Abstract

Rumput Knop (Hyptis capitata Jacq.)  banyak dimanfaatkan sebagai obat tradisional  oleh masyarakat suku Toraja.  Informasi  ilmiah tentang pemanfaatan Rumput Knop dan kandungan metabolit sekundernya yang berkhasiat sebagai obat tradisional  masih terbatas.  Penelitian ini dilakukan untuk mengumpulkan informasi dari masyarakat suku Toraja mengenai khasiat Rumput Knop dalam mengatasi berbagai penyakit, variasi organ tumbuhan yang dimanfaatkan sebagai obat dan cara pengolahannya. Data pemanfaatan rumput knop diperoleh melalui wawancara. Untuk mendukung data khasiat, dilakukan uji fitokimia kualitatif meliputi uji alkaloid, triterpenoid, steroid, saponin, tannin dan flavonoid. Rumput knop diaplikasikan untuk beberapa penyakit seperti sakit perut, sakit kepala, demam dan luka terbuka. Organ tumbuhan yang sering digunakan dalam pengobatan adalah daun muda. Cara pengolahan yang paling banyak dilakukan adalah perebusan. Berdasarkan hasil tersebut, maka organ yang dipilih untuk dilakukan skrining fitokimia adalah daun dan metode ekstraksi metabolit sekundernya dengan metode infusa. Skrining fitokimia menunjukkan infusa daun Rumput Knop mengandung flavonoid, steroid dan tannin.  Kata Kunci: Etnobotani, Infusa, rumput knop, nutrasetika

Abstract

Rumput Knop (Hyptis capitata Jacq.) memiliki sejarah etnobotani. Informasi mengenai keamanan pemanfaatan rumput Knop sebagai obat tradisional perlu untuk dilakukan. Uji keamanan dilakukan dengan uji toksisitas. Uji toksisitas akut dengan Brine Shrimp Lethality Test (BSLT) dilakukan sebagai uji toksisitas awal. Batang rumput Knop diekstrak dengan  metode maserasi, menggunakan penyari etanol. Ekstrak etanol batang rumput Knop dibuat dengan berbagai konsentrasi, yakni 1000, 500, 250, 125 dan 62,5 µg/ml. Larva Artemia salina (L.) diberi paparan ekstrak etanol batang rumput Knop selama 24 jam. Toksisitas ekstrak ditentukan berdasarkan jumlah larva yang mati setelah paparan ekstrak. Jumlah larva yang mati, dianalisis dengan analisis probit untuk menentukan nilai LC50. Kandungan senyawa fitokimia dalam ekstrak etanol batang rumput Knop diuji dengan uji fitokimia kualitatif. Besar nilai LC50 ekstrak etanol batang rumput Knop adalah 880,579 µg/ml. Ekstrak etanol batang rumput Knop mengandung steroid.

Abstract

AbstrakKegiatan Pengabdian kepada Masyarakat dengan topik “Pelatihan tentang Pengenalan, Pemeriksaan, dan Penjaminan Mutu Bahan Obat Tradisional (BOT) bagi Guru Biologi SMA Daerah Istimewa Yogyakarta” dilakukan mengingat banyaknya kasus keracunan akibat konsumsi obat tradisional. Guru Biologi SMA menjadi sasaran pelatihan karena telah memiliki latar belakang keilmuan dan keterampilan yang memadai serta sering menghadapi pertanyaan orangtua murid dan masyarakat perihal pemanfaatan BOT. Pelatihan diharapkan memberikan solusi terhadap permasalahan di atas dengan menerapkan teknologi tepat guna yang sederhana, sehingga dapat memberikan pemahaman tentang BOT berkualitas berdasarkan Cara Pembuatan Obat Tradisional yang Baik (CPOTB). BOT yang dipilih yaitu sambiloto (Andrographis paniculata), mahkota dewa (Phaleria macrocarpa), dan pule (Alstonia scholaris) karena banyak digunakan dalam pengobatan diabetes mellitus. Peserta diberikan keterampilan mengenali, mengidentifikasi, dan melakukan pemeriksaan mutu secara sederhana, namun ilmiah. Peserta menyatakan mampu menerapkan keterampilan yang diperoleh dan bersedia mengikuti pelatihan lanjutan di masa mendatang. Kata kunci: Pengenalan, Pemeriksaan, Penjaminan Mutu, Bahan Obat Tradisional  AbstractCommunity Service Activity with the topic “Training on Determination, Identification, and Quality Control of Traditional Medicine Ingredients to Biology High School Teachers in Yogyakarta Special Province” was done due to the increase of traditional medicine intoxications. Biology High School teachers were targeted as the participants, because they had scientific background and skills related to the problem. The activity was done to give better solution to the problem using simple and appropriate technology, hence it can give knowledge on high quality of traditional medicine based on Good Production of Traditional Medicine. Traditional medicines utilised were sambiloto (Andrographis paniculata), mahkota dewa (Phaleria macrocarpa), and pule (Alstonia scholaris), since these traditional medicines were mostly practiced to cure diabetes. Participants were also given skills on determination, identification, and quality control. Participants stated that they were able to apply the skills obtained and were ready to be included in the continual training in the future. Key words: Determination, Identification, Quality Control, Traditional Medicine

Abstract

Larvicidal potential of Hyptis capitata grown in Indonesia has not been extensively studied. Its leaves are extracted with the maceration method using ethanol as the solvent. Furthermore, this study aims to use the ethanolic leaf extract of the H. capitata for larvicidal assays against instar III/IV larvae of Culex quinquefasciatus with different concentrations, including 1000, 500, 250, 125, and 62.5 µg/mL. Fractionation of the extract was carried out by vacuum liquid chromatography, and obtained four fractions, namely fractions F1, F2, F3, F4. Fractions were also used for the larvicidal assay. The constituents of the extract were then analyzed with the GC-MS method to predict the components involved in its toxicity. Larvicidal data obtained were analyzed using regression analysis to determine the LC50 value. Analysis of variance was carried out with one-way ANOVA using Tuckey HSD-test on the SPSS ver. 26 at 95% confidence and significance P<0.05. H. capitata ethanolic leaf extract has a higher level of toxicity than its fraction, with LC50 value 990.90 µg/mL. H. capitata ethanolic leaf extract are capable of providing a toxic effect on larvae, with a mortality percentage of up to 50% more. Some compounds that were assumed to play a role in its toxicity include pentadecanoic acid, 2,6,10,14-tetramethyl-, methyl ester; 1-heptadecyne; 9-tetradecen-1-ol, acetate; oxyrane, deodecyl-; 9,12,15-octadecatrienal; and 6,11-dimethyl-2,6,10-dodecatrien-1-ol. These finding indicate that the ethanolic leaf extract of H. capitata has the potential to be developed as a biolarvicidal agent against C. quinquefasciatus.

Abstract

Pesut merupakan mamalia perairan bersifat fakultatif yang dapat ditemui di wilayah Asia Tenggara. Populasi pesut, terutama pada perairan tawar menghadapi berbagai tekanan untuk dapat bertahan hidup sehingga digolongkan sebagai Critically Endangered pada daftar merah IUCN. Pemahaman mengenai sejarah evolusi dan aspek apa yang memfasilitasi adaptasi pesut pada perairan tawar belum banyak diketahui. Adaptasi penuh terhadap perairan tawar pada beberapa spesies lumba-lumba menunjukan adanya seleksi positif pada gen NADH subunit 2 (NADH2) di DNA mitokondria yang diperkirakan berkaitan dengan mekanisme translokasi jalur proton untuk efisiensi energi yang diperlukan dalam kolonisasi perairan tawar. Oleh sebab itu, penelitian ini dilakukan untuk mengetahui apakah gen NADH subunit 2 (NADH2) pada DNA mitokondria memfasilitasi kolonisasi pesut pada perairan tawar seperti dijumpai pada Cetacea perairan tawar lain. Melalui pendekatan bioinformatika sederhana, hasil dari penelitian ini menunjukan tidak adanya subtitusi basa yang merubah susunan asam amino Threonin menjadi Alanin pada urutan asam amino ke 297 gen NADH2 pada populasi pesut yang mendiami perairan tawar dan pesisir. Hal ini menunjukan tidak adanya seleksi adaptif pada gen NADH2 yang diperkirakan terjadi akibat kolonisasi perairan tawar yang relatif baru,  sehingga perubahan asam amino belum teramati pada populasi pesut.

Abstract

Roses are one of the commodities in the Sapuangin area, Pemalang, Klaten. The fluctuating selling price and short shelf life make fresh roses only a secondary product. Therefore, a training is needed to process rose flowers into products with high economic value and longer shelf life. This community service aims to enhance the community's knowledge in processing various products from rose flowers. The activities include survey, socialization, training implementation, and evaluation. The training consists of 7 topics from June to July 2023, where participants receive oral explanations and practical demonstrations of the materials provided. The evaluation includes assessing the participants' knowledge through questionnaires given at the beginning and end of the community service, as well as satisfaction evaluation. The entire community service runs smoothly, with participants able to practice making raw materials, solid soap, bath salts, aromatherapy candles, rosewater, and hand sanitizer. Participants also learn about the different types of roses. The average score of the questionnaires from 15 respondents before the training was 21.33 ± 2.61, increasing by 53.49% to 32.74 ± 3.90 from 19 respondents. 68.42% of participants expressed very satisfied, and 31.58% expressed satisfied with participating in the community service activities. In conclusion, the community service has improved the knowledge and skills of the Sapuangin Village community in creating products from rose flowers that have higher economic value and longer shelf life. Keywords — roses, soap, candles, hand sanitizer, rosewater   Abstrak— Mawar merupakan salah satu komoditi di kawasan Sapuangin, Pemalang, Klaten. Harga jual yang tidak menentu dan masa simpan pendek menyebabkan mawar segar hanya menjadi produk sampingan. Oleh karena itu diperlukan kegiatan pelatihan pengolahan bunga mawar menjadi produk dengan nilai ekonomis tinggi dan masa simpan panjang. Pengabdian pada masyarakat ini bertujuan untuk meningkatkan pengetahuan masyarakat dalam mengolah bunga mawar menjadi berbagai macam produk. Kegiatan meliputi tahap survey, sosialisasi, pelaksanaan pelatihan, dan evaluasi. Pelatihan terdiri dari 7 materi mulai Juni-Juli 2023, dimana peserta diberi penjelasan secara lisan dan mempraktekkan secara langsung materi yang diberikan. Evaluasi meliputi evaluasi tingkat pengetahuan melalui kuesioner yang diberikan di awal dan di akhir kegiatan pengabdian, serta evaluasi kepuasan. Seluruh kegiatan pengabdian berjalan lancar, peserta dapat mempraktekkan pembuatan simplisia, sabun padat, garam mandi, lilin aromaterapi, air mawar dan handsanitizer. Peserta juga mengetahui jenis-jenis bunga mawar. Nilai rata-rata kuesioner dari 15 responden sebelum pelatihan adalah 21,33 ± 2,61, meningkat sebesar 53,49% menjadi 32,74 ± 3,90 dari 19 responden. 68,42% peserta menyatakan sangat puas dan 31,58% menyatakan puas mengikuti kegiatan pengabdian. Dapat disimpulkan bahwa kegiatan pengabdian dapat meningkatkan pengetahuan dan kemampuan masyarakat Desa Sapuangin dalam membuat produk dari bunga mawar yang memiliki nilai ekonomis lebih tinggi dengan masa simpan panjang.

Abstract

Sapuangin area is one of supporting areas for Mount Merapi National Park (TNGM). The endemic Merapi orchid is widely cultivated by people in the Sapuangin, using conventional cultivation. Sapuangin’s youth, who are members of a tourism group (POKWIS) have never received training regarding orchid propagation using in vitro culture principles. This is important to support orchid conservation activities carried out by POKWIS in the Sapuangin greenhouse. Based on requests from the community, lecturers and educational laboratory staff concentrating on Technobio-Industrial Studies, Faculty of Biotechnology, Universitas Atma JayaYogyakarta, provided a series of training on in vitro orchid culture, for household scale. This activity is expected to provide knowledge and skills regarding orchid cultivation techniques for the community. The activity was held on 12-20 December 2023. The training activity was divided into several topics, namely introduction to orchid types and their uses, principles of in vitro orchid culture, introduction to tools, methods for sterilizing and sowing orchid seeds, pollination/pollination, overplanting, and acclimatization. The training carried out can provide additional knowledge for the community regarding in vitro orchid cultivation techniques. The implementation of the training was considered very useful and by community needs, the training resource was competent, the implementation time was by the plan, and was supported by complete facilities and infrastructure. The community hopes for ongoing guidance and assistance regarding orchid propagation in the future.

Abstract

Hyptis capitata Jacq. known as Sumambu plants in Sulawesi, has phytopharmaceutical importance. H. capitata extracts were evaluated for their phytochemical properties, antioxidant activity, and cytotoxicity. Using the maceration yielded five types of extracts: root chloroform (RC), root methanol (RM), leaf chloroform (LC), leaf methanol (LM), and leaf ethanol (LE). Phytochemical properties were identified by qualifying procedure and digital image analysis for quantifying Red-Green-Blue (RGB) percentage and hex colour code. 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay was used to determine the half-maximal inhibitory concentration (IC50). Cytotoxicity screening of each extract was performed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay against HeLa and 4T1 cells. Gas Chromatography-Mass Spectrometry (GC-MS) assay was used to identify the phytochemical compounds of the extracts with the most promising potential. Alkaloids were the major constituents of the phytochemicals of RC, RM, LE, LC, and LM. RM and LM have potency and weak free radical scavenging activities, with IC50 value 31.08 and 58.03 µg/mL, respectively. The IC50 of RC and RM against HeLa cells were 84.21 ± 0.63 and 172.10 ± 02.90 µg/mL, respectively. Meanwhile, the cytotoxicity of RC and RM against 4T1 cells were 86.42 ± 0.80 and 182.82 ± 7.00 µg/mL, respectively. It means RC and RM exhibit a moderate level of cytotoxicity in both HeLa and 4T1 cells. LM shows moderate cytotoxicity, but it is limited to 4T1 cells with an IC50 value of 181.86 ± 12.68 µg/mL. The cytotoxicity level of extracts was lower than doxorubicin. Campesterol, ferruginol, stigmasterol, cis-13-octadecenoic acid methyl esters, and methyl palmitate were predicted to play a role in the antioxidant activity and cytotoxicity of RC, RM, and LM. RC, RM, and LM possess the potential for development as anticancer agents. Moreover, RM shows promise as an antioxidant due to its notable radical scavenging activity. Further research is required to explore the cytotoxic effects of RC, RM, and LM on normal cells and to assess their toxicity in experimental animals.

Abstract

Sansevieria, commonly known as Dracaena trifasciata. Apart from serving as an ornamental and air-purifying plant, it also exhibits medicinal properties, making it a potential candidate for the development of novel natural drugs. The demand for natural antibacterial agents has significantly increased due to the rising incidence of resistance among pathogenic bacteria, such as Staphylococcus aureus. This research conducted to determine the antibacterial and antioxidant activities of D. trifasciata leaf extracts. The D. trifasciata leaf material was sequentially extracted using sonication, beginning with chloroform and then followed by ethanol. The Kirby-Bauer method was employed to conduct the antibacterial test, wherein the inhibition zones generated by the chloroform and ethanolic extracts were measured. The test bacteria used in this study was Staphylococcus aureus. The assessment of the antioxidant activity was conducted using the DPPH (2,2-diphenyl-1-picrylhydrazyl) method. Phytochemical compound identification in the extracts was conducted using GC-MS analysis. The ethanolic extract showed a higher yield percentage compared to the chloroform extract. Both extracts exhibited moderate antibacterial activity. The chloroform extract exhibited an inhibition zone of 73.33 mm2, while the ethanolic extract showed an inhibition zone of 110 mm2. The chloroform extract had a minimum inhibitory concentration (MIC) of 500 mg/mL and a minimum bactericidal concentration (MBC) of 1000 mg/mL. The ethanolic extract had an MIC of 31.25 mg/mL and an MBC of 62.50 mg/mL. In terms of antioxidant activity, the chloroform extract had an IC50 of 370.8±0.07 µg/mL, while the ethanolic extract had an IC50 of 647.4±0.12 µg/mL. GC-MS analysis revealed 47 compounds in the chloroform extract and 49 compounds in the ethanolic extract. Based on the results of yield, antibacterial and compound identification, the ethanolic extract of D. trifasciata leaves exhibited greater potential compared to the chloroform extract. Its antibacterial activity showed more promise than its antioxidant activity

Abstract

Kegiatan pengabdian masyarakat ini dilakukan di Paroki St. Maria Bunda Penasihat Baik, Wates. Tujuan kegiatan adalah meningkatkan kesadaran dan pemahaman masyarakat tentang prinsip-prinsip Laudato Si' serta pemanfaatan dan budidaya tanaman berenuk (Crescentia cujete) serta pembuatan ekoenzim sebagai salah satu bentuk pengolahan sampah organik. Metode yang digunakan meliputi survei lokasi, sosialisasi, pelatihan, dan evaluasi. Pelatihan mencakup pembuatan produk berbasis berenuk seperti sari buah, masker-jelly, teh daun berenuk, serta pemanfaatan sampah organik menjadi ekoenzim. Hasil kegiatan menunjukkan peningkatan pengetahuan warga gereja tentang pemanfaatan tanaman berenuk sebagai sumber senyawa fitokimia yang bermanfaat serta metode pengolahan sampah menjadi produk bernilai guna. Evaluasi menunjukkan tingkat kepuasan masyarakat yang tinggi terhadap pelaksanaan kegiatan ini. Program ini tidak hanya meningkatkan kesadaran lingkungan tetapi juga memberikan keterampilan baru yang aplikatif kepada masyarakat setempat.
