Abstract

The Indonesian region has quite high air humidity and is ideal for the growth of straw mushrooms. To obtain pure cultures of straw mushrooms, you can go through the pure culture or G0 culture stage, then from G0 sub-cultures are carried out into G1, G2, G3 and G4. This research aims to obtain the best pure culture media for the growth of G3 FP005 Faperta Unsika mycelia. The research was carried out at the Biotechnology and Plant Breeding Laboratory, Faculty of Agriculture, Singaperbangsa University, Karawang, from April to May 2023. The research method used was a single factor Completely Randomized Design (CRD) with 7 treatments and 5 replications: PDA 100% (A), Charcoal Husk 100 % (B), 100% Husk (C), 75% PDA + 25% Husk Charcoal (D), 80% PDA + 20% Husk Charcoal (E), 50% Husk Charcoal + 50% Husk Charcoal (F), and PDA 50% + Charcoal Husk 25% + Husk 25% (G). Treatments were analyzed using variance and F test at 5% level, to find out the best treatment followed by further DMRT (Duncan Multiple Range Test) at 5% level. The results of the research were that there was a real effect of using several pure cultures on the growth of G3 FP005 mycelia. PDA 75% + Charcoal Husk 25% media gave the highest diameter at 2 dai (2.24 cm), 3 dai (4.78) and 4 dai (7.19 cm), the highest growth rate was (2.52 cm), diameter regression with R2 0.898 then P value 0.052047, and growth rate regression with R2 0.68 then P value 0.410604.

Abstract

One way to increase the genetic diversity of Ciherang as the superior variety is to cross Ciherang with the donor B11143D line as a New Plant Type (NPT) rice. This study aimed to obtain Ciherang X B11143D lines with the best vegetative characteristics in the field. The research was conducted in rice fields in Talagasari Village, Telagasari District, Karawang Regency, from May to September 2023. This experiment used a single-factor Randomized Block Design (RBD) with three replications of 22 treatments, consisting of 19 Ciherang X B11143D lines and three comparison varieties. The effect of treatment was studied using analysis of variance. The results showed that the rice lines significantly influenced the vegetative characters of Ciherang X B11143D lines in Telagasari, Karawang Regency. Based on the observed characters, several lines were selected i.e. 124.2.3, 94.3.3, and 20.4.4 lines. Those three lines chosen as backcrossing lines performed similarly compared to Ciherang as recurrent parent and inherited several important traits for rising productivity from the B11143D line as donor parent, namely the length and area of flag leaf, the total number of tillers, and the number of productive tillers, which were significantly higher than Ciherang.

Abstract

Kadmium (Cd) merupakan salah satu logam berat yang berada di lingkungan serta memiliki efek toksik yang tinggi dalam konsentrasi yang rendah. Lingkungan yang terkontaminasi Cd dapat diperbaiki kembali dengan sistem pengolahan limbah logam berat dengan pendekatan bioremediasi. Bakteri penghasil eksopolisakarida (EPS) yang berperan dalam mengkelat logam dapat digunakan sebagai agen bioremediasi seperti Azotobacter dan Pseudomonas. Penelitian bertujuan untuk menunjukkan efektivitas pupuk hayati terhadap populasi Azotobacter dan Pseudomonas pada media yang terkontaminasi logam berat Cd. Metode yang digunakan adalah metode eksperimental dengan Rancangan Acak Kelompok (RAK) faktor tunggal yang terdiri atas 9 perlakuan dengan 3 kali ulangan. Perhitungan data dianalisis menggunakan analisis uji F pada taraf 5%. Apabila hasil uji F perlakuan menunjukkan hasil perbedaan yang berbeda nyata, maka dilakukan analisis uji lanjut Duncan Multiple Range Test (DMRT) pada taraf 5%. Hasil penelitian menunjukkan bahwa perlakuan berpengaruh nyata terhadap populasi Azotobacter dan Pseudomonas, serta akumulasi Cd pada akar. Perlakuan D (8 ppm Cd (25 ml Azotobacter)) memberikan hasil tertinggi terhadap populasi total Azotobacter (407,547 CFU/ml) dan akumulasi logam Cd pada akar paling rendah (0,649 mg/kg). Sedangkan perlakuan B (8 ppm Cd (50 ml/l Pseudomonas + 25 ml/l Azotobacter) memberikan hasil tertinggi terhadap populasi total Pseudomonas (353,703 CFU/ml). Perlakuan Inokulasi pupuk hayati Azotobacter dan Pseudomonas mampu resisten terhadap media yang terkontaninasi logam Cd hingga 16 ppm, mampu menurunkan akumulasi logam Cd pada akar tanaman, dan dapat digunakan dalam bioremediasi logam Cd pada media yang terkontaminasi Cd.

Abstract

Pencemaran air oleh logam berat menyebabkan kualitas air menurun dan mengancam kelangsungan hidup organisme karena dapat menimbulkan efek toksik. Memanfaatkan agen hayati seperti Azotobacter sp. dan Pseudomonas sp. dapat digunakan sebagai cara alternatif untuk menanggulangi pencemaran air dan mendukung pertumbuhan tanaman. Penelitian ini bertujuan untuk mendapatkan kombinasi pupuk hayati yang tepat terhadap pertumbuhan tanaman apu-apu (Pistia stratiotes L.) pada media tanam yang terkontaminasi logam berat. Penelitian dilaksanakan di rumah kaca Universitas Singaperbangsa Karawang, Jalan HS. Ronggo Waluyo, Telukjambe Timur, Karawang. Metode yang digunakan adalah Rancangan Acak Kelompok (RAK) faktor tunggal dengan 9 perlakuan dan 3 ulangan sehingga terdapat 27 unit percobaan terdiri dari: A (kontrol tanpa Pb dan tanpa pupuk hayati), B (10 ppm Pb (50 ml Pseudomonas sp. + 25 ml Azotobacter sp.)), C (10 ppm Pb (50 ml Pseudomonas sp.)), D (10 ppm Pb (25 ml Azotobacter sp.)), E (10 ppm Pb tanpa pupuk hayati), F (20 ppm Pb (75 ml Pseudomonas sp. + 50 ml Azotobacter sp.)), G (20 ppm Pb (75 ml Pseudomonas sp. + 50 ml Azotobacter sp.)), H (20 ppm Pb (50 ml Azotobacter sp.)), I (20 ppm Pb tanpa pupuk hayati). Hasil percobaan dianalisis dengan uji F, apabila berpengaruh nyata maka diuji lanjut Duncan Multiple Range Test (DMRT) taraf 5%. Hasil penelitian menunjukkan bahwa perlakuan B (10 ppm Pb (50 ml Pseudomonas sp. + 25 ml Azotobacter sp.)) berpengaruh nyata terhadap semua parameter pengamatan. Perlakuan ini memberikan hasil terbaik pada tinggi tanaman (17,523 cm), jumlah daun (38,443 helai), panjang akar (32,256 cm).
