Abstract

Surface sterilization is a very important key for reaching a successful explants introduction to the in vitro environment. However, to get the best protocol of explants surface sterilization is not easy. Each explants has its characteristics and the contaminants (which usually are bacteria and fungus) are vary depending on its source. In order to know the best sterilization protocol of Phaleria macrocarpa leaves, some sterilizing agents were tested. This experiment suggests that ethylene bis dithiocarbamate may eradicate fungal infection by metalloenzymes inhibition, which is showed by 91.7% survivals. In the other hand, streptomycin sulfate may eliminate bacterial infection by inhibition of protein synthesis, which is showed by 95.8% to 98.3% survivals depending on the exposure time. Abstract

Tomato (Lycopersicon esculentum Mill.) is used as vegetable or fruit by people around the world. The effective and efficient propagation of tomato is needed due to the high demand, while its availability is not enough to meet the consumer demand. In vitro flowering and fruiting is useful in order to produce high quality hybrid seeds. Also, this can be produced in all seasons. Beside, this technique is beneficial to improve genetic diversity in tomato. In addition, tomato has natures that make it compatible as a plant model. This experiment intended to find the best combination of plant growth regulators or plant retardants to induce flower and fruit from tomato plantlets. The results will be beneficial to overcome self-fertilization in tomato, as well as to promote higher genetic biodiversity in tomato. To do so, some plant growth regulators (6-Benzylaminopurine (BAP), Indoleacetic acid (IAA), and Gibberellin (GA3)) and retardants (Ancymidol and Paclobutrazol) were used to find the best combination in inducing in vitro flowering and fruiting. The results showed that 1 mg.L-1 BAP was the best candidate plant growth regulator to produce the in vitro flowers and fruits from the treated plantlets.

Abstract

Tomatoes are one of the most popular vegetable plants globally because of the many benefts that can be obtained from tomatoes. However, its availability is still insuffcient for market demand. Therefore, researchers began to develop tissue culture methods to cope with market demand. Although there have been several studies on tomato flowering in vitro, information about this method is minimal and needs further development. In vitro flowering is a technique in tissue culture for flowering plants in vitro. One of the benefts is cultivating plants in a relatively short time under controlled conditions to prevent disease transmission between plants. Therefore, we conducted a literature study to compare the results of these studies to know an effective and efcient method of factors that increase the in vitro flowering plants of tomatoes. In vitro flowering methods generally focus on flowering media, especially cytokinin types such as 6-Benzylaminopurine (BAP) and kinetin, and the concentration of those hormones, also light intensity used to induce flowering of tomato plants. Factors that affect in vitro flowering of tomato plants are tomato plant varieties, hormones, and light intensity.

Abstract

Buah tomat merupakan salah satu buah yang sering dijumpai di Indonesia. Permintaan tomat juga terus meningkat dari tahun ke tahun di Indonesia. Keterbatasan lahan menjadi kendala dalam memenuhi permintaan tersebut. Oleh karena itu, tomat dapat dikembangkan melalui media kultur jaringan untuk menghasilkan buah yang sehat dan cepat. Salah satu faktor yang menentukan keberhasilan dalam budidaya tomat melalui media kultur jaringan adalah intensitas cahaya yang digunakan. Metode multiple linear regression diimplementasikan untuk dapat memperkirakan intensitas cahaya yang tepat. Fitur yang akan menjadi prediktor dalam penelitian ini adalah jenis media tanam dan panjang kecambah yang diharapkan. Berdasarkan evaluasi yang telah dilakukan, nilai mean absolute error (MAE) yang dihasilkan yaitu 0,87 dengan rasio data training dan testing adalah 90:10. Hal ini menandakan bahwa sistem cukup akurat dalam melakukan prediksi.

Abstract

Kelompok warga RT 03 RW 07 Mejoyo Surabaya adalah masyarakat yang aktif dan rajin. Kampung ini selalu berprestasi khususnya dalam hal lingkungan hidup dan Usaha Mikro Kecil dan Menengah (UMKM). Program pengabdian masyarakat yang didanai DIKTI ini bertujuan untuk memberdayakan warga kampung dalam hal pertanian sayur organik dengan memanfaatkan lahan sempit yang merupakan sepetak tanah fasilitas umum dari Universitas Surabaya (UBAYA). Selain itu, hasil budidaya sayur diolah sehingga terdapat nilai tambah dan solusi jika panen berlebihan atau hasil panen tidak laku terjual. Agar rintisan usaha ini sukses, warga juga dibekali dengan manajemen keuangan dan pemasaran. Peningkatan kemampuan masyarakat dalam hal pertanian organik vertikultur diukur dengan memberikan pre test dan post test. Hasilnya adalah adanya peningkatan keterampilan warga, yaitu sebesar 16%. Sedangkan keterlibatan mahasiswa Merdeka Belajar Kampus Merdeka (MBKM) di dalam program ini dinilai sangat efektif untuk memperkaya pengalaman mahasiswa dalam mengabdi di masyarakat. Hal ini tertuang dalam nilai keterlibatan masing-masing siswa yaitu antara A dan B.

Abstract

RT 03 RW 07 Mejoyo merupakan kampung smart yang mendapat beberapa penghargaan karena produktivitas mereka, seperti menang dalam lomba UMKM antar kampung. Lahan fasum yang mulai dimanfaatkan dengan tanaman vertikultur. Namun hal itu belum optimal, hasil panen tanaman vertikultur dan produk UMKM belum terjual dengan luas, serta beberapa warga masih menganggur. Untuk itu dilakukan pelatihan marketing yang terdiri dari pelatihan digital marketing, kemasan, membuat konten promosi, cara foto produk yang baik, hingga pelatihan pengurusan legalitas. Metode pelatihan yang digunakan adalah workshop yaitu pemaparan dua arah dan pelatihan langsung di tempat. Sehingga warga langsung bisa mempraktikkan materi dengan langsung didampingi oleh narasumber. Hasil dari workshop adalah 89% warga menyatakan materi sangat memuaskan, dan 94% menyatakan penyampaikan materi sangat memuaskan. Pada akhirnya diharapkan semua produk warga RT 03 RW 07 terjual dengan luas, berkembang, dan warga mendapatkan profit.

Abstract

Introduction: Plant Growth Regulators (PGRs) play a role in regulating organogenesis and morphogenesis in shoots, roots, and callus formation. Color spectrum of light is one of the quality light factors that affects plant physiological processes. This study aimed to determine the effect of cytokinin and auxin on Murashige-Skoog (MS) medium and light on callus induction and proliferation of porang (Amorphophallus muelleri Blume.) var. Madiun1. Methods: This study used completely randomized design, with the first factor was PGRs (combination auxin and cytokinin) and the second was color spectrum of light (white light, blue light, and its combination) during incubation. Variables observed were emergence time, color, texture, structure and calli growth, also shoots emerging from calli. Results: The results showed an interaction between PGRs with a combination of light color spectrum on callus growth. The fastest callus growth occurred in combination 5.0 mg.l-1 6-benzyl amino purine (BAP) with 0.2 mg.l-1 naphthalene acetic acid (NAA) which was incubated in a combination of white and blue light for 16 hour irradiation. The combination 5.0 mg.l-1 BAP with 0.2 mg.l-1 NAA was able to induce callus emergence time, and the shoots appearing were faster, whereas combination of white and blue light was able to accelerate callus emergence from bulbil and adventitious shoots emergence. Conclusion: The combination of white and blue light color spectrum for 16 hours irradiation can accelerate callus emergence from bulbil and adventitious shoots emerging from calli, and interaction with combination of 5.0 mg.l-1 BAP and 0.2 mg.l-1 NAA can accelerate porang’s callus growth.

Abstract

Endophytic bacteria play an important role for plants in assisting the efficient absorption of nutrients from the surrounding environment. Studies on endophytic bacteria in Avicennia marina plants are still relatively rare. This research aims to determine abilities of three bacterial endophytes and a control Bacillus subtilis infecting the host plant A. marina and conducted in ekowisata mangrove Gunung Anyar, Surabaya. Each endophyte isolates at the concentration of 102, 103, and 104 CFU/mL were used for inoculation, whereas each concentration was applied to 3 plants (triplicate). Every ± 1 m tall A. marina plant was injected with 2 × 500 µL of bacterial solution. Another three plants were chosen and injected with Lactose-Broth (LB) sterile medium. The leaf length and width of 2-4 leaves per plant were measured at day 0 before inoculation and after two weeks of inoculation. Data analysis using paired t-test resulted in no change in leaf area average before and after treatments. Despite the result, we observed that some of the treated plants produced flowers, whereas no flower was observed on control plants. In addition, there were infection zones detected around the infection area. Furthermore, we have observed Indole Acetic Acid (IAA) and siderophore production in all bacterial isolates. The results of this study show that acceleration in the development of A. marina, evidenced by a phase shift from vegetative to generative occurring within a two-week timeframe.
