Abstract

Isolation and identification one of cytotoxic fraction have been conducted from the leaves of Andong(Cordyline terminalis Kunth). The fraction having LC50 values of 41,64 ppm contains saponin. Major isolate (7.5 mgwhite amorphous solid) was obtained after a series of chromatographic separations (gravity column chromatography,and preparative high performance liquid chromatography). Identification of the isolate using mass spectrometry withpositive electrospray showed MW of 866 as calculated from the ion peaks m/z 889[M + Na]+, and 867[M + H]+.The ion peaks at m/z 721[(M + H) - 146]+, 703[(M + H) - 164]+, 575[(M + H) - 292]+, 557[(M + H) - 310]+ , 429[(M+ H) - 438]+, and 411 [(M + H) - 456]+ of its fragments indicate the presence of three sugars (two terminal sugarsand one central sugar) from methylpentose moiety with MW 164 eash linked to an agyicone of MW of 428. Protonmagnetic resonance spectrum of the isolate in pyridine-d5 showed characteristic proton signals for three steroidmethyls (two angular methyls and one secondary methyl) at d 1.37 (s), 0.84 (s) and 1.02 (d, J = 6.6 Hz); anexomethylene group at d 4.79 ppm and 4.71 (each br s); an ethylene group at d 5.52 ppm (br d, J = 5.4 Hz); signalsof the protons linked to C26 at d 4.01 and 4.44 ppm (each d, J = 12.0 Hz), and three anomeric protons at d 6.43 ppm(br s); 5.56 ppm (br s) and 4.57 ppm (d, J = 7.0 Hz). From the above data it can be indicated that the isolate resultedis spirostan steroidal saponin.

Abstract

The research about extraction of natural dies from the mixtures of areca nut, betel leaf, gambier, and theeffect of addition of KMnO4 towards surface coloration of albasia wood (Paraserianthes falcataria) was carried out.The extraction of mixture of natural dye was conducted by heating in water for two hours. 10,00 gram of eachsamples of betel leaf, areca nut, and gambier were refluxed and resulted in 28,60%, 40,10%, and 30,20%respectively. According to fitochemical test, the betel leaf extract contain flavonoid, tannin, and carotenoid, whereasthe areca nut and gambier extracts contain flavonoid and tannin.The optimum mass ratio and mass of KMnO4 for coloration were resulted in optimum mass 10:10:5 and0,25 gram respectively. This showed that the addition of KMnO4 strengthen the bond between dye and wood fiberand sharpen the resulted color. This was proven by color endurance test by soaking the object in 1% of detergentwater for 15 minutes. The dye without KMnO4 was easy to fade out, whereas the dye with KMnO4 was moreresistant. The resulted color varies from reddish brown, brown, to deep brown.

Abstract

Wood is one of the few materials that is highly exploited by humans, resulting in increasing usage of low quality wood. High demand and usage on high-quality wood causing heavy reduction of its availability. To increase artistic value of low quality wood, staining on the surface of the wood need to be done. This research aims to apply a mix of natural pigments (Gambier- Piper betle leaves- Areca seeds) and NaCl salt as the mordant on the surface of Albesia wood fiber and test the physical and chemical dye properties with 1% detergent. This study uses a mixture of natural dyes (Gambier- Piper betle leaves- Areca nut) with various concentrations and salt as auxiliaries, carried out with the simultaneous method of mordanting. This study was divided into 7 groups. Differences in mean between groups were tested using One Way ANOVA followed by Post Hoc LSD, which revealed statistically significant test when p value < >0.05. The results showed that there were significant differences of natural dyes mixed applications (Gambier- Piper betle leaves- Areca seeds) in the ratio of 5.0: 3.0: 3.0 to 0.5 g of salt as auxiliaries and 10 minutes of immersion with the simultaneous method of mordanting. Ratio of the dye mixture, on the surface of the wood causes brown color and has the highest absorption average of 0.19 gram and the durability of the dye showed the highest seen from the appearance of color and mix of the missing mass. It can be concluded that the best application is with a mixture of natural dyes and the addition of 0.5 g of NaCl salt on the surface of Albisia wood fiber with the simultaneous method of mordanting and also based on the durability of the dye test is a ratio of 5.0: 3.0: 3.0.

Abstract

The Community Service activity about the manner of making natural mosquito antilarvae liquids from Piper betle L leaf was conducted on the 24st of October 2010 to the mothers of PKK of Tibubiu village, Kerambitan subdistrict, Tabanan district. Making natural mosquito antilarvae liquids from Piper betle L leaf used the simple formulation technique. The liquids were made by using petroleum as a solvent and Piper betle L leaf as a antilarvae agent. The result of this activity and evaluation showed that the mosquito antilarvae liquids product is as good as chemical agent like “abate”. All participants showed enthusiastic during the activity and they will try to make these liquids. All participants, moreover understand about making liquids and they want to try at home. Such public service is interesting to because the peoples have improved knowledge, skill, and possibility to open of new entrepreneur in village so that it can be vacancy and increase income to community.

Abstract

The study about determination of Pb and Cr content in nile fishes (Oreochromis niloticus) found atEstuaria Dam (Badung River Downtream) has been carried out. Some samples were taken every week duringhalf a month. Five grams of dried sample was destructed with a mixture of 10 mL concentrated H2SO4 and40 mL concentrated HNO3. The composition of Pb and Cr in the samples was analyzed Atomic AbsorptionSpectrofotometer using Standar Addition Method.The results showed that the content of Pb in the nile fishes was about 10.1910-10.7710 mg/kg wetsamples and it was over the maximum level of 2.0 mg/kg. On the other hand, the content of Cr wasfound(1.3460-2.9640 mg/kg wet samples) to be out of level as well (maximum 0.4 mg/kg).

Abstract

Lidah mertua plant  has the potential as a natural antioxidant and the antioxidant activity of a plant depends on the number of active compounds extracted. This study aims to determine the yield, total phenol content, total flavonoid content, and IC50 value of lidahmertua leaf extract macerated with ethanol, ethyl acetate, and n-hexane as solvents, and to determine the compounds contained in the lidahmertua leaf extract which have activity antioxidants. The results of the phytochemical test of the lidahmertua leaf extract showed positive results containing polyphenolic compounds, flavonoids, saponins, alkaloids, and steroids. The maceration yield of each solvent was 12.13%, 10.15% and 8.11%, respectively. While the measurement results of the total phenol content of each solvent were 6.96; 3.67; and 1.94 mg GAE/g. The results of the measurement of the flavonoid content of each solvent were 5.82; 3.28; and 1.71 mg QE/g. The IC50 values for each solvent were 162.135 mg/L, 184.561 mg/L and 206.265 mg/L. The extract which has the highest total phenol content, total flavonoid content, and antioxidant activity is in ethanol solvent.

Abstract

Rimpang andong merah (Cordyline fruticosa) ialah tumbuhan obat yang mengandung saponin yang dikenal selaku antiinflamasi. Tujuan riset ini adalah menguji aktivitas antiinflamasi ekstrak etanol rimpang andong merah. Analisis fitokimia dilakukan secara kualitatif dengan uji fitokimia menggunakan reagen pereaksi warna sedangkan uji aktivitas antiinflamasi dengan menggunakan 25 ekor tikus Wistar yang diinduksi dengan karagenan kemudian dikelompokkan menjadi 5 yaitu; (P1) kontrol negatif, (P2) kontrol positif, serta (P3, P4, dan P5) kelompok ekstrak uji dengan dosis 125, 250, dan 500 mg/kg BB. Hasil uji fitokimia menunjukkan ekstrak tersebut terdapat senyawa golongan alkaloid, steroid, fenolik, flavonoid, dan saponin. Dari hasil uji aktivitas antiinflamasi, pemberian ekstrak dosis 125 mg/kg BB memberikan hambatan inflamasi sebesar 65,876% sebaliknya dosis 250 mg/kg BB dapat menghambat inflamasi sebesar 59,994% dan dosis 500 mg/kg BB memberikan hambatan inflamasi sebesar 73,908% selama 360 menit pengamatan. Hasil uji analisis probit memberikan nilai ED50 sebesar 158,48 mg/kg BB. Kata kunci: aktivitas antiinflamasi, andong merah, ekstrak, rimpang. ABSTRACT Rhizome of red andong (Cordyline fruticosa (L.) A. Chev) is a medicinal plant that contains saponins known to be anti-inflammatory. The research aimed to assess the anti-inflammatory activity of the rhizome ethanol extract of red andong. Phytochemical analysis was done qualitatively by using phytochemical reagents. Anti-inflammatory activity was evaluated using 25 Sprague Dawley male rats which had been divided into five groups: negative control (P1), positive control (P2), and group P3, P4, and P5 given extract at doses of 125; 250; and 500 mg/kgBW, respectively. A phytochemical study revealed that the rhizome ethanol extract consisted of alkaloids, steroids, phenolic, flavonoids and saponins compounds. The anti-inflammatory activity test showed that the administration of the extract at a dose of 125 mg/kgBW resulted in an inflammatory inhibition of using 65,876%, on the other hand, a dose of 250 mg/kgBW could inhibit inflammation by 59,994%, and a dose of 500 mg/kgBW had the inflammatory inhibition of 3,908% for 360 minutes of observation. The results of the probit analysis gave an ED50 value of 158,48 mg/kgBW. Keywords: anti-inflammatory activity, extract, rhizome, red andong.

Abstract

Background: Sembung leaves (Blumea balsamifera L.) leaves are proven to contain flavonoid compositions that contain antihyperglycemic properties. This study aimed to evaluate effect of sembung leaves ethanol extract towards serum blood glucose of hyperglicemia wistar rats.Method: This study includes the maintenance of test animals, fractionation and purification using TLC, column chromatography, and LCMS, free radical scavenging activity of 1.1-diphenyl-2-picrillhidrazil (DPPH) as well as experimental methods with pre-post test control group design. Twenty male Wistar rats (Rattus norvegicus) were divided into five groups, the negative control group (aquades), the positive control (glibenclamide 0.18 mg/day/200gbw) and the test group with varying doses of 50 mg/kgbw, 100 mg/kgbw, and 150 mg/kgbw for 14 days and carried out measurement of blood sugar levels in rats. Rats collected data were analyzed using the One Way ANOVA test and continued with the LSD test.Results: Result of LCMS management is flavonoid composition consisting of syringetin with mz=347. In the free radical scavenging activity of DPPH ethylacetate fraction of Sembembal leaf extract IC50 value is 0.5369 μg/ml or 536.9 ppm. Data on rat blood regression levels were analyzed using the One Way ANOVA test and continued with the LSD test each showing a value of p=0.000. This value indicates a significant difference between each group (p<0.05).Conclusion: Ethanol fraction of sembung leaves can reduce protein levels in male wistar hyperglycemia rats. The secondary metabolism of syringetin contained in sembung leaves which contains antioxidants. Latar belakang: Blumea balsamifera (daun sembung) terbukti memiliki senyawa flavonoid dengan khasiat antihiperglikemik. Penelitian ini guna mengkaji efek pemberian ekstrak etanol daun sembung terhadap penurunan kadar glukosa darah pada tikus wistar jantan yang hiperglikemia.Metode: Penelitian ini meliputi pemeliharaan hewan uji, fraksinasi, dan pemurnian menggunakan metode KLT, kromatografi kolom, dan LCMS, uji aktivitas pengikatan radikal bebas 1,1-diphenyl-2-picrylhydrazyl (DPPH) serta metode eksperimental dengan pre-post test control group design. Dua puluh ekor tikus wistar jantan (Rattus norvegicus) dibagi menjadi lima kelompok, kelompok kontrol negatif (aquades), kontrol positif (glibenclamide 0,18 mg/hari/200gBB)  dan kelompok uji dengan variasi dosis fraksi etilasetat ekstrak etanol daun sembung 50 mg/kgbb, 100 mg/kgbb dan 150 mg/kgbb selama 14 hari dan dilakukan pengukuran kadar gula darah pada tikus. Data kadar glukosa tikus dianalisis dengan menggunakan uji One Way ANOVA dan dilanjutkan dengan uji LSD. Pada penelitian ini didapatkan hasil dari fragementasi LCMS adalah senyawa flavonoid yang berupa syringetin dengan mz=347.Hasil: Pada evaluasi aktivitas pengikatan radikal bebas DPPH fraksi etilasetat ekstrak etanol daun sembung memiliki nilai IC50 adalah 0,5369 µg/ml atau 536,9 ppm. Data kadar glukosa darah tikus yang dianalisis dengan menggunakan uji One Way ANOVA dan dilanjutkan dengan uji LSD masing-masing menunjukkan nilai p=0,000. Nilai ini memperlihatkan perbedaan signifikan antar tiap kelompok intervensi (p<0,05).Simpulan:  Fraksi ekstak etanol daun sembung dapat menurunkan kadar glukosa darah pada tikus wistar jantan yang hiperglikemia. Penurunan glukosa darah ini diduga berasal dari metabolit sekunder syringetin yang terkandung dalam daun sembung yang berperan sebagai antioksidan.

Abstract

ABSTRAK: Jahe merah adalah umbi yang mengandung antioksidan berupa terpenoid, polifenol dan minyak atsiri yang sangat berguna untuk menurunkan oksidan dalam tubuh manusia. Tujuan dari penelitian ini yakni menentukan IC50 fraksi dari ekstrak jahe merah, kadar total flavonoid dan fenol yang terdapat pada jahe merah dengan aktifitas antioksidan terkuat, dan mengidentifikasi senyawanya. Metode DPPH digunakan untuk menentukan aktivitas antioksidan dan metode Folin-Ciocalteu untuk menentukan total flavonoid dan fenol serta identifikasi senyawa aktif menggunakan LC MS/MS. Nilai IC50 fraksi air, n-heksan, dan fraksi kloroform secara berutut-turut 52,92 µg/ml, 149,92 µg/ml, dan 108,10 µg/ml. dengan total flavonoid dan fenol fraksi air 34,9045 mg/g QE dan 2,37 mg/g GAE. Hasil identifikasi LCMS/MS menunjukkan fraksi air jahe merah mengandung Senyawa antara lain 6-gingerol, Zerumbone, 6-shogaol dan 10-gingerol yang muncul pada waktu retensi 3.89; 5.90; 8.60 dan 10.11. Disimpulkan bahwa fraksi air jahe merah mempunyai aktivitas antioksidan terkuat dengan kandungan senyawa aktif yang berkontribusi sebagai antioksidan. ABSTRACT: Red ginger is a spice that contains antioxidant compounds of terpenoids, polyphenols, and essential oils which are very useful for reducing oxidants in the human body. The IC50 determination of fraction from extract red ginger, the total levels of flavonoids and phenols found in red ginger with the strongest antioxidant activity, and to identify the compounds. The antioxidant activity determination using DPPH methods and the total flavonoids and phenols determination using Folin-Ciocalteu methods meanwhile the identification of active compounds using LC-MS / MS. The antioxidant activity test showed that the IC50 values ??of the water fraction, n-hexane, and chloroform fraction were 52,92 µg/ml, 149,92 µg/ml, and 108,10 µg/ml, respectively. with total flavonoids and phenols, water fraction 34.9045 mg/g QE, and 2.37 mg / g GAE. The results of LCMS / MS identification showed that the red ginger water fraction contained compounds are 6-gingerol, Zerumbone, 6-shogaol, and 10-gingerol which appeared at retention time of 3.89; 5.90; 8.60, and 10.11. Conclusion is red ginger water fraction has the strongest antioxidant activity with active compounds that contribute as antioxidants.

Abstract

Kecombrang (Etlingera elatior Jack) is a plant that has potential for hypolipidemia. Kecombrang has secondary metabolites consisting of tannins, saponins, alkaloids, steroids, terpenoids, phenolics, and flavonoids. The purpose of this study was to determine the hypolipidemic activity of kecombrang flower methanol extract in obesed Wistar rats fed a high-fat diet. The test animals used in this study were 18 female white rats aged two months, weight of ± 150 grams. The test animals were divided into three groups, namely normal control (only given standard feed), high-fat control (obesed rats with high-fat diet), and flower treatment group (obesed rats with a high-fat diet + flower methanol extract 100 mg/kgBW) orally for 30 days. The results showed that kecombrang flowers reduced total cholesterol levels by 55.5%, triglycerides (TG) by 57.4%, LDL by 76.73%, and increased HDL by 42.96%. The kecombrang flower has the potential for hypolipidemia which is characterized by a decrease in total cholesterol, TG, LDL, and can increase HDL levels in obese rats significantly (p<0.05).
