Abstract

Oil palm OG hybrid genetic potentials are high in term of vegetative and generative traits, such as: low height increment, compact palm, high free fatty acid and high carotenes. However, OG hybrid reproduction is hindered due to its low germination rate. So, alternative method for germination is needed, which is in vitro embryo zygotic culture. This study objective was to reveal the best medium composition for OG hybrid embryo culture by modificated several oil palm basic medium from oil palm tissue culture protocol and to understand the role of plant growth regulators, activated charcoal, and sucrose on in vitro growth and development of OG hybrid embryo. The design used was Completely Randomized Design (CRD) with 16 replication and 4 embryos for each replicates, while the treatments were: A1 = M129 (control); A2 = M129 + activated charcoal; A3 = M129+ activated charcoal + sucrose; A4 = ½ M129 + activated charcoal + sucrose; A5 = M129 + GA3 ; and A6 = M129 + GA3 + NAA. From the study we found that the best medium for OG hybrid open pollinated clone's embryo culture was M129 with GA3 and NAA (A6). This medium gave percentage of in vitro germination up to 14.06%, and later on the plantlets from this medium gave the best vegetative growth performance such as high number of leaves and primer roots.

Abstract

An oil palm variety with high vitamin E has an added value because of its benefit as pharmaceutical and nutraceutical source. The measurement of the vitamin E content in CPO from eight varieties of oil palm is an effort to obtain high vitamin E varieties with the optimum oil yield. The varieties used in this experiment were DxP PPKS 718, DxP PPKS 239, DxP PPKS 540, DxP Yangambi, DxP Lame, DxP Avros, DxP Simalungun, and DxP Langkat, which were planted in a demonstration block, located at Kebun Adolina PTPN IV. The result showed that the average of the vitamin E from 8 varieties ranged from 477.36 ppm up to 582.78 ppm. The DxP Yangambi has the potency to be improved as the candidate of DxP variety with high vitamin E added value due to its highest vitamin E content. On the other hand, the DxP PPKS 540 is appropriate as candidate of ortets for high vitamin E clones regarding the vitamin E content; which is the highest over the whole samples

Abstract

Contamination in the in vitro culture is a critical problem causing the failure of seed production. Contamination in the oil palm plantlet is detrimental, considering that oil palm propagation is difficult and takes a long time. This research aimed to study the effect of sterilization during acclimatization of the contaminated oil palm plantlets by fungi on viability and to determine the optimum viability achieved from the contaminated materials. The materials used were contaminated plantlets of oil palm with roots, four leaves, and a height of about 17 cm. The plantlets were removed from the tube and cleaned with running tap water, then were sterilized, with treatments P1: soaking in benomyl-mancozeb-sodium hypochlorite and mannitol and rinsing with aquadest, P2: soaking in benomyl-mancozeb, P3: soaking in mancozeb. Cleaning plantlets under running tap water was carried out as a control treatment. The results showed that at 10 weeks after acclimatization, the survival rate of plantlets in each treatment (P1, P2, and P3) was significantly higher than that of the control. Sterilization methods affect the time new leaves emerge, leaf condition after sterilization treatment, and shoot height. The lowest fungal contamination after treatments was found in P2, followed by P3. After 3 months, plantlet survival rate decreased, with the highest survival rate in treatment P3 (32.3%) followed by treatment P2 (22.5%). In conclusion, acclimatization of contaminated oil palm plantlets can be carried out using a suitable sterilization treatment. Sterilization affects the survival rate and growth of in vitro-contaminated oil palm plantlets during acclimatization.
