Abstract

The purpose of this study was to determine toxic symptoms (loss of reflex), enzyme activity of SGOT and SGPT, and liver histopathology of experimental mice (Mus musculus) exposed to a combination formulation of 96% ethanol extract spray of Solanum melongena L. and Cananga adorata flower oil. True experimental research design with Post Test Only Control Group Design, this study used 4 treatment groups and 1 control group, data collected in the form of toxic symptoms (loss of reflex), SGOT enzymatic activity, and liver histopathological picture of experimental mice (Mus musculus). Which was analyzed by One Way Anova and Descriptive test. The results showed that the changes in toxic physical symptoms (loss of reflexes) of experimental mice (Mus musculus) after 30 minutes of spraying were strange reactions to walking around without direction, sensitivity to touch, changes in social interaction colliding with each other, lethargy, eye opacity and excessive blinking, respiratory rate. increased, which returned to normal at 4 hours to 168 hours after spraying. The activity of the SGOT and SGPT enzymes after 168 hours of spraying showed that the higher the dosage of the formulation, the higher the activity of the SGOT and SGPT enzymes, but the increase was still within normal limits, the histopathological results of liver morphology grading at various research doses showed Normal and Mild. Conclusion: Solanum melongena L. and Cananga adorata flower oil combined did not cause pathological effects in experimental mice (Mus musculus).

Abstract

Before using medicinal plant products, quality control is required to determine their quality and consistency. Yacon leaves (Smallanthus sonchifolius) in preclinical trials have pharmacological effects to lower postprandial glucose with enhydrin as the marker compound. This study aimed to develop and validate a TLC-Densitometry method for determining enhydrin in Yacon leave extract. The stationary phase was TLC plates of silica gel GF254, while a mobile phase was the mixture of chloroform:hexane in the ratio of 10:1 was selected for the chromatographic system. Then, the TLC separation was followed by densitometric analysis at 210 nm. The method was validated in terms of linearity, accuracy, precision, limit of detection (LOD), and limit of quantitation (LOQ), and found to be linear, with a correlation coefficient (R2) 0.9998. The LOD and LOQ values calculated from the calibration curve were 80.57 and 244.1µg/mL, respectively. The method also exhibited acceptable precision, with relative standard deviation values lower than 3%. The accuracy of the method was evaluated based on the recovery percentages, and found to be within an acceptable range (97–107%).

Abstract

Cancer has a high prevalence and mortality in the world. Cancer treatment is still hampered by high levels of side effects, drug resistance and the lack of affordable prices for anticancer drugs. It is necessary to develop new anticancer drugs to help overcome this problem. One of the plants that has the potential to be developed as an anticancer drug is Sungkai (Peronema canescens Jack). This study aims to determine the cytotoxic activity of sungkai leaf and identify its metabolomic profile. The sungkai leaves were macerated for 24 hours with 96% ethanol. Metabolomic profiles were analyzed with Ultra High-Performance Liquid Chromatography-High Resolution Mass Spectrometry (UHPLC-HRMS). Chemical structure identification was performed by MS-DIAL and MS-FINDER platforms. The ethanol extract of sungkai leaves was analyzed for its cytotoxic activity using the MTT test on A549 lung cancer cells and its selectivity on normal HDFa fibroblast cells. The ethanol extract of sungkai showed IC50 105,21 µg/mL on A549 cells and no cytotoxic activity against normal HDFa cells. Based on the metabolomic analysis, 7 furano terpenoid compounds were detected in the ethanol extract, namely peronemin A2; A3; B1; B2; B3: C1; and D1 along with other compounds. In conclusion, the ethanol extract of P. canescens leaves has cytotoxic and selective activity against A549 lung cancer cells, and potential to be further developed as an anticancer drug candidate. Peronemins and other substances like flavonoids and polyphenols may be linked to the cytotoxic properties of sungkai leaves

Abstract

The Sesquiterpene lactones, enhydrin and uvedalin, are the chemotype and subtype compounds of the yacon leaf. They are known to have anti-diabetic activity, but their mechanisms are not clear. One of the mechanisms of anti-diabetic agents is to inhibit the activity of the dipeptidyl peptidase 4 enzyme (DPP-4). The research aimed to determine the inhibition of the extract, enhydrin, and uvedalin isolated from the yacon leaf against the DPP-4 enzyme activity. Yakon leaves were extracted using 70% ethanol by the maceration method. Enhydrin and uvedalin were obtained from previous research. The inhibitory activity of the DPP-4 enzyme was then determined by the fluorescence assay method using a multi-well plate reader. Sitagliptin was used as standard inhibitor of DPP-4 enzyme. The ethanol extract of yacon leaves inhibits the DPP-4 enzyme with an IC50 of 856.9 ppm. The percentage inhibition of the DPP-4 enzyme by the enhydrin and uvedalin at a concentration of 250 ppm was 2.37 and 35.16%, respectively. The inhibitory potency of the pure isolated compounds was not substantially greater than that of the crude extract itself. This led us to the conclusion that the overall contribution of enhydrin and uvedalin to the extract's DPP-4 inhibitory activity is actually modest, suggesting the presence of other active compounds within the extract. However, it may provide a new approach to the treatment of type 2 diabetes.
