Abstract

Aims of this research were to know genetic correlation of birth and weaning weight of indigenous sheep. Materials used were 240 birth and weaning weight recorded from 100 days of the indigenous lambs age that were born from 40 dams mated by 8 rams. Nested classification model was used to estimate to genetic correlation. Result of the research showed that the estimated value of genetic correlation from sire, dam, and sire-dam components were 0,32 ± 0,31; 0,52 ± 0,54, and 0,36 ± 0,52, respectively. It could be summarized that genetic correlation between birth and weaning weight of the sheep had a moderate positive. Birth weight characteristic could be used for criteria selection with non-direct selectionmethod for weaning weight.

Abstract

Abstract. The aim of the study was to know the genetic characteristic and polymorphysm of Indonesian local ducks including Magelang, Tegal, Mojosari, Bali and Alabio duck based on Single Nucleotide Polymorphism (SNP) analysis in D-loop region mtDNA. The long term aim was to set the spesific genetic marker based on SNP D-loop region mtDNA which could differentiate local ducks in Indonesia. In the future, it could be used as selection tool for local duck conservation, and refinement strategy as well as the improvement of genetic quality by utilizing the available native duck germplasm. There were 20 ducks for each duck population and were taken 3 ml of its blood as sample. DNA Isolation Kit high pure PCR template preparation (Geneaid) was uded for Genome DNA isolation.  Amplification with PCR technique used primer DL-AnasPF (L56) as forward and DL-AnasPR (H773) as reverse. Next, PCR product or amplicon were sequenced. Sequence result were analyzed with SNP technique and observed the similarity and difference of its nucleotide sequence between individual and population. The result of the study showed that genome DNA from local duck in Indonesia was successfully isolated. DNA fragment of 718 bp was amplified with primer pair of DL-AnasPF and DL-AnasPR. Nucleotide sequence was 469 nt and analyzed with SNP technique. It was compared with standard nucleotide sequence of Anas platyrhynchos (HM010684.1) in Gen Bank. The result of nucleotide sequence similarity percentage was 99.68±0.56%. Single Nucleotide Polymorphism D-loop region mtDNA Indonesian local duck was 0.32±0.56%.  Some SNP was found in Magelang duck C (Klawu blorok), F (Cemani black),  G (Gambiran), H (Jarakan kalung), I (Jowo plain) and K (Plain white) also Tegal duck 8, 1, 2, 5, 2, 8 and 2 SNP respectively. It could be concluded that polymorphic genetic characteristic similarity were existed in Indonesia local duck populations which was shown by its big standard deviation SNP in D-loop region mtDNA. Magelang duck with different feather color relatively more polymorphic to another local duck in Indonesia. Single Nucleotide Polymorphism which was achieved could be used as genetic marker that differentiate genetic characteristic of Indonesian local ducks.Key words:  genetic characteristic, local duck, Single Nucleotide Polymorphism (SNP), D-loop mtDNAAbstrak.  Penelitian ini bertujuan untuk mengetahui karakteristik genetik dan polimorfisme itik lokal Indonesia yaitu itik Magelang, Tegal, Mojosari, Bali dan Alabio berdasarkan analisis Single Nucleotide Polymorphism (SNP) daerah D-loop mtDNA. Tujuan jangka panjangnya adalah menetapkan marker atau penanda genetik berdasarkan SNP daerah D-loop mtDNA spesifik yang dapat membedakan itik-itik lokal yang ada di Indonesia. Selanjutnya digunakan sebagai  alat bantu seleksi untuk konservasi, pembibitan  dan pengembangbiakan itik lokal.  Populasi masing-masing jenis itik lokal yang digunakan sebanyak 20 ekor untuk diambil 3 ml sampel darahnya. Isolasi DNA genom menggunakan DNA Isolation Kithigh pure PCR template preparation (Geneaid). Amplifikasi dengan teknik PCR menggunakan pasangan primer DL-AnasPF (L56) sebagai forward dan DL-AnasPR (H773) sebagai reverse. Produk PCR atau amplikon yang diperoleh disekuensing. Hasil sekuensing dianalisis dengan teknik SNP dan diamati kesamaan dan perbedaan urutan nukleotida antar individu itik dan antar populasi.  Hasil penelitian menunjukkan bahwa DNA genom dari itik lokal di Indonesia berhasil diisolasi. Amplifikasi dengan teknik PCR berhasil memperoleh fragmen berukuran 718 bp. Urutan nukleotida hasil sekuensing sebesar 469 nt dianalisis dengan teknik SNP dan dibandingkan dengan urutan nukleotida standar dari itik Anas platyrhynchos (HM010684.1) yang ada di Gen Bank, diperoleh persentase kesamaan urutan nukleotid sebesar 99,68±0,56%. Single Nucleotide Polymorphism daerah D-loop mtDNA pada itik lokal di Indonesia sebesar 0,32±0,56%. Sejumlah SNP ditemukan pada itik Magelang C (Klawu blorok), F (Hitam cemani),  G (Gambiran), H (Jarakan kalung), I (Jowo polos) dan K (Putih polos) serta itik Tegal  masing-masing 8, 1, 2, 5, 2, 8 serta 2 SNP. Kesimpulan dari penelitian ini adalah terdapat karakteristik genetik yang polimorfik pada populasi itik lokal di Indonesia, ditunjukkan dengan adanya simpang baku SNP pada daerah D-loop mtDNA yang relatif besar. Itik Magelang dengan warna bulu yang berbeda relatif lebih polimorfik dibandingkan dengan itik lokal lainnya di Indonesia.  Single Nucleotide Polymorphism yang diperoleh dapat digunakan sebagai penanda genetik yang dapat membedakan karakteristik genetik yang dimiliki oleh itik lokal di Indonesia.Kata kunci:  karakteristik genetik, itik lokal, Single NucleotidePolymorphism (SNP),  D-loop mtDNA

Abstract

The aims of this research was to identify the qualitative and quantitative difference of the phenotypes of native mallard and Muscovy, and Muscovy the genetic diversity between Muscovy and mallard using RFLP method with growth hormone primer (GH) gene. The materials were 30-week old male and female ducks from 5 strains—Magelang, Mojosari and Tegal ducks, white-feathered, and black and white-feathered Muscovy. Muscovy Completely Randomized Design was used to analyze parameters 5 strains of ducks based on gender and with 5 replications. The parameters measured were local and Muscovy duck phenotypes and genetic diversity (number of alleles) based on GH gene, heterozygosity and genetic distance. Qualitative properties were analyzed descriptively and quantitative properties were analyzed using Anova followed by HSD in case of significant differences, whereas RFLP analysis was used to determine the allele frequencies, genotype frequencies, genetic diversity, and genetic distance of local ducks. Result showed phenotypic differences between Anas platyrhynchos and Cairina moschata. The dominant feather colorof Muscovy was black and white while mallard was brown. The bill color Muscovy Muscovy was pink with dark brown but black in mallard. The dominant shank’s color in both Muscovy and mallard was black. Body size of mallard was larger than that of Muscovy Muscovy, and Magelang ducks weigherd more than Tegal and Mojosari Duck. The PCR-RFLP results showed lower heterozygosity of mallard compared to Muscovy based on GH gene, and genetic distance of the Tegal duck was closer to Mojosari and Magelang ducks, whereas Magelang and Mojosari ducks had a considerable genetic distance based on the GH gene.

Abstract

The aim of present study was to investigate myostatin gene polymorphism and its association with weaning weight and body weight in Batur sheep, 30 heads of Batur all lambs were fed by concentrated feed. Body weight measured monthly after weaning to the six months of age. DNA Extraction used 200 ul of whole blood samples. To amplify exon 3 region of MSTN gene a specific primer designed using the Primer3 software. The 25 μl volume contained 25 ng of genomic DNA, 12.5 μl 2x Reaction mix of each primer. The cycling protocol was 5 minutes at 95°C as initial denaturation, 35 cycles of denaturing at 94°C for 45 seconds, annealing at 73.9 for 45 seconds, extending at 72°C for 40 s, with final extension at 72°C for 10 minutes. Eleven polymorphic sites were observed in the in 3rd exon region transversions at c.*121 G instead of A, del-T at c.*129, one individual at c.*139 and one individual at c.*158 positions however, one individual sequence disrupted reading frame in whole MSTN sequenced, also weaning weight and 6-month weight of Batur lambs were 21.13±5.70 and 31.64±7.06 to the homozygous whilst 19.99±4.33 and 30.50±5.44 for heterozygote respectively but there is no significe differences seen above all the studied parameters. Homozygous AA lambs had fairly low weaning weight and body weight compared to the heterozygous AB lambs but might have a higher carcass weight, further investigation is needed into the interaction of MSTN with other genes involved in muscle growth.

Abstract

This research was aimed to investigate heritability score (h2) of milk yield, kid birth weight, and milk quality that included density, fat content, protein, lactose, water and non-fat dry matter of Saanen goat according to the production record of first-lactating individual in the Centre of Pedigree Breeding and Cattle Forage Baturraden. The materials for this research were 180 Saanen does with a first-lactating record. The estimated genetic parameter was heritability score using a paternal half-sib correlation method. Result showed that the heritability of milk yield, kid birth weight, density, fat content, protein, lactose, water and NFDM was 0,32+0,23; 0,25+0,26; 0,15+0,17; 0,11+0,16; 0,12+0,16; 0,10+0,14; 0,10+0,16 and 0,11+0,16, respectively. A high h2 was obtained from heritability score of milk yield, and a moderate h2 was from kid birth weight and milk quality. In conclusion, the h2 score of milk yield of Saanen goats was relatively high, while the h2 score of kid birth weight and milk quality was relatively moderate.

Abstract

Pelaksanaan Kolaborasi Sosial Membangun Masyarakat (Kosabangsa) pada kegiatan ini bertujuan untuk meningkatkan dan menerapkan ilmu pengetahuan dan teknologi mengenai pengomposan limbah ternak kambing dan limbah baglog jamur tiram untuk meningkatkan produksi pangan sekaligus dapat meningkatkan ketahanan pangan masyarakat. Metode yang digunakan pada pelaksanaan kegiatan ini adalah metode Participatory Action Research (PAR) dengan sasaran kegiatan adalah gabungan kelompok tani (Gapoktan) Tani Makmur Desa Tlaga, Kecamatan Gumelar, Kabupaten Banyumas. Kegiatan berupa transfer teknologi melalui kegiatan penyuluhan, pelatihan, demplot dan pendampingan serta pemberian peralatan teknolgi dan inovasi kepada gapoktan Tani Makmur. Hasil kegiatan ini adalah meningkatnya pengetahuan, ketrampilan dan meningkatnya kesuburan tanah yang pada khirnya dapat meningkatkan produktivitas tanaman pangan di Desa Tlaga.

Abstract

This study aims to determine the increase in production capacity through genetic quality improvement using selection techniques with the production capability test method in Tegal ducks. The research was conducted using an experimental method, using a nested pattern experimental design, male as treatment, female as sub-treatment, offspring as replicates, and production characteristics as observations. The selection population of Tegal ducks consisted of 10 males, 50 females, and their offspring. Production characteristics recorded included egg weight, hatching weight, and egg production. Egg production measured was Hen Day Production (HDP) at the initial laying period for 90 days. Assessment of genetic quality using individual breeding values (BV). Each individual's breeding value is calculated, and then the estimated breeding value (EBV) results are arranged based on their rank. Selection of parent candidates is done by maintaining 25, 50, and 75% of the total population. Selection results are obtained by estimating the selection response using different selection intensities. The results showed that the average and standard deviation of the characteristics of egg weight, hatching weight, and percentage of egg production in Tegal ducks were 67.76 ± 4.57 g, 40.40 ± 2.16 g, and 63.33 ± 10.89 %, respectively. Heritability values (h2) and standard error of egg weight characteristics, hatching weight, and percentage of egg production in Tegal ducks were 0.47 ± 0.032, 0.39 ± 0.0589, and 0.512 ± 0.071, respectively. The assessment for selection response was conducted based on three factors - egg weight, hatching weight, and percentage of egg production each week. The proportions were maintained at 25%, 50%, and 75%, respectively. The egg weight proportions were 0.46, 0.29, and 0.15, while the hatching weight proportions were 0.71, 0.45, and 0.23, respectively. The percentage of egg production each week was 47.92%, 30.18%, and 15.85%, respectively. The study’s results revealed that the intensity of selection and selection response of the Tegal duck were higher when smaller proportions were used in the selection process. It should be noted that the production characteristics used in the selection process were different, which resulted in varying heritability values and selection responses. Overall, these findings suggest that careful consideration of the selection factors is necessary to achieve optimal results in the selection process of the Tegal duck

Abstract

The purpose of this study is to identify the non-genetic variables that have a substantial impact on the milk output per lactation of FH dairy cows at BBPTUHPT Baturraden and to construct variables' correction factors. The National Dairy Cow and Forage Breeding Center (BBPTUHPT) of Baturraden provided the secondary data for the study, which included 1,942 unique records of the amount of milk produced per lactation by 1,015 FH dairy cows born between 2000 and 2014 (a total of 1,015 lactation records). Milk output per lactation, the number of milking days (100–600), the age at calving (575–2,993 days), the lactation phase (lactation 1-6), and the season of birth were among the studied variables. The F test was used to examine the impact of non-genetic factors on the amount of milk cows produce per lactation (ANOVA). The impact of season on milk output per lactation was examined using a student t-test. Utilizing the multivariate least squares method, correction factors were created. Age at calving, which ranges from 1750 to 2000 days, milking days, which range from 300 to 350, and the dry season serve as the primary benchmarks for constructing correction factors. The R program was used to generate and run statistical tests and graphic representation. The findings indicated that the age of calving and lactation period had a very strong correlation (r= 0.94). The number of milking days, age at calving, and season at birth all significantly affected milk output per lactation, with the variance contributing 84.16 percent to the overall variation, according to the results of multivariate analysis (P < 0.01). Actual milk production had a mean (standard deviation) of 3710.55 kg, while adjusted milk production had a mean (standard deviation) of 5167.91 kg. The adjustment parameters can lower the variation in milk production each lactation by 57.92%. (43.00 percent vs 18.09 percent). Conclusion: Non-genetic variability was successfully reduced by correcting milk production data on the number of days of milking, age at calving, and season at birth.

Abstract
