodles by adding pigment red spinach leaves extract. The extraction method used the maceration method by ethanol solvent. Anthocyanin levels on an extract that obtained by using a UV-Vis spectrophotometer were 33.34%. Anthocyanin retention of red spinach leaves extract on wet noodle processing at storage times of 0, 6, 12, 18, 24, 30, 36, 42, and 48 hours was 100%, 75.87%, 75.37%, 74.95%, 74.62%, 74.12%, 73.28%, 71.70%, and 70.61%. Organoleptic quality of wet noodles that was added with extract has “like” to “really like” grade with score detail of the best color of 4.37 at 18 hours storage, whereas the odor, texture, and taste for 0 hours storage have the highest score of 4.63, 4.63, and 4.57, respectively. Abstract

Diazinon (o, o-diethyl-o [2-isopropyl-6-methyl pyrimidynyl] phosphorotioate) is one of the organophosphate group pesticides widely used to protect cabbage plants (Brassica olarecea). Pesticide residues left in plants can bind to the cholinesterase enzyme and cause system disruption nerve. Analysis of diazinon residues in cabbage plants needs to be done to determine the level of toxicity caused. The electrochemical biosensor method as an electrochemical sensor uses a working electrode of platinum wire coated with the enzyme butybholchesteresterase (BChE) and cellulose acetate (SA) membrane and glutaraldehyde (GA) can be used for the analysis of pesticide residues. The test results of the SA electrode composition of the 10% GA 20% membrane obtained Nernst factor prices 28.1 µV / decade with a measurement range of 10-9 - 10-6 M, detection limit value of 1.29 x 10-7 M response time of 30 - 300 seconds. Abstract

Young coconut coir (Cocos nucifera Linn.) waste has the potential to be processed into a useful product, namely as a source of natural dyes, especially in cotton fabrics. Processing is carried out through a maceration process for 72 hours using 96% ethanol as solvent. The obtained extract was applied on cotton cloth with various concentrations of the extract solution, 5, 10, 15, 20, and 25%, and various soaking times of the fabric in the extract for 60, 90, 120, 150, and 180 minutes. The extract was analyzed using Uv-Vis Spekctrofotometer it showed that the ethanol extract or dye that is absorbed into the cotton fabric with various concentrations is 3.43, 4.82, 6.61, 7.79, and 9.03%, respectively. The various soaking times show that on the other hand that the extract absorbed into the fabric was successfully obtained around 7.75, 9.64, 10.81, 11.65, and 12.04%. The results show that the concentration of the extract solution and the increasing length of soaking time can increase the percentage of extract or dye absorbed in cotton fabrics

Abstract

Johar plant is a type of plant from the Fabaceae family which is widely used in traditional medicine such as malaria, itching and diabetes medicine. This study aims to determine the antibacterial activity of Johar stem bark extract with different levels of solvent polarity on the growth of Staphylococcus aureus and Escherichia coli bacteria and to determine the active compound class by TLC-Bioautography. The extraction of active compounds used a multilevel maceration method starting with n-hexane (non polar), followed by ethyl acetate (semi-polar) and ethanol (polar) solvents. Antibacterial activity testing was carried out by diffusion well method with a concentration variant of 25% and 50%. The results showed that n-hexane, ethyl acetate, and ethanol extract had antibacterial activity against the two test bacteria. Ethyl acetate and ethanol extract from Johar stem bark at a concentration of 50% were classified as very strong antibacterials with inhibition zone diameters against S. aureus, which were 22.02±0.84 mm and 20.16±0.23 mm, respectively. The results of the TLC-Bioautography test showed that the three test extracts had strong antibacterial activity against S. aureus and E. coli. In the n-hexane extract with n-hexane: ethyl acetate (9:1) eluent using Lieberman-Burchard spray reagent, it was suspected that triterpenoid compounds were present. In ethyl acetate extract with n-hexane: ethyl acetate (6:4) eluent and ethanol extract with chloroform: methanol (8:2) eluent using FeCl3 1% spray reagent, it was suspected that the tannin compound was present in both extracts.
