Abstract

Background. Andrographolide as an anti-inflammatory inhibit activation of NF-κβ, the production of TNF-α, IL-12, pressing the release inducibele nitic oxide synthase (iNOS), inhibiting the release of COX-2 in human fibroblast cells and also prevents the production of oxygen radicals. TNF- α stimulates the migration of immune cells to get to the site of infection, contribute to the formation of granulomas, and can control the disease progresificity. Methods. The study aims to analyze the effect of andrographolide on the expression of TNF-α and tuberculosis granuloma in mice infected with CFA. Laboratory tests in the laboratory of histology and pathology anatomy medical faculty of UNS. Samples are 30 individuals’ mice, consist of 10 rats as control, 10 rats injected with CFA, and 10 rats injected with CFA+ andrographolide. Histopathology and immunohistochemistry of lung tissue granuloma examinated in the laboratory of pathology anatomy medical faculty of UNS. Results. The study design was purely experimental or (true experiment, with randomized post test only control group design). Andrographolide lowering the average expression of TNF-α compared to the CFA group, based on the description of the variable expression of TNF-α. The results of the analysis of three different variations or the average using Kruskal Wallis test showed that there are differences in the average number 3 granuloma with P=0.003. It means different average number of granulomas in the control group, CFA and CFA+ Andrographolide completely different convincingly. Compared with the average number of granulomas in the control group, the group CFA has a tendency average number of granuloma higher (increased), then the average number of granuloma in group CFA + Andrographolide has an average lower than the group CFA or mean average granuloma amount can be reduced by giving Andrographolide paniculata extract. Conclusion. Andrographolide 14.8% paniculata extract shown to decrease the expression of TNF-α induction of pulmonary tuberculosis in mice the CFA. Andrographolide 14.8% paniculata extract shown to reduce the number of lung granuloma in mice induced tuberculosis CFA. There is a positive correlation between the expression of strong TNF-α by the number of granulomas in the lungs in mice induced tuberculosis CFA. (J Respir Indo 2018; 38(2): 75-82)

Abstract

BACKGROUND: Anthrax is a zoonotic disease caused by Bacillus anthracis, whose endospores stimulate the release of pro-inflammatory cytokines and promote oxidative stress. Propolis, a natural resource that can be found in Indonesia, has been proven to have anti-apoptotic and antioxidant role which might be a potential adjuvant therapy for anthrax treatment. Hence in this study we aimed to investigate effect of propolis as an anti-inflammatory, anti-apoptotic, and antioxidant in anthrax rats model by examining the level of tumor necrosis factor (TNF)-α, caspase-3, and malondialdehyde (MDA), respectively.METHODS: This was an experimental post-test only study with 40 male rats weighed 180-200 g that were induced by anthrax spores injected subcutaneously. The rats were divided into one control positive group and four intervention groups that were administered with 200 mg/kgBW propolis extract for 7 to 14 days. The levels of serum TNF-α, caspase-3, and MDA were measured using enzyme-linked immunosorbent assay (ELISA) and analyzed with bivariate analysis.RESULTS: TNF-α, caspase-3, and MDA level were found lower in anthrax rats model given ethanol extract of propolis than the control group. The lowest concentration of TNF-α value was found in group administered with propolis extract 200 mg/kgBW for 7 days before the anthrax induction (6.136±0.205 pg/mL). The results were similar to the MDA serum and caspase-3 which were the lowest when the propolis was administered 7 days earlier (1.893±0.188 nmol/mL and 2.040±0.067 ng/mL). There was significant difference in the TNF-α, caspase-3, and MDA serum levels (p≤0.001) compared to control group.CONCLUSION: Propolis has anti-apoptotic and antioxidant effects which can be used as complementary therapy in anthrax infection.KEYWORDS: propolis, anthrax, anti-apoptotic, antioxidants

Abstract

Introduction. The relationship between serum uric acid (SUA) level and ischemic heart disease abides controversial and still has not been established as a cardiovascular risk factor. The cooperative interaction between those two factors is not fully understood. Prior epidemiological evidences of the causal relationship is still argumentative. There were various studies using the same methods yet the outcome were different. This study aims to conduct a meta-analysis to synthesize the results of recent studies in order to obtain data quantitatively and also accurately. Methods. This study protocol was registered with PROSPERO (CRD42020210948), and conducted according to PRISMA guidelines, tracing studies published in vulnerable periods from January 2010 to May 2020. The Cochrane Library, EBSCO, Medline/PubMed, ProQuest and Science Direct are sources of published studies. Meta-analysis was conducted to synthesize the associations between SUA level and severity of coronary artery stenosis, using random effect model to account for possible study heterogeneity. Heterogeneity was assessed using I2 , and the meta-analysis was performed using comprehensive meta-analysis version 3 (CMA3) software. Results. Five studies (n = 601 patients) identified a correlation between serum uric acid levels and Gensini scores (r = 0.548; p <0.001) in ACS patients. Heterogeneity bias was found in the analysis, whereas publication bias was not found. There was a moderate positive correlation between serum uric acid levels and the severity of coronary artery stenosis, with a correlation coefficient of 0.548 (p value <0.001).
