Abstract

Background: Diabetes mellitus is a chronic metabolic disease that causes increased free radical activity in the body. Excessive free radicals contribute to diabetes complications. The free radicals will react with lipids and then they can form malondialdehyde. This free radicals can be neutralized by antioxidants. Breadfruit (Atrocarpus altilis F.) leaf contains flavonoids and phenolics which serve as antioxidants. This research aimed to determine the effect of breadfruit leaf extract on malondialdehyde levels in diabetic rats. Methods: This was an experimental research using posttest only control group design. Subjects were 30 male white rats, three months old with average body weight of 160-200 g. Subjects were randomly divided into five treatment groups: Normal group, negative control group, treatment group I, treatment group II, and treatment group III. All subjects of the groups, except normal group, were induced by 65 mg/kgBW streptozotocin (STZ) and 230 mg/kgBW nicotinamide (NA). Each subjects in normal and negative control group were administered 2 ml/kgBW/day Natrium Carboxymethyl Cellulose (Na-CMC), whereas treatment groups I, II, and III were administered breadfruit leaf extract at doses of 200 mg/kgBW/day, 400 mg/kgBW/day, and 800 mg/kgBW/day, respectively. The treatments were done for 14 days. Malondialdehyde levels were measured from blood samples collected via retro-orbital veins. Data were analyzed using Kruskal-Wallis test followed by Mann-Whitney U tests (a=0.05). Results: Kruskal-Wallis test showed a significant difference of MDA levels among five groups (p<0.001). Statistically significant differences of MDA levels were further found in all pairs of two-group comparisons, except between the negative control group and the treatment group I (p=0.690). Conclusion: Breadfruit leaf extract could decrease malondialdehyde levels in diabetic rats induced by STZ-NA. The dose of 800 mg/kgBW/day of breadfruit leaf extract showed the greatest effect. Abstract

Introduction: Low Density Lipoprotein (LDL) cholesterol level related with Coronary Heart Disease (CHD) which is the most common death cause in the world. Beans contains flavonoid, phytosterol, saponin, niasin, vitamin C, and fiber which have hypolipidemic effect. This study aimed to determine the effect of beans extract to the LDL cholesterol level in white rats hypercholesterolemia model. Methods: This study was a laboratory experimental research using the posttest only controlled group design, that have done in Histology Laboratorium of Sebelas Maret University Surakarta. Samples of research were 30 male white rats, Wistar strain. Control Group (KK) was given pellets and aquades for 28 days. Group 1 until 4 (KP1-4) were given duck egg yolk 5 gram/200 gram BW white rat/day and PTU 0,01% for 28 days. On days 15-28, Group 2 (KP2) was given simvastatin 0,72 mg/200 gram BW white rat/day, Group 3 was (KP3) given beans extract 100 mg/200 gram BW white rat/day, and Group 4 (KP4) was given beans extract 150 mg/200 gram BW white rat/day. The datas of LDL cholesterol level were analyzed with One-way ANOVA test and Post Hoc test. Result: Result of this study shows that LDL cholesterol levels of KK = 33.83 ± 14.442 mg/dl, KP1 = 54.00 ± 27.943 mg/dl, KP2 = 18.83 ± 7.672 mg/dl, KP3 = 77.33 ± 20.954 mg/dl and KP4 = 80.17 ± 31.923 mg/dl. The statistical analysis with One- way ANOVA shows a significant difference in LDL cholesterol level among the five groups of treatment with p = 0.000 (p < 0.05). Post Hoc Test shows a significant difference in LDL cholesterol level between KP1 – KP2 with p = 0.011 (p < 0.05) and non significant difference in LDL cholesterol level between KK – KP1 with p = 0.129, KP1 – KP3 with p = 0.081, and KP1 – KP4 with p = 0.052 (p > 0.05). Conclusions: The beans extract (Phaseolus vulgaris L.) does not give any effect to decrease of the LDL cholesterol level in white rats (Rattus norvegicus) hypercholesterolemia model. Abstract

Introduction: Coronary Heart Disease is the disease that often suffered by Indonesian people which known with high cholesterol rate.Brotowali’s stem contains saponin, flavonoid and polifenol, which potential to inhibit blood  total cholesterolincrease. The research is for to know more the influence of the content of the brotowali’s stem extract against total blood cholesterol levels. Method: This  research is as experimental laboratoric research with Randomized Controlled Trial design that post-test only control group design, have done in Histology Laboratorium of the Sebelas Maret University, Surakarta. The subject are 32 male white rats, Wistar strain, aged 2 months, weighing more or less than 200 grams. Brotowali’s stem extract made by using maceration method in Biological Pharmacy Laboratory, Pharmacy Faculty, Gadjah Mada University. The rat were divided randomly into 4 groups, each group consisting of 8 rats. In the negative control group was only given regular feed, propiltiourasil (PTU) and distilled water, the positive control group was given 5 gramduck egg yolk per 4,5 ml of aquadest, regular feed, PTU and distilled water. Whereas in the treatment group I and II was given 5 gram/4,5 ml of duck egg yolk suspense, regular feed, PTU, water and given 50 mg/200 gr and 100 mg/200gr extract of the brotowali’s stem. Rats were fasted during 12 hours and then their blood cholesterol levels checked after the 21 days treatment period. The sample measured with TMS 24i Premium spectrophotometer with 600 nm wave. The data were analyzed by one-way ANOVA with SPSS for Windows version 22. Result:Total cholesterol levels of negative control group is 91,00±19,46 mg/dl,on the positive control group is 104,12±13,66 mg/dl,on treatment group I is 91,50±16,08 mg/dl and on treatment group 2 is 84,62±13,45 mg/dl.The result of  one-way ANOVA test showed there is no significant total cholesterol blood level difference between 4 groups of rats with p=0,120. Conclusion:Brotowali’s stem extract has not give an effect on total cholesterol blood levels of Rattus norvegicus with high cholesterol intake. Abstract

Asetaminofen, hepatotoksin yang menimbulkan kerusakan sel hepar. Kerusakan tersebut akibat terbentuknya metabolit reaktif N-asetil-p-benzo kuinon (NAPQI) pada dosis besar tidak diikat seluruhnya oleh enzim glutation-s- transferase. Zat NAPQI me-rupakan radikal bebas yang menyebabkan peroksida lipid dengan peningkatan malodialdehide (MDA). Akupunktur titik paraver-tebral thorax (Th) VIII-X sedermatom hepar sehingga memper-baiki mikrosirkulasi hepar. Tujuan penelitian ini adalah menge-tahui pengaruh akupunktur titik paravertebral thorax VIII-X ter-hadap kadar MDA darah kelinci yang diinduksi asetaminofen per oral. Penelitian eksperimental yang menggunakan 30 ekor kelinci. galur Australia, umur 3 bulan, dibagi 3 kelompok: K1-kontrol, KII (asetaminofen 1,05 gram/1,5 kg BB, 3 kali selama 3 minggu), K III (asetaminofen 1,05 gram/1,5 kg BB, 3 kali dalam 3 minggu dan akupunktur 2 hari sekali (10 kali). Pemeriksaannya dilakukan 48 jam setelah penelitian berakhir. Data dianalisa menggunakan ANOVA dan LSD. Hasilnya menunjukkan perbedaan bermakna (p<0,05) kadar MDA (p=0,000) semua kelompok, akupunktur me-nurunkan MDA (K3 = 0.703 m mol/dl) dibanding asetaminofen (K2 = 0.987 m mol/dl). Akupunktur titik paravertebralis thorax VIII-X dapat menormalkan secara ber-makna MDA dan kolesterol darah kelinci yang diinduksi asetaminofen.

Abstract

Penggunaan kortikosteroid dosis rendah pada penatalaksanaan sepsis tahap awal masih diperdebatkan. Pemberian kortikosteroid dosis rendah pada sepsis dapat mengurangi respons inflamasi sitemik, menghambat produksi sitokin proinflamasi dan mediator inflamasi, serta menurunkan adhesi leukosit ke endotel. Pro-adrenomedullin (pro-ADM) merupakan biomarker keparahan sepsis. Penelitian ini bertujuan untuk menganalisis efek penggunaan kortikosteroid dosis rendah pada kadar pro-ADM serum pada mencit Balb/C model sepsis tahap awal. Penelitian ini merupakan penelitian eksperimental laboratoris, dengan sampel 30 ekor mencit Balb/C jantan yang dibagi menjadi kelompok kontrol, sepsis, dan sepsis+kortikosteroid dosis rendah. Penelitian dilakukan di Laboratorium Histologi dan Biomedik, Fakultas Kedokteran, Universitas Sebelas Maret Surakarta, selama periode April?September 2012. Untuk membuat model sepsis, hewan coba diinokulasi lipopolisakarisa/LPS (E. coli) dosis 0,1 mg/mencit secara intraperitoneal (i.p). Kelompok tikus kontrol tidak diinokulasi selama penelitian. Kortikosteroid dosis rendah yang digunakan adalah metil prednisolon dosis 0,05 mg/mencit/12 jam secara i.p. Mencit kontrol tidak diinokulasi selama penelitian. Kadar pro-ADM serum ditentukan menggunakan enzyme-linked immunosorbent assay (ELISA). Uji kadar pro-ADM serum menggunakan analysis of variance (ANOVA) dan untuk menentukan perbedaan kemaknaan digunakan p<0,05. Kortikosteroid dosis rendah secara bermakna menurunkan kadar pro-ADM dibandingkan dengan kelompok sepsis (33,0±2,9 pg/mL vs 48,5±6,1 pg/mL; p<0,001). Simpulan, penggunaan kortikosteroid dosis rendah menurunkan kadar pro-ADM pada hewan coba model sepsis tahap awal. Low-dose corticosteroids in sepsis can reduce the systemic inflammatory response, inhibit the production of pro-inflammatory cytokine, inflammatory mediators, and decrease adhesion of leukocytes to the endothelium. Pro-adrenomedullin (pro-ADM) is a biomarker of sepsis severity. This study aimed to analyze the effects of the use of low-dose corticosteroids on the levels of pro-ADM in the serum in Balb/C mice model of sepsis at the early stage. This study was an experimental research laboratory, with 30 male Balb/C mice which were divided into control, sepsis, and sepsis+low-dose corticosteroids groups. The study was conducted at the Histology and Biomedical Laboratory, Faculty of Medicine, Sebelas Maret University, Surakarta, during the period of April to September 2012. Sepsis was induced in the male Balb/C mice by inoculation through an intraperitoneally (i.p) injection of lipopolysaccharide/LPS (E. coli) with 0.1 mg/mice/i.p. dose for sepsis mice model. Control mice were not inoculated during the study. Low-dose corticosteroids used was methyl prednisolone at a dose of 0.05 mg/mice/12 hours/i.p. Pro-ADM serum level was measured with enzyme-linked immunosorbent assay (ELISA). One way analysis of variance (ANOVA) for pro-ADM serum level and p<0.05 were used to determine the significant differences. Low-dose corticosteroids significantly decreased pro-ADM level (33.0±2.9 pg/mL vs 48.5±6.1 pg/mL, p<0.001) compared to the sepsis group. In conclusion, the use of low-dose corticosteroids reduce levels of pro-ADM in the early stages of sepsis. Abstract

Background: Polycystic Ovary Syndrome (PCOS) is an endocrine disorder that most often causes infertility. It occurs due to the failure of folliculogenesis, thus causing non-ovulation. Letrozole is a 3rd generation drug of non-ste­roidal aromatase inhibitors, which can increase ovulation rates. Electroacupuncture is the insert­ion and manipu­lation of needles into acupunc­ture points (acu­points) that are selected speci­fically based on the meridian system and con­nected to the electrode stimulator. This study aimed to determine the differences of the effects of letrozole and letrozoleelectroacupuncture (letrozole-EA) on the improve­ment of folliculo­genesis in women with PCOS.Subjects and Method: This was an analytic observational study with a case-control design. This study was conducted at the Gynecology Polyclinic and Sekar Clinic, Dr. Moewardi Hos­pital, Surakarta. A sample of 30 infertile women aged 20-45 years was selected by fixed disease sampling. The criteria of the PCOS were deter­mined based on Rotterdam criteria. The depen­dent variable was the diameter of the follicle. The independent vari­ables were the administration of letrozole and letrozole-EA. Transvaginal ultra­sound of Voluson P6 was used to measure the folliculo­genesis. The data were analyzed by independent t-test.Results: On the second day after treatment, the diameter of the follicles in the letrozole-EA group (mean= 6.92; SD= 1.60) was bigger than the letrozole group (mean=5.66; SD=1.11), and it was statistically significant (p=0.004).Conclusion: Combination of letrozole and letrozole-EA therapy is more effective in increasing follicular diameter development than letrozole therapy only.Keywords: polycystic ovary syndrome, folli­culogenesis, Letrozole, electro-acupunctureCorrespondence: Yulyanti. Abstract

The incidence of COVID-19 in Indonesia is significantly increasing. In Indonesia, there were 965.283 confirmed cases and 27.453 people died of covid as of January 22, 2021. Meanwhile, the 7.505 confirmed cases and 352 death were from Surakarta. In this pandemic situation, the community must protect itself from being infected by the virus. Prevention is done by implementating the 5M health protocol, which are wearing mask, social distancing, and hand washing. Those health protocols are also part of clean and healthy lifestyle (PHBS). Special needs children (ABK) are those who have different characteristics than any other common children. Special health consideration should be given for those special needs child, including PHBS because of the special needs they are having and probable difficulties into implementing PHBS. The consideration can be given by their teacher, caretaker, parents, or trained social health workers in the hope of special needs children become independent into implementing the PHBS. The aim of this program is implementating social service program and helping special needs children to become independent in implementating PHBS and health protocol. The program was held by lecturing, demonstration, simulation, and supervised practice. This program hopefully will prevent either health issues or prevent the spreading of infective disease, specifically in this pandemic situation.
