Abstract

Stem cells are cells that became the beginning of the growth  to others cell that constract the whole body of organisms, including human. Stem cells are from embryonic, fetal, or adult cells, which are capable to proliferate to cells in a long time, which this cell doesn’t have specific function, stem cells are able to differentiate  into specific cell that build tissues and organ systems. Stem cells have charateristic including :  undifferentiated cells, this cell is self-renewal, cells can differentiate into more than one type of cells (multipotent/pluripotent). Types of stem cells include embryonic stem cells derived from embryo, or inner cell mass (ICM) and adult stem cells found in bone marrow, cord blood, umbilical cord, peripheral blood, liver, skin, brain, dental pulp, fatty tissue. The uniqueness of stem cells give new hope as an alternative therapy or cells therapy for various types especially degenerative diseases. Keywords: stem cell, cell therapy

Abstract

Wallet bird nest is a nest that can be consumed by humans, which is made from male swallow's saliva. Male swallow bird makes a nest with its saliva for 35 days. The purpose of this study was to determine the number of TPC (Total Plate Count), nitrite and L * a * b. The material of this study is wallet bird's nest. The method used is Completely Randomized Design (CRD) with 3 treatments and 4 replications P0 = Without treatment, P1 = room temperature, P2 = Freezer no vacuum, P3 = Vacuum freezer. The results showed that the best TPC test results obtained by room temperature treatment were 461.25 ? 39.167 x 104, and the best nitrite test was obtained in vacuum freezer treatment ie 1.42 ? 0.059, and the best L * a * b color test on room temperature treatment 1.50 ? 0.520. Conclusion of the results of this study is the best treatment in this study is the storage of swallow's nests at freezer temperature using vacum packaging seen from the TPC parameters, nitrite and redness level using the L * a * b * color meter instrument.

Abstract

Background: Breast cancer is the most common type of cancer and the second leading cause of death in the world. One of the supporting therapeutic efforts to overcome cancer is through food, mainly fruits. Ananas comosus has been investigated for its potential as an anti-cancer. It is a source of antioxidants from the content of Vitamin C and flavonoids that work by capturing free radicals, resulting in inhibition of cancer cell proliferation.Objective :This study was conducted to determine the antioxidant activity and cytotoxicity of water extract of Ananas comosus on T-47D breast cancer cell lines.Method: The process of extracting pineapple flesh is done using water as a solvent, then tested its antioxidant activity using DPPH and cytotoxicity using MTT assay on T-47D.Result: IC50 value for antioxidant activity was 463.369 μg / mL and the IC50 for the cytotoxicity assay was 488.003 μg / mL.Conclusion : The water extract of Ananas comosus have very weak antioxidant activity and moderate cytotoxicity properties that have potential as chemo preventive agents in T-47D breast cancer cell lines.

Abstract

Dengue is an infectious disease caused by dengue virus. Dengue endemic region includes America, Western Pacific, Africa, East Mediterranian, and South East Asia including Indonesia. An early diagnostic system specific for Indonesia is needed to control dengue in Indonesia. In this research, cloning of Non Structural 1 (NS1) gene from dengue virus type 3 (Indonesian strain D3-1703) into pYES2/CT vector was performed. In the long run, NS1 recombinant protein will be expressed in Saccharomyces cerevisiae for diagnostic materials. Polymerase Chain Reaction (PCR) amplification of NS1 gene fragments were done with optimal annealing temperature at 55 ºC. NS1 gene fragment and pYES2/CT were cut by BamH I and Not I enzymes. The digested pYES2/CT was dephosphosrylated using Calf Intestine Alkaline Phospatase enzyme. Ligation with the vector:insert ratio of 1:12 and 1:20 resulted in 6 and 5 recombinant colony candidates respectively. Restriction enzyme and PCR verifications showed that 5 recombinant plasmids contained NS1 gene. Sequencing of the first 600 bp of one recombinant plasmid was performed. The blastn analysis showed that it had a 99% identity with dengue virus type 3 strain FW06. Finally, it was shown that NS1 clone within pYES2/CT was in the correct Open Reading Frame and ready to be expressed in S. cerevisiae.

Abstract

Obesity is one of the risks of Diabetes Mellitus (DM) that requires health screening measures. Different characteristics of rural and urban residents in terms of lifestyle behaviour have an impact on the risk of DM. One of the risk indicators can be seen from the aspects of body mass index (BMI), body fat, and blood glucose levels. The purpose of this paper is to determine the profile of body mass index, body fat, and blood glucose levels between urban and rural residents. Additionally, education and consultation will be provided. Activities were carried out in the form of health screening in Malang City and Madiun Regency for 1 month with a cross sectional design and total sampling according to the presence of participants. Additionally, education and consultation were conducted. The instruments used include a blood glucose meter, HBF-375 body composition meter, and educational videos. Based on the results of measurements in 20 rural people obtained a mean BMI of 23.11kg/m2 (14-61 kg/m2), mean body fat 27.72% (8-41%) and mean blood glucose levels 89.5mg (75-158mg), while measurements in 15 urban people obtained a mean BMI of 27.59 kg/m2 (16-38 kg/m2), mean body fat 32.85% (17-44%) and mean blood glucose levels 123mg (99-216mg). It was concluded that the mean body mass index, body fat percentage, and instantaneous blood glucose levels were higher in urban than rural communities. This can be used as a basis for educational management to prevent the risk of metabolic syndrome. It is necessary to develop intervention management in managing the risk of metabolic syndrome in further community service activities by taking into account regional characteristics.

Abstract

Abstrak Anak usia sekolah merupakan kelompok yang rentan akan kekurangan zat gizi terutama zat energi. Hal ini terjadi karena anak usia sekolah mengalami puncak pertumbuhan terutama pada penambahan tinggi dan berat badan. Sehingga, kecukupan dan pemenuhan zat gizi menjadi hal yang penting dipenuhi pada masa tersebut. Sebanyak 10-20% kecukupan harian anak terpenuhi oleh jajanan anak di sekolah. Meski demikian, faktor nilai gizi dan higienitas jajanan perlu menjadi perhatian. Program edukasi ini bertujuan untuk memberikan edukasi bagi pihak terkait guna meningkatkan pengetahuan mengenai jajanan sehat dan higienis bagi anak sekolah dasar. Sebanyak 27 peserta mengikuti kegiatan yang terdiri dari guru sekolah dasar, wali murid, dan pengelola kantin di SDN Kiduldalem 2 Kota Malang. Materi yang diberikan pada program edukasi ini adalah terkait pola makan bergizi seimbang bagi usia anak sekolah dan jenis jajanan sehat di kantin sekolah. Hasil rerata nilai pre-test dari peserta adalah 6,70 dan post-test 9,00 (dari rentang 10). Terdapat peningkatan 2.30 poin setelah dilaksanakan program edukasi dan sebanyak 55,5% (15 peserta) mendapatkan nilai post-test 10 pada akhir kegiatan. Hasil uji statistik menunjukkan terdapat perbedaan signifikan antara nilai pre-post test yang menunjukkan bahwa kegiatan edukasi berlangsung efektif dan materi dapat dipahami oleh semua peserta.Kata kunci: Jajanan Sekolah, Cemaran Makanan, Higienitas, Edukasi Gizi.

Abstract

Sampah menjadi salah satu sumber permasalahan yang memiliki dampak yang signifikan pada lingkungan sehingga diperlukan upaya pengelolaan yang baik. Bank sampah Desa Nglambangan mampu meningkatkan kesejahteraan hidup masyarakat sejak didirikan. Kondisi pandemi covid-19 menyebabkan kegiatan bank sampah berhenti beroperasi dan hal ini berkelanjutan hingga saat ini pasca covid-19 karena kegiatan ini tidak dijadikan prioritas pemerintah desa. Desa Nglambangan merupakan salah satu desa yang berada di Kabupaten Madiun yang memiliki tingkat penderita penyakit kronik cukup tinggi dibandingkan dengan desa lain. Selama ini, penderita penyakit kronik seperti diabetes melitus menggunakan asuransi BPJS untuk mendapatkan layanan kesehatan yang dibutuhkan. Asuransi bank sampah merupakan salah satu pilihan yang dapat dikembangkan dalam upaya mendukung program penanganan penyakit kronik. Edukasi bank sampah meliputi manajemen keuangan bank sampah dan konsep asuransi bank sampah menjadi alternatif atas permasalahan tersebut. Tujuan kegiatan pengabdian masyarakat ini adalah untuk meningkatkan pemahaman dan pengetahuan masyarakat akan pentingnya manajemen dalam bank sampah dan mengenalkan konsep asuransi bank sampah. Hasil dari kegiatan pengabdian masyarakat ini adalah edukasi manajemen bank sampah sebagai sarana asuransi kesehatan di Desa Ngalambangan Kecamatan Wungu Kabupaten Madiun telah berhasil meningkatkan pemahaman dan pengetahuan masyarakat. Hasil pre test dan post test menunjukkan pengetahuan dan pemahaman masyarakat akan pentingnya pengelolaan bank sampah dan asuransi meningkat. Kegiatan edukasi mengenai bank sampah tersebut juga dapat dilaksanakan secara berkelanjutan dengan melibatkan pemangku kepentingan untuk menjamin keberlanjutan bank sampah di masa yang akan datang. Abstract

Tempe is one of the popular foods in Indonesia which comes from fermented soybeans with the help of Rhizopus oligosporus. The development of tempeh with non-soy ingredients is starting to be carried out frequently. Cowpea (Vigna unguiculata subsp. unguiculata) is one of the nuts that is often consumed by Indonesian people. Cowpeas can be an alternative substrate for tempeh fermentation, because of their nutritional content, including vitamins and minerals, which are quite good for the body. However, some of these minerals are still bound to the phytic acid compound so it needs to be explained by the fermentation process. This research aimed to determine the ash content, mineral content of iron, calcium, and total mold in cowpea tempeh at a fermentation time of 35, 45, and 54 hours using standard methods. The research design used was a Completely Randomized Design (CRD) with repetition 3 times for each treatment for fermentation duration P1 (35 hours), P2 (45 hours), and P3 (54 hours). The ash and calcium content of cowpea tempeh decreased with fermentation times of 45 and 54 hours compared to 35 hours. In terms of iron content, the amount was almost the same at all fermentation times. Meanwhile, the total number of molds increased with fermentation times of 45 and 54 hours. Based on the results of observations for further research, the fermentation time for making cowpea tempeh used in this optimization was determined to be 35 hours. A fermentation time of 35 hours produces good cowpea tempeh, high calcium content, and efficient time and energy.

Abstract

High disaster risk in Indonesia necessitates the development of emergency food products (EFP). One potential EFP is cookies made from foxtail millet and snakehead fish. This study aimed to examine the sensory, chemical, and physical properties of EFP cookies with different formulations. This research used a completely randomized design. Variations in the composition of wheat flour, foxtail millet flour, and snakehead fish flour were P0(100:0:0), P1(8:76:16), P2(8:66:26), and P3(8:56:36). All treatments were analyzed for organoleptic quality using hedonic tests and proximate analysis, water activity analysis using an aw meter, and physical quality analysis using tensile strength. The Zeleny method was applied to identify the most optimal treatment, revealing that P1 emerged as the best formulation. P1 has the color, aroma, and taste that the panelists like, while maintaining a neutral rating in texture. P1 contains 241.51 kcal/50 grams of energy, 10.97% protein, 41.16% fat, 47.86% carbohydrates, 1.93% ash content, 4.93% water content, 0.55 water activity, and 17.16 N breaking strength. These results are in accordance with EFP and SNI 2973:2011 standards. The recommended daily consumption is 18 pieces (each piece weighing 25 grams) if not accompanied by any other food, or only 4-5 pieces if consumed as a snack.
