Abstract

Gene encoding GRA1 protein is potent DNA-vaccine candidate against toxoplasmosis. The aim of the researchwas to clone the gene encoding GRA1 protein of tachyzoite Toxoplasma gondii local isolate by DNA recombinanttechnology. Tachyzoite was grown in Balb/c mice in vivo. Messenger RNA was isolated from total RNA and itwas used to synthesis cDNA. Complementary DNA encoding GRA1 protein of tachyzoite Toxoplasma gondii localisolate was amplified and cloned in a prokaryote cloning vector. The recombinant GRA1-encoding gene was thendigesting using EcoRI restriction endonuclease and sequencing. The result showed that the recombinant GRA1-encoding gene consisted of DNA sequences encoding all signal peptide and mature peptide of GRA1 protein.Alignment of recombinant GRA1 sequence to gene encoding GRA1 protein of Toxoplasma gondii RH isolate showed100% homologous.Keywords: GRA1 protein, Toxoplasma gondii, tachyzoite, cloning, cDNA

Abstract

Rhoptry protein belongs to an excretory and secretory antigens (ESAs) that play an important role during activepenetration of parasite into the cell target. This protein an able Toxoplasma gondii to actively penetrate targetedcell, meanwhile ESAs protein stimulates intracellular vacuole modification. It is, therefore, after the parasitesuccessfully enter the cell target then Granule (GRA) proteins are responsible for the formation of parasitophorusvacuole, which is protect the fusion with other intracellular compartments such as lysosomal vacuole. Consequently,this parasite is being able to survive and multiply at the cell target. The current study was aimed to clone andsequens cDNA encoding for ROP-2 of local isolated T. gondii tachizoite through DNA recombinant technique.Total ribonucleic acid (RNA) was isolated from tachyzoites of local isolated T. gondii that were grown up in Balb/c mice. Messenger RNA was isolated from total RNA using PolyAtract mRNA Isolation System. Messenger RNA wasused as a template for synthesis cDNA using Riboclone cDNA Synthesis System AMV-RT. EcoRI adaptor fromRiboclone EcoRI Adaptor Ligation System was added to Complementary DNA and than ligated to pUC19. Recombinantplasmid was transformed into E. coli (XL1-Blue). The transformed E. coli XL-1 Blue were plated on LB agarcontaining X-Gal, IPTG and ampicillin. Recombinant clones (white colony) were picked up and grown up in theLB medium at 37oC overnight. Expression of recombinant protein was analysed by immunoblotting in order toidentify cDNA recombinant wich is express ESA of T. gondii local isolate. Recombinant plasmid were isolatedusing alkalilysis method and were elektroforated in 1% agarose gel. The isolated DNA recombinant plasmid wascut using Eco RI and then sequenced through Big Dye Terminator Mix AB1 377A Sequencer using M13 Forward andM13 Reverse primers. The conclusion of this results showed that the recombinant clone was coding for excretoryand secretory protein which has molecular weight of 54 kDa. The DNA alignments of sequence from the clonedgene showed 97% homology with gene encoding for ROP-2 of T. gondii RH isolate.Keywords: Toxoplasma gondii, tachizoite, ESA, complementary DNA, ROP2

Abstract

Penggunaan kortikosteroid dosis rendah pada penatalaksanaan sepsis tahap awal masih diperdebatkan. Soluble receptor for advanced glycation end products (sRAGE) merupakan biomarker keparahan dan keluaran yang buruk pada sepsis. Penelitian ini bertujuan untuk menganalisis efek penggunaan kortikosteroid dosis rendah terhadap kadar sRAGE serum pada mencit Balb/C model sepsis tahap awal. Penelitian ini merupakan penelitian eksperimental laboratoris dengan sampel 30 ekor mencit Balb/C jantan dibagi menjadi kelompok kontrol, sepsis, dan sepsis+kortikosteroid dosis rendah. Penelitian dilakukan di laboratorium Histologi dan Biomedik Fakultas Kedokteran Universitas Sebelas Maret Surakarta periode Juni-Desember 2013. Untuk membuat model sepsis, hewan coba diinokulasi lipopolisakarisa/LPS (E. coli) dosis 0,1 mg/mencit secara intraperitoneal (i.p.). Kelompok mencit kontrol tidak diinokulasi selama penelitian. Kortikosteroid dosis rendah yang digunakan adalah metil prednisolon dosis 0,05 mg/mencit/hari secara i.p. Mencit kontrol tidak diinokulasi selama penelitian. Kadar sRAGE kelompok kontrol 54,29±16,28 pg/mL, sepsis 78,12±13,38 pg/mL, dan kortikosteroid dosis rendah 63,39±11,07 pg/mL. Kortikosteroid dosis rendah secara bermakna menurunkan kadar sRAGE (p=0,044) dibanding dengan kelompok sepsis. Simpulan, penggunaan kortikosteroid dosis rendah menurunkan kadar sRAGE pada hewan coba model sepsis tahap awal. Soluble receptor for advanced glycation end products (sRAGE) is a biomarker of severity and poor outcome of sepsis. This study aimed to analyze the effects of the use of low-dose corticosteroids on sRAGE serum levels in Balb/C mice model of early sepsis. This study was an experimental research laboratory study with 30 male Balb/C mice which divided into control, sepsis and sepsis+low-dose corticosteroids groups. The study was conducted at Histology and Biomedical Laboratory, Faculty of Medicine, Sebelas Maret University, Surakarta, from June to December 2013. Sepsis was induced in the male Balb/C mice by inoculation with an intraperitoneally (i.p.) injection of lipopoly-saccharide/LPS (E. coli) with a dose of 0.1 mg/mice/i.p.for sepsis mice model. Control mice were not inoculated during the study. Low-dose corticosteroids used was methyl prednisolone at a dose of 0.05 mg/mice/day/i.p. Levels of sRAGE  54.29±16.28 pg/mL in control group, 78.12±13.38 pg/mL in sepsis group, and 63.39±11.07 pg/mL in low-dose corticosteroids group. Low-dose corticosteroids significantly decreased sRAGE level (p=0.044) compared to the sepsis group. Abstract

Jamblang (Syzygium cumini L.) leaves contain several phytochemical compounds. These phytochemicals are thought to have roles as antihyperglycemic, antihyperlipidemic, and antioxidant agents. This study aims to prove that there is an effect of giving ethanolic extract of jamblang leaves on body weight (BW) of metabolic syndrome (MS) Wistar rats induced by high-fat high-fructrose diet (HFFD) and injection of Streptozotocin (STZ)-Nicotinamide (Na), to determine different doses effect on the weight of Wistar rats. Laboratory experimental research with pre-post test control group. Samples were 8 weeks male Wistar (Rattus noverigicus) weighing 150-200 grams chosen by purposive random sampling method. Thirty rats were divided into 5 groups, each consisting of 6 rats. Normal group, metabolic syndrome group, 3 treatment groups MS were given jamblang leaf extract 100 mg/KgBW, 150mg/KgBW, 200mg/KgBW for 28 days. Data were analyzed by one way ANOVA test and repeated ANOVA test. The study showed an ethanolic extract of Jamblang leaves can reduce weight loss in Wistar rats with a metabolic syndrome model, and that the best dose used in this experiment is 150 mg/kgBW per day in Wistar rats with a metabolic syndrome model.

Abstract

Background: There has been an increasing prevalence of metabolic syndrome (MS) caused by life style such as sedentary behavior and western diet. Metabolic syndrome causes degeneration in ren’s structure and processes the elimination of by product in metabolism, which is uric acid. Moringa oleifera Lam. leaves contain antioxidant that can repair damages caused by MS. Studies about improvement of ren’s structure and uric acid level related to Moringa oleifera Lam. leaves’ consumption has not been found yet. Therefore, this study was intended to examine the effects of ethanolic extract in Moringa oleifera Lam. leaves to uric acid and glomerular surface area in male wistar rat (Rattus norvegicus) with induced metabolic syndrome model.Method: This study is an experimental laboratory study. The subjects of this study consisted of 30 rats which were divided into 5 groups with 6 in each group. K1 is control group, K2 is MS group, and K3, K4, and K5 are MS groups given variety of ethanolic extract doses. The induction of MS was done by giving high-fat diet in 28 days and injection of streptozotocin-nicotinamide (STZ-NA) in the 25th day. Rats in group K3, K4, and K5 were given doses of 150, 250, and 350 mg/kgBW in 28 days.Results: The administration of high-fat diet for 28 days and injection of STZ-NA caused MS condition in rats.  Repeated ANOVA and One-Way Anova test showed that the administration of ethanolic extract in Moringa oleifera Lam. leaves with doses of 150, 250, and 350 mg/kgBW in 28 days decreased uric acid significantly (p= 0.001; p=0.001; p= 0.001). Another result also found that ethanolic extract from Moringa oleifera Lam. leaves with doses of 250 and 350 mg/kgBW increased area of glomerular surface area in rats significantly.Conclusion: The administration of ethanolic extract from Moringa oleifera Lam. leaves with doses of 150, 250, and 350 mg/kgBW for 28 days decreased uric acid level in rats. Abstract

Introduction: Metabolic syndrome is characterized by hypertension, central obesity, insulin resistance, and dyslipidemia. Hylocereus polyrhizus is known for its potential in treating metabolic syndrome. Method: Laboratory experimental study with pretest-posttest control group design. Thirty rats were divided into 5 groups: K1 and K2 as negative and positive control, K3, K4, and K5 as groups receiving red dragon fruit extract at doses of 60, 120, and 180 mg/200 gBW. Data were analyzed using one-way ANOVA, Tukey HSD test, paired T-test, and Pearson correlation. Results: The highest HDL levels were observed in K5, while the lowest was in K1. Pearson correlation revealed a strong relationship between the dose of Hylocereus polyrhizus and HDL levels. Conclusion: Hylocereus polyrhizus extract could significantly increase HDL level. In this study, the optimal dose was 180 mg/200 gBW.

Abstract

Sindrom metabolik ditandai oleh hipertensi, obesitas sentral, resistensi insulin, dan dislipidemia, dan dapat meningkatkan risiko stroke serta penyakit jantung koroner. Kehadiran dan tingginya kadar enzim SGPT dan SGOT dalam darah menunjukkan adanya gangguan fungsi hati. Pemberian ekstrak daun Moringa berdampak pada penurunan nilai serum SGPT dan SGOT pada tikus Wistar dalam model sindrom metabolik. Penelitian eksperimental laboratorium dengan desain pretest-posttest dan kontrol posttest hanya. Sebanyak 30 tikus Wistar dibagi menjadi 5 kelompok sampel, yaitu: P1 sebagai kontrol normal, P2 sebagai kontrol sindrom metabolik tanpa pemberian ekstrak etanol daun Moringa, P3, P4, dan P5 sebagai kelompok sindrom metabolik yang diberikan ekstrak etanol daun Moringa selama 28 hari (mulai dari hari ke-28 hingga ke-56) dengan dosis bertingkat 150 mg/KgBB, 250 mg/KgBB, dan 350 mg/KgBB per hari. Induksi sindrom metabolik dilakukan dengan pakan tinggi lemak selama 28 hari pertama dan injeksi STZ-NA dilakukan pada hari ke-25. Analisis statistik yang akan digunakan adalah ANOVA satu arah dan post-hoc Tukey HSD untuk data yang terdistribusi normal, serta menggunakan uji Kruskal-Wallis untuk data yang tidak terdistribusi normal diikuti oleh uji Mann-Whitney. Data penelitian adalah data kuantitatif deskriptif. Pemberian ekstrak etanol daun Moringa 150 mg/kg BB/hari (kelompok P3); 250 mg/kg BB/hari (kelompok P4) dan 350 mg/kg BB/hari (kelompok P5) menghasilkan penurunan kadar SGPT dan SGOT, dan dosis 350 mg/kg BB/hari memberikan hasil terbaik. Pemberian ekstrak etanol daun Moringa dapat menurunkan kadar SGPT dan SGOT dalam plasma darah.

Abstract

Penelitian Penelitian ini bertujuan untuk mempelajari apoptosis sel germinal pada ductus seminiferous dan pengaruh dosis ekstrak etanolik akar kelor terhadap spermatogenesis pada sindrom metabolik. Enzim caspase-3, yang berperan sebagai eksekutor utama apoptosis, digunakan untuk mengukur tingkat apoptosis yang terjadi. Apoptosis sel germinal yang tinggi pada ductus seminiferous menunjukkan peningkatan infertilitas. Penelitian eksperimental laboratorik ini menggunakan 30 tikus Wistar model sindrom metabolik yang diinduksi dengan diet tinggi lemak dan STZ-Na, serta 6 ekor kontrol normal. Kelompok perlakuan menerima ekstrak etanolik akar kelor dengan dosis berbeda (150, 250, dan 350 mg/kgBB/hari). Hasil penelitian menunjukkan penurunan ekspresi caspase-3 pada kelompok dengan dosis 150 mg/kgBB/hari (K3), sedangkan dosis 250 mg/kgBB/hari (K4) dan 350 mg/kgBB/hari (K5) meningkatkan ekspresi caspase-3. Selain itu, terjadi peningkatan kerusakan sel germinal dan penurunan spermatogenesis pada kelompok sindrom metabolik yang diberi ekstrak akar kelor, yang tercermin pada penurunan nilai Johnsen score. Peningkatan caspase-3 dan penurunan skor Johnsen menunjukkan peningkatan apoptosis dan penurunan spermatogenesis, yang berpotensi meningkatkan infertilitas.

Abstract

Pendahuluan: Sindrom metabolik ditandai dengan hipertensi, obesitas sentral, resistensi insulin, dan dislipidemia. Hylocereus polyrhizus dipercaya dapat mengobati sindrom metabolik. Metode: Penelitian eksperimental laboratorik dengan desain pretest dan posttest control group. Sebanyak 30 tikus dibagi menjadi 5 kelompok: K1 dan K2 kontrol negatif dan positif, kemudian K3, K4, K5 kelompok yang diberi ekstrak Hylocereus polyrhizus dengan dosis berturut-turut 60, 120, 180 mg/200 gBB. Data dianalisis dengan uji one-way ANOVA, uji Tukey HSD, uji T berpasangan, dan uji korelasi Pearson. Hasil: Kadar HDL tertinggi terdapat pada K5 dan terendah pada K1. Uji korelasi Pearson menunjukkan dosis Hylocereus polyrhizus mempunyai hubungan sangat kuat dengan peningkatan kadar HDL. Simpulan: Dosis ekstrak Hylocereus polyrhizus 60, 120, 180 mg/200 gBB dapat meningkatkan kadar HDL secara signifikan. Pada penelitian ini, dosis paling optimal adalah 180 mg/200 gBB.
