Abstract

Kemasan biokomposit adalah kemasan yang dibuat dari campuran dari beberapa bahan alam. Dalam makalah ini dipaparkan hasil penelitian kemasan biokomposit yang menggunakan campuran kulit luar biji kopi dengan polimer alam polivinil alkohol (PVA) yang berasal dari sintesis bakteri. Kulit luar kopi merupakan limbah dari industri pengolahan kopi yang terdiri dari campuran kulit luar dan kulit tipis (silver skin) yang terlepas pada saat proses pembersihan. Kulit luar kopi mengandung kandungan serat selulosa yang dapat menjadi bahan pengisi dalam pembuatan kemasan biokomposit berbasis PVA. Polivinil alkohol (PVA) memiliki kekuatan tarik yang baik sehingga sering dimanfaatkan dalam pembuatan kemasan, meskipun demikian PVA memiliki kelemahan yaitu mudah menyerap air sehingga mudah terurai ketika kontak dengan air. Dalam penelitian ini, kulit luar kopi ditambahkan sebagai material pengisi untuk meningkatkan sifat fisik dan mekanik komposit berbasis PVA komersial supaya lebih tahan air, tidak mudah terurai, dan aman bagi lingkungan. Yang menjadi faktor dalam penelitian ini adalah rasio perlakuan PVA dan limbah kopi (100:0, 90:10, 80:20, 70:30, 60:40, 50:50) serta penambahan plasticizer gliserol dengan konsentrasi yang berbeda (2% dan 4%) campuran kemudian dicampur dan dipanaskan sebelum dicetak dengan menggunakan aplikator. Pada tahap berikutnya dilakukan penambahan asam sitrat untuk meningkatkan ketahanan air dari kemasan biokomposit. Penambahan kulit luar kopi dapat meningkatkan karakteristik mekanik kemasan biokomposit berbasis PVA (rasio 90:10) dengan penambahan gliserol dan asam sitrat sebanyak 2%. Biokomposit yang dihasilkan memiliki penurunan daya serap air sebanyak sebanyak 70% dan waktu urai 2 kali lebih panjang dibanding kontrol kemasan dengan menggunakan 100% PVA. Selain itu juga memiliki tekstur yang baik, dengan nilai kuat tarik sebesar 12,48±1,60 MPa, elongasi sebesar 301,99±32,18%, dan modulus Young sebesar 4,16±0,08 MPa.

Abstract

Tomato fruit (Lycopersicum esculentum Mill.) is a food material with a high water content so it is easy to decompose. This shows that efforts to prevent reduction in food quality can be developed in the form of edible coating from aloe vera gel with concentration of 0%, 30%, 40%, and 50%, and carrageenan with concentration of 1%, 2%, and 3%. The aims of this research were to determine the concentration of aloe vera gel and carrageenan based on the characteristics of edible film and to determine the effect of edible coating and storage time on the quality of tomato. The first stage of this research is making edible film based on analysis parameters such as water vapor transmission rate (WVTR), tensile strength, elongation, and thickness. The selected concentration used in the second stage of this research for making edible coating was 40% of aloe vera gel and 3% of carrageenan. The selected concentration has a WVTR value of 1.758±0.20 g.mm/m2.hour, tensile strength value of 0.248±0.00 N/cm2, elongation value of 25.164±0.10%, and thickness value of 0.074±0.00 mm. Tomato samples coated with edible coating and control tomatoes is stored for 9 days at room temperature (24-25oC and 65–70% relative humidity aproximately) and samples are analyzed every 3 days based on analysis parameters such as vitamin C content, weight loss, hardness level, pH, ohue, lycopene content, and total dissolved solids value. The application of edible coating and storage time can inhibit the decrease of vitamin C levels, lycopene levels, weight loss, hardness, also can inhibit the increase in pH value and total dissolved solids value. Abstract

Indigo Carmine (IC) is a synthetic dye that might promote irritations, vomiting, and diarrhea when orally consumed in high concentrations. Copper is a dangerous heavy metal that tends to accumulate in organisms' cells as a pollutant. Hyperproduction of IC and accumulation of copper in water could cause an environmental problem that must be adequately addressed. Kaolin is found abundant on Bangka Island. Based on XRD analysis, Kaolin Island from Bangka Island consists of two important minerals, Kaolinite and Rectorite. Kaolin has chemical and physical properties, which make it useful in several applications, including as an adsorbent. This study aims to evaluate how Kaolin can adsorb copper ion from acid wastewater (pH < 0.5) and determine natural Kaolin's ability to adsorb IC dye from an aqueous solution. Many factors, including mixing time and number of doses of Kaolin, influence Kaolin's ability to absorb IC. The longer the stirring time, the more IC is absorbed. The optimal temperature for IC adsorption is 25˚C. The optimal kaolin dose for IC adsorption is 20% (m/v). Copper concentration after application of Kaolin tends to decrease, with optimal at 25.09% (m/v) dosage for 19.23% copper removal.

Abstract

Kecombrang (Etlingera elatior) is a plant that contains bioactive compounds such as alkaloids, phenolics, flavonoids, triterpenoids, saponins, and glycosides which are beneficial for body health. This research aimed to determine the antioxidant and antibacterial activity of extracts of kecombrang flowers, stems, and tubers. Powder from each part of the kecombrang plant (flowers, tubers, and stems) was extracted with 80% ethanol solvent using the maceration method for 24 hours at room temperature. The chemical compound composition of the selected extracts was determined using the GC-MS method. Kecombrang flower extract has antioxidant activity (IC50) of 34.13 ± 0.86 ppm, total phenolics of 28.38 ± 1.12 mg GAE/g, and total flavonoids of 2.15 ± 0.03 mg QE/g. Tuber extracts had antioxidant activity (IC50) of 35.09 ± 1.1 ppm, total phenolics of 18.79 ± 0.91 mg GAE/g, and total flavonoids of 0.96 ± 0.05 mg QE/g. Meanwhile, stem extract has antioxidant activity (IC50) of 38.35 ± 1.32 ppm, total phenolics of 14.49 ± 0.56 mg GAE/g, and total flavonoids of 0.77 ± 0.03 mg QE/g.  The antibacterial activity assay against E.coli, kecombrang flower extract showed an inhibition zone of 23.08 ± 0.49 mm with a MIC value of 0.53% and MBC 2.14%, tuber extract showed an inhibition zone of 21.87 ± 0.29 mm with a MIC value of 0.51% and an MBC of 2.04%, the stem extract showed an inhibition zone of 9.68 ± 0.35 mm with a MIC value of 0.41% and a MBC of 1.66%. While  the antibacterial activity assay against S.aureus, kecombrang flower extract showed an inhibition zone of 23.97 ± 0.50 mm with a MIC value of 0.63% and MBC 2.50%, tuber extract showed an inhibition zone of 22.86 ± 0.51 mm with a MIC value of 0.40% and MBC 1.58% and the stem extract showed an inhibition zone of 21.65 ± 0.42 mm with a MIC value of 0.54% and MBC 2.17%. The antibacterial activity of the three types of kecombrang extract is classified as strong. The results of GC-MS analysis of kecombrang flower extract showed the presence of the compounds 3-ethyl-2-cyclohexen-1-ol, 1,2-Benzenediol, and 1,4-Benzenediol which are antibacterial, and 2-methoxy-4-vinylphenol which is an antioxidant.

Abstract

This study aims to evaluate the antioxidant activity and stability of the extract of pigmented rice. Pigmented rice powder from 6 different varieties including Subang, Jowo Melik, Andel Abang, Bukittinggi, Cempo Merah, and Cempo Ireng) was macerated using ethanol with concentrations of 96% and 70%. Next, the rice extract was tested for stability to pH (1, 3, 5, 7, 9) and temperature (20°, 40°, 60°, 80°C). Results revealed that extract of Subang black glutinous rice using 96% ethanol resulted the highest antioxidant activity of 11.11±2.04 mg FeSO4/g, with an overall anthocyanin, flavonoids, phenolic contents of respectively 28.22±1.14 mg CE/g, 2.91±1.13 mg QE/g, and 3.46±1.59 mg GAE/g. The physiochemical properties of the rice extract were affected by the difference of pH. The increase in pH resulted in a decrease in antioxidant activity, total anthocyanin, and the values of color attributes (L*, a*, b*, C*, and ºHue) of the rice extracts. Temperature did not affect the physiochemical properties of the extract. The antioxidant activity, total anthocyanin, and color attributes of the rice extract were typically stable when the temperature rose from 20°C to 80°C. The extract at pH 1 and 80ºC treatment exhibited slightly higher yield based on antioxidant activity, total anthocyanin content, a* value, and ºHue with the values of 16.92±1.73 mg FeSO4/g, 33.90±2.33 mg CE/g, 21.55±0.54, and 30.04±0.57, respectively. Keywords: Anthocyanin, antioxidant activity, pH, pigmented rice, temperature.

Abstract

Red melinjo (Gnetum gnemon L.) peels have high carotenoid content and have the potential to be a natural food colorant. But it is difficult to dissolve in water, easily degraded by heat, and light. Microencapsulation is the process of coating core material which is easily damaged by using coating material. The purpose of this research is to protect, maintain, and improve the stability and solubility of carotenoid from red melinjo skin extracts. This research consist of two stages, preliminary and main research. In the first stage preliminary research, red melinjo peels were extracted using ethyl acetate and its characteristics were analyzed. In the second stage preliminary research, microencapsulation of extract using spray drying method, then its characteristics were analyzed. In the main research, the determination of the number of microcapsules and the ratio of citric acid and tartaric acid based on the characteristics of the best effervescent powder drink. Characteristics of red melinjo peels extract, water content 18.47%, yield 1.37%, total carotenoid 19.41 mg/g sample, total phenolic 27.86 mg GAE/g sample, and total flavonoid 30.60 mg QE/g sample. Characteristics of microcapsules, encapsulation efficiency 62.10%, solubility of microcapsule 93.44%, total carotenoid 0.29 mg/g sample, total phenolic 1.2 mg GAE/g sample, and total flavonoid 2.23 mg QE/g sample, moisture content 6.18%, yield 65.72%, and vitamin C content 404.37 mg/100g. Selected effervescent powder drink with the treatment of 0.5 gram microcapsules and 1: 2 acid ratio has the highest total carotenoid content, which is 0.05 mg/g sample, pH 6.03, foam volume 129.17 ml, soluble time 156.17 second, water content 8.99%, and the color is red based on °Hue 29.08. The chemical composition are water content 8.99%, ash content 21%, protein content 0.06%, carbohydrate content 69.79%, fat content 0.16%, and vitamin C content of 2.35 mg/100ml.

Abstract

Melinjo peel has a high content of polyphenol compounds, is expected to inhibit the activity of the enzyme α-glucosidase. The activity of α-glucosidase is related to the absorption of glucose and blood sugar. The content of polyphenols in the melinjo peel can be utilized more optimally by making the juice of melinjo fruit peel and processed as a jelly drink. Making jelly drinks requires hydrocolloids as a gelling material. The objective of the research are to determine the type of melinjo peel juice that has the best α-glucosidase inhibition activity and determine the ratio of melinjo peel juice  and the best percentage of hydrocoloid in the making of jelly drinks. The first stage of research is to determine the type of melinjo peel  red, yellow, and green that will be selected into the best melinjo skin juice that has inhibitory activity αglucosidase and also has the best antioxidant activity. The second stage of research is making the best jelly drinks with treatment ratio between water and melinjo peel juice and percentage of the amount of hydrocoloid used. IC 50 of α-glucosidase inhibition activity yellow melinjo peels juice is 22,393 ppm as the highest activity. Based on organoleptic, color, texture, pH, total soluble solid, and syneresis tests that were done to all samples of jelly drink, the selected jelly drink is the one with 50:50 ratio of yellow melinjo peels juice to water and concentration of hydrocoloid of 0.20%. The jelly drink has IC 50  Î±-glucosidase inhibition of 30,974 ppm, antioxidant activity of 12.4054 mg VCE/100mL, phenolic total of 0.4037 mg GAE/g, flavonoid total of 0.0173 mg QE/g. ABSTRAKKulit melinjo memiliki kandungan senyawa polifenol yang tinggi, diharapkan dapat menghambat aktivitas enzim α-glukosidase. Aktivitas α-glukosidase berhubungan dengan penyerapan glukosa dan kadar gula di dalam darah. Kandungan polifenol pada kulit melinjo dapat dimanfaatkan secara lebih optimal dengan cara mengambil sari kulit buah melinjo dan diolah sebagai minuman jeli. Pembuatan minuman jeli memerlukan hidrokoloid sebagai bahan pembentuk jel. Penelitian ini bertujuan untuk menentukan jenis sari kulit melinjo yang memiliki aktivitas penghambatan α-glukosidase terbaik serta menentukan rasio sari kulit melinjo dan persentase hidrokoloid terbaik dalam pembuatan minuman jeli. Tahap pertama penelitian adalah menentukan jenis kulit melinjo merah, kuning, dan hijau yang akan dipilih menjadi sari kulit melinjo yang memiliki aktivitas penghambatan α-glukosidase dan juga memiliki aktivitas antioksidan terbaik. Tahap kedua penelitian adalah pembuatan minuman jeli dengan perlakuan rasio antara sari kulit melinjo dan air serta konsentrasi  hirokoloid  yang dipakai. Aktivitas penghambatan α-glukosidase tertinggi diperoleh dari sari kulit melinjo kuning d engan nilai IC50 sebesar 22.393 ppm. Berdasarkan uji organoleptik, warna, tekstur, pH, total padatan terlarut, dan sineresis, minuman jeli terpilih dengan rasio sari kulit melinjo kuning:air 50:50 dan konsentrasi hidrokoloid  0,20%. Minuman jeli sari kulit melinjo kuning memiliki nilai IC50 penghambatan  α-glukosidase 30974 ppm, aktivitas antioksidan 12,4054 mg VCE/100mL, total fenolik  0,4037 mg GAE/g, total flavonoid 0,0173 mg QE/g.Kata kunci : aktivitas antioksidan, α-glukosidase, hidrokoloid, kulit melinjo, minuman jeli

Abstract

Yellow melinjo (Gnetum gnemon Linn.) peel extract contains active components such as resveratrol, tannin, steroid, flavonoid, and saponin. Ginger (Zingiber officinale Rosc.) also contains active components such as gingerol, paradol, and shogaol. These components in ginger and yellow melinjo peels are reported to have the activity to inhibit α-glucosidase. This research aim was to utilize yellow melinjo peel extract and ginger steep as functional drink that might have the activity to inhibit α-glucosidase. This research was divided into two stages. The preliminary stage was done to characterize yellow melinjo peel extract and ginger steep through activity inhibition of α-glucosidase, total phenolic, total flavonoid, and antioxidant activity. Main research was done to apply yellow melinjo peel extract and ginger steep into functional drink that use different concentration of yellow melinjo peel extract (0,12%, 0,16%, and 0,20%) and also using different concentration of stevia sweetener (0,3%; 0,4%; and 0,5%). All functional drinks’ formula was analyzed in organoleptic (scoring and hedonic), color, pH, and total soluble solids tests. The selected functional drink was analyzed for its activity of α-glucosidase inhibition, α-glucosidase inhibition kinetics, total phenolic, total flavonoid, and antioxidant activity. Yellow melinjo peel extract has activity inhibition of α-glucosidase (IC50)99.87 ppm, total phenolic 21.36 mg GAE/g, total flavonoid 11.97 mg QE/g, and antioxidant activity (IC50)1386.80 ppm. The ginger steep has activity inhibition of α-glucosidase (IC50)247811.5 ppm, total phenolic 0.29 mg GAE/g, total flavonoid 0.08 mg QE/g, and antioxidant activity IC50 60227.61 ppm. The selected functional drink from the hedonic test is the one with 0,20% extract and 0,5% stevia sweetener. The selected functional drink has inhibition of α-glucosidase IC50 194125 ppm, antioxidant activity (IC50)55497.12 ppm, total phenolic 0.64 mg GAE/mL, and total flavonoid 0.21 mg QE/mL.

Abstract

Functional drink from guava (Psidium guajava Linn.) leaves are beneficial for human health, as a medicine for diarrhea, cough, antidiabetic, and antioxidants. Guava leaf in Indonesia has many varieties. This research uses guava leaves ”˜Kristal’, ”˜Sukun Merah’, and ”˜Australia’ varieties. The purpose of this study was the utilization of guava leaves extract in functional drinks with the addition of stevia leaves as a sweetener. This research was divided into 2 stages. Preliminary stage was carried out to determine guava leaves extract that will be used in functional beverages based on α-glucosidase inhibition activity and antioxidant activity. The main research was carried out by making functional drinks using different concentration of guava leaves extract (0.15; 0.175; and 0.2%) and different concentration of stevia leaves powder (0.025; 0.05; and 0.075%). All functional drink formulations were tested for sensory test (scoring and hedonic), pH, total soluble solids, and color tests. The functional drink was selected based on the results of the hedonic tests and analyzed for total phenolic, total flavonoid, antioxidant activity, and α-glucosidase inhibition activity. Selected guava leaves extract was ”˜Sukun Merah’ with α-glucosidase inhibition activity (IC50) of 38.18 ppm and antioxidant activity (IC50) of 21.69 ppm. The concentration of guava leaves extract and stevia leaves powder significantly affected the pH and °Hue of functional drinks. The chosen functional drink formulation is the one with 0.175% guava leaves extract and 0.075% stevia leaves powder. Selected functional drink have a total phenolic of 5.42 mg GAE/mL, total flavonoid of 0.07 mg QE/mL, antioxidant activity (IC50) of 3041.04 ppm, and α-glucosidase inhibition activity of 17066.65 ppm.Abstrak Daun jambu biji (Psidium guajava Linn.) telah dimanfaatkan bagi kesehatan tubuh manusia, seperti sebagai obat diare, batuk, antidiabetes, dan antioksidan. Di Indonesia tanaman jambu biji  memiliki berbagai varietas. Penelitian ini menggunakan daun jambu biji varietas Kristal, Sukun Merah, dan Australia. Tujuan penelitian ini adalah pemanfaatan ekstrak daun jambu biji dalam minuman fungsional dengan penambahan daun stevia sebagai pemanis. Penelitian ini dibagi menjadi 2 tahap. Penelitian pendahuluan dilakukan untuk menentukan varietas daun jambu biji yang akan digunakan pada pembuatan minuman fungsional berdasarkan aktivitas inhibisi α-glukosidase dan aktivitas antioksidan. Pada Penelitian utama dilakukan pembuatan minuman fungsional dengan perlakuan konsentrasi ekstrak daun jambu biji (0,15; 0,175; dan 0,2%) serta konsentrasi serbuk daun stevia (0,025; 0,05; 0,075%). Seluruh formulasi minuman fungsional dianalisis meliputi uji sensori (scoring dan hedonik), uji pH, uji total padatan terlarut, dan uji warna. Minuman fungsional terpilih ditentukan berdasarkan  parameter hedonik, total fenolik, total flavonoid, aktivitas antioksidan, dan aktivitas inhibisi α-glukosidase. Ekstrak daun jambu biji terpilih yaitu varietas Sukun Merah dengan aktivitas inhibisi α-glukosidase (IC50) sebesar 38,18 ppm dan aktivitas antioksidan (IC50) sebesar 21,69 ppm. Konsentrasi ekstrak daun jambu biji dan serbuk daun stevia berpengaruh signifikan terhadap pH dan °Hue minuman fungsional. Formulasi minuman fungsional terpilih yaitu konsentrasi ekstrak daun jambu biji sebesar 0,175% dan konsentrasi serbuk daun stevia sebesar 0,075%. Minuman fungsional terpilih memiliki total fenolik sebesar 5,42 mg GAE/mL, total flavonoid sebesar 0,07 mg QE/mL, aktivitas antioksidan (IC50) sebesar 3.041,04 ppm, dan aktivitas inhibisi α-glukosidase (IC50) sebesar 17.066,65 ppm. Minuman terpilih berpotensi menjadi minuman fungsional dengan manfaat sebagai antioksidan dan antidiabetes.
