Abstract

Improvement of nutrient intake was needed to increase cattle productivity. Probiotics and organic Cr, as feed additives, might have beneficially improve ruminant microbial ecosystem, and optimize digestion process by rumen microbes. The objective of this study was to examine the effects of combination between probiotic or mixed probiotcs and organic Cr on rumen fermentation activity. An in vitro study was held based on randomized block design with 5 treatments and 3 replications. The treatments were (A) Control ration, (B) Control ration + probiotic TSD 10, (C) Control ration + probiotic TSD 10 + organic Cr, (D) Control ration + mixed probiotics, and (E) Control ration + mixed probiotics + organic Cr. Organic Cr concentration was supplemented 2 ppm, whereas probiotics supplement contained 109 cfu/g. Supplementation of probiotic and organic Cr increased dry matter and organic matter digestibility but decreased total VFA and NH3 concentration. Treatment E (combination between mixed probiotics and organic Cr) gave the highest dry matter (52.45 %) and organic matter (51.96 %) digestibilities. It also tended to increase NH3 and total VFA production. Supplementation of single or mixed probiotics showed no difference for dry matter and organic matter digestibilities, and proportion of individual VFA. Mixed probiotics gave higher VFA and NH3 concentration compared to single probiotic. Probiotics supplementation resulted in a tendency for higher acetate proportion, while organic Cr supplementation resulted in a tendency of higher propionate proportion. Abstract

ABSTRAKPemberian pelet formula pada landak Jawa (Hystrix javanica F. Cuvier, 1823) : pengaruhnyaterhadap konsumsi, konversi pakan, dan kecernaan pada kondisi pra budidaya. Penelitiantelah dilakukan untuk mengetahui pengaruh pemberian pelet formula pada landak Jawa terhadapkonsumsi, konversi pakan, dan kemampuan cerna. Penelitian berlangsung selama 40 hari (12hari preliminary dan 28 hari masa pengumpulan data) di Penangkaran Mamalia Kecil, BidangZoologi, Pusat Penelitian Biologi – LIPI, Cibinong. Selama penelitian, delapan ekor landakmasing-masing ditempatkan di dalam kandang metabolik berukuran 1,4 m x 1,2 m x 1,0 m (panjangx lebar x tinggi). Rancangan penelitian adalah rancangan acak lengkap terdiri dari 4 perlakuanransum dan dua ulangan yaitu ransum kontrol (T0), ransum diberi pelet formula 50 g/ekor/hari(T1), ransum diberi pelet formula 100 g/ekor/hari (T2), dan ransum diberi pelet formula 150 g/ekor/hari (T3). Hasil penelitian menunjukkan tingkat kesukaan landak Jawa terhadap jenispakan penelitian berturut-turut adalah bengkuang, ketimun, talas belitung, pelet formula,kangkung, jagung manis, daun kitengis, kelapa, ubi jalar, dan kunyit. Konsumsi nutrien, kecualilemak pada perlakuan pemberian pelet formula (T1, T2, T3), lebih tinggi dibanding perlakuankontrol (T0). Konversi pakan pada landak T0 lebih tinggi dibandingkan pada landak T1, T2,dan T3, masing-masing 12,45%; 8,15%; 6,98%;, dan 5,58%. Kecernaan bahan kering adalah94,44% (T0); 95,79% (T1), 95,08% (T2); dan 97,16% (T3), sedangkan kecernaan bahan organikadalah 94,55% (T0), 96,16% (T1); 95,64% (T2); dan 97,45% (T3). Total digestible nutrien(TDN) tinggi pada semua perlakuan, yaitu 90,40% (T0); 92,69 % (T1); 92,03% (T2); dan 94,25%(T3) yang menggambarkan landak mampu mencerna semua nutrien pakan dengan baik.Kata kunci: pelet formula, konsumsi, konversi pakan, kecernaan, Hystrix javanica

Abstract

A total of 137 strains of Actinomycetes were isolated from 14 soil samples collected in 3 part of Lombok Island, Indonesia  using  SDS-YE (SY) and Rehydration and Centrifugation  (RC)  isolation methods  and Humic Acic Vitamin (HV) agar as isolation media. All the isolates were identified by morphological characteristic and by analysis of 16S rRNA gene sequence. On the basis of their morphology and 16S rRNA gene sequence, 67% of isolates were belonged  to the Streptomyces Group and 33% of isolates were belonged to the Rare-Actinomycetes (Non-Streptomyces) Group. Nine families and 15 genera were found from the samples. It is indicated the richness of actinomycetes in these area. The genus  Streptomyces  is the most abundant in all soil samples, occupying 67% of all isolates by using SY method; while the genusActinoplanes  is mainly found by using RC as isolation method. Two strains belong to the genus Virgisporangium and Catenulispora that showed taxonomic interest need further study for describing as ne species.   Keywords: Actinomycetes, Streptomyces Group, Non-Streptomyces Group, Diversity

Abstract

A total of 268 strains of Actinomycetes were isolated from soil and leaf-litter samples collected in Jambi (Sumatera) and Cibinong (West Java) using three selective isolation methods. All the isolates were identified by morphological characteristic and by analysis of 16S rDNA sequence. On the basis of their morphology and 16S rDNA sequence, 164 isolates were belonged to the Streptomyces Group and 104 isolates were belonged to the Rare-Actinomycetes (Non-Streptomyces) Group. Furthermore, 40% of isolates in Streptomyces Group and 62% of isolates in Non-Streptomyces Group are supposed to be new taxa. It is indicated the richness of Actinomycetes in these two area. The genus Streptomyces is the most abundant in soil samples, occupying 60- 75% of all isolates; while the genus Actinoplanes is mainly found in leaf-litter samples (70%). The use of selective isolation media is important to elucidate the microbial diversity.Key words: Actinomycetes, microbial diversity

Abstract

Amino acid protection is one of the efforts to improve livestock feed quality. Lysine is one of the limiting amino acids, which is easily degraded in the rumen, and thus requires a protection. This study aimed to determine the effect of protected lysine on cattle's digestive system and ruminal ecology in vitro. This study used encapsulation using xanthan gum and tannin (2% w/v) to protect lysine. The treatments consisted of P0 = unprotected amino acids; P1 = amino acids + 2 g xanthan gum; P2 = amino acids + 2 g tannin; P3 = amino acids + 2 g xanthan gum + 2 g tannin. Parameters observed included nutrient content, chemical activity, degradability of organic matter (OM) and crude protein (CP), and rumen fermentability (pH, N-NH3, VFA, gas kinetics, and CH4). A combination of xanthan gum and tannin coatings (P3) resulted in a higher effect (p<0.05) on rumen fermentation compared to others. The Undegraded DM and CP values in the rumen were 48% and 32%, respectively, and in the pepsin-HCl were 15% and 89%. All treatments did not change the rumen fermentability. In conclusion, protected amino acids has a effect on the cattle digestion system without disturbing the rumen ecology. Combining xanthan gum and tannin coatings provides the best results than using a single type of coating. Key words: amino acids, encapsulation, rumen fermentation, tannin, xanthan gum

Abstract

The Borneo earless monitor lizard (Lanthanotus borneensis) is one of the endemic animals of Indonesia. In 2019, the IUCN listed L. borneensis as an ‘endangered’ species. The declining population of the L. borneensis is a major concern for conservation efforts. One of the efforts to conserve endangered wild animals is to know the gut microbiome profile and the relationship between the host and the bacterial community in the body, which is associated with an influence on health. This research aims to determine the gut microbiome profile of animals with metagenomic analysis using Next-Generation Sequencing (NGS). This research was carried out by providing three feed types: earthworms, shrimp, and fish. Sample feces was collected by cloacal swabs and the DNA was sequenced using the Illumina NGS technique in the V4 16S rRNA region to examine the gut microbiome. The results of the metagenomic analysis showed differences in bacterial abundance in each feed treatment. The L. borneensis treated with earthworms and fish were dominated at the phylum level by Proteobacteria and Bacteriodota, while in shrimp, the phyla were Proteobacteria and Firmicutes. Index Shanon, Simpson, and the rarefaction curve also showed the highest bacterial diversity found in the fish treatment. This study can be used as a reference in designing suitable feed formulations for effective captive breeding for the conservation of L. borneensis

Abstract

This study investigates in depth the strategies used by senior high school students in an English as a Foreign Language (EFL) context to reduce speaking anxiety. Speaking anxiety is a common problem faced by many students, and this study aims to identify the main causes of such anxiety as well as effective strategies applied by students in the classroom. Through semi-structured interviews with a number of students, this study uncovered several key themes, including fear of communicating, fear of negative evaluation from teachers and peers, and various strategies used to overcome anxiety, such as thorough preparation before speaking, relaxation techniques to calm down, and social support from classmates. The findings from this study are expected to provide practical recommendations for educators, so that they can create a more supportive and inclusive learning environment, which in turn can help reduce students' speaking anxiety and increase their confidence in communicating in English. Abstract

In Liquid Organic Fertilizer (LOF), the problem that is often found is that the nutrients contained do not reach quality standards. Azolla and beef bone meal are known to contain macronutrients needed by plants, in addition to maximizing the existing content, functional bacteria are also added. This study aims to analyze the effect of the addition of azolla and beef bone meal on macro nutrient content, C-organic, coliform, and viability. The research method uses a completely randomized design (CRD) with 6 treatments and 3 replicates, namely (P1) 25% Azolla + 15% beef bone meal, (P2) 25% Azolla + 20% beef bone meal, (P3) 25% Azolla + 25% beef bone meal, (P4) 25% azolla + 15% beef bone meal + 10% functional bacteria, (P5) 25% azolla + 20% beef bone meal + 10% functional bacteria, (P6) 25% azolla + 25% beef bone meal + 10% functional bacteria. The research data were analyzed statistically using variance analysis and Duncan's multiple range test with the IBM SPSS Statistics 25 program. The results showed that the addition of 25% azolla + 25% beef bone meal (P3) had a significant effect (P≤0.05) on increasing macro nutrient content and functional bacteria viabilty, C-organic, coliform.

Abstract

Rice bran is a by-product of the rice milling process and has been well used as livestock feed. Rice bran is often adulterated with rice husk. The objective of this study was to evaluate the in vitro ruminal fermentation characteristics of rice bran with various compositions of rice husk and assess the relationship between rice husk addition and rice bran quality. The experiment was arranged in a completely randomized design with rice husk addition as a factor and three replications. Data of proximate value, gas production, ruminal degradability, and volatile fatty acid production were analyzed by analysis of variance. Moreover, significant effects of each treatment in the in vitro fermentation were further analyzed by Duncan's multiple range test (P<0.05). It was shown that the addition of rice husk to rice bran could increase acetic acid level, but it reduced potential gas production, gas production rate, organic matter and dry matter digestibility, and propionic acid level. Interestingly, the linear regression of dry matter digestibility, organic matter digestibility, and potential gas production showed the high adjusted R2 values. Moreover, this study also revealed that 10% of rice husk substitution on rice bran could significantly reduce the dry matter digestibility.
