Abstract

Dipeptidyl Peptidase-4 is a epithelial transmembrane protein of many cell type such as gastrointestinal tract, biliary tract, pancreas, kidney, thymus, uterus, prostate and capillary endothelial cell. It has a cytoplasmic domain (amino acids 1-6), transmembrane domain  (amino acid 7-28) and extracellular domain (amino acids 29-766). Dipeptidyl Peptidase-4 also found in soluble form of serum and other body fluid. Soluble DPP-4 has not a cytoplasmic domain and transmembrane domain. The level of soluble dipeptidyl peptidase-4 increased in diabetes. In diabetes, Hydrogen Peroxide as Reactive Oxygen Species (ROS) involved in transmembrane DPP-4 shedding to produce soluble DPP-4. Hydrogen peroxide can be converted into hydroxyl radicals (•OH) which are highly reactive. Hydroxyl radicals can oxidize  fatty acid such as linoleic acid which is found mainly in Low Density Lipoprotein (LDL). This study was aimed to elucidate the molecular mechanism of oxidation to shed  transmembrane DPP-4 to produce soluble DPP-4. The DPP-4 model used is DPP-4 with 10-766 amino acid derived from Uniprot and 3D Jigsaw program. Oxidized linoleic acids were derived from NCBI pubchem. DPP-4 as a receptor was docked with oxidized linoleic acids as ligand by patchdock program. And the result show that 13-HODE more easily interact acid with DPP-4than  the other oxidized linoleic. That  oxidants had the active site on DPP-4 (Threonine 36 and Alanine 37) that allow the shedding of transmembrane DPP-4 to produce soluble DPP-4. But that oxidant also interact with the other  site  of DPP-4 that may destroy the catalytic domain.Keywords: DPP-4; oxidized linoleic acid; shedding

Abstract

Okra (Abelmoschus esculentus) is reported to have various biological functions such as antioxidant, anticancer and anti-inflammation. However, only few studies have been reported immunomodulatory activities of okra to prevent disease caused by bacteria infection. In this study, the immunomodulatory activities of polysaccharides from okra pods were investigated further through the spleen weight and B-lymphocytes proliferation in mice infected by Staphylococcus aureus. Okra polysaccharides were obtained by water extraction and ethanol precipitation. Okra polysaccharides with doses of 25, 50, 75, and 100 mg/kg BW were orally administrated to mice with or without Staphylococcus aureus infection. Spleen weight was evaluated in both treatment group and control group. B-lymphocytes proliferation was evaluated by MTT assay using LPS induction. Results showed that okra polysaccharide at the doses of 50, 75, and 100 mg/kg increased spleen weight (p < 0.05) significantly. While at the dose of 75 and 100 mg/kg, it increased the B-lymphocytes proliferation (p < 0.05) significantly. There is positive correlation between the spleen weight and B-lymphocytes proliferation by 73.3%. These result reveal that okra polysaccharide could improve the immune response and be utilized as a novel candidate of neutraceutical.

Abstract

Background: Keputih Village is located in eastern Surabaya. The boundary of this village is the river. Areas on the river banks are overgrown with wild plants and poorly maintained. One way to overcome the dysfunctional land around the river is by cultivating medicinal plants with verticulture techniques. Purpose: The implementation of this program is to overcome the dysfunctional land around the Keputih Village’s river by cultivating medicinal plants in verticulture techniques and provide training in the manufacture of herbal products from the cultivation of medicinal plants and provide financial management training to the local residents. Methods: This activity was carried out in 4 stages. Phase I was training in making pots and verticulture shelves as well as planting medicinal plants, Phase II was mentoring in the field, Phase III was training on making herbal products from plants and training on simple bookkeeping, Phase IV was product display. Results: Based on the results of this training showed that the knowledge of the participants related to the training material has increased which shown from the results of the posttest with average of 79,31 and product display. Conclusion: In this activity, beside gaining knowledge, participants can apply this knowledge to increase family income.

Abstract

The effect of cutting and adding gibberellin on the duration of stem and flower bud forming of Rosa sp had been done. This research purpose to shorthen the duration of stem and flower bud forming. The research sample were Rose plant with yellow colour and the same length and number of branches. The cutting was done on 0 cm, 5 cm, and 10 cm from the apex. In this research, we used gibberellins with 50 ppm and 100 ppm concentrations and as a control we used aquadest. The result showed that the best treatment to shorthen the duration of stem bud forming were the cutting 10 cm from the apex and with adding of gibberellin on 100 ppm concentration. And the best treatment to shorthen the duration of flower bud forming were with the cutting on 10 cm from the apex and with adding of gibberellins on 50 ppm concentration.

Abstract

This study was aimed to know the effects of oyster mushroom supplement to milk secretion and mammary alveolus diameter of mice. The research was carried out under laboratory condition at the laboratory of Reproduction Biology, Faculty of Mathematic and Sciences, Airlangga University by using Complete Random Method. The study was designed by using totally 32 mice post partum (PP) and devided into four groups of treatment which consisted 8 female mice post partum of each group. The groups are: (a) the control groups, the groups that were given 0.2 ml aquadest, (b) the treatments of oyster mushroom solution with 3 variation of mushroom concentration, there are 2%, 4% and 6%. The oyster mushroom solution was given by gavage. The treatments were started on 3th to 12th day of lactation period. The increasing of milk secretion were showed by the increasing of body weight of offsprings. The data were collected from the difference result of the balance of weight body of offspring and the data were observed on 4th, 6th, 8th, 10th, and 12th of lactation periode and then on 13th day, the mice (five mice for each group) were killed to make histological preparat of mammary glands. The datas were analyzed by ANOVA and LSD test (a = 0.05). The results of this research showed that oyster mushroom supplement can increase milk secretion and mammary alveolus diameter of mice. Oyster mushroom with 6% concentration gave the best effects And there was a positive corelation between the weight of oyster mushroom that was used with the increasing of the oyster mushroom effects.

Abstract

The purpose of this research was conducted to evaluate the effects of “daun wungu” extracts to decrease total cholesterol, LDL (Low Density Lipoprotein) and HDL (High Density Lipoprotein) cholesterol of ovariectomized mice. This research used 24 ovariectomized mice and 8 normal mice. The ovariectomized mice were grouped in 3 groups for K1 (the treatment with aquadest, 0.05 ml), K2 (the treatment with fish oil, 0.05 ml) and P (the treatment with “daun wungu” extracts, 0.5 mg in 0.05 ml fish oil). All treatments were done for 20 days. At the end of the treatments, the blood (0.5 ml) was taken from cardiac by tuberculin disposable syringe. And then with fotometry, serum total, LDL and HDL cholesterol were measured. The result of this research showed that daun wungu extracts (0.5 mg in 0.05 ml fish oil) could not decrease HDL cholesterol, but it could decrease total and LDL cholesterol of blood serum in ovariectomized mice.

Abstract

Extract of testis contains the testis specific proteins. The testis proteins may raise immune responses. The immune responses are the antibodies against testis specific anti-protein. The reaction between the antibodies and the sperms can inhibit fertilization. This process has contraception effects. This research used 24 female and 20 male mice strain Balb/c. The experiment used the complete random design with 4 concentration treatments (0, 500, 1000, and 2000 μg extracts of testis) and 3 replications. Mice were injected 3 times with time interval 21 days. After 15 days from the rising of vaginal plug, 3 mice were dissected and counted the implantation. The others mice were waited until delivered of the children. The datas were analyzed by ANOVA and LSD (α = 5%). The results showed that extract of testis did not influence the amount of implantation at the uterus endometrial, but it decreased the amount of children. The effective concentration of extract testis is 2000 μg. So, this study showed that the intraperitoneum injection of the extract of testis had contraception effects.

Abstract

Estrogenic effects of handeuleum leaf {Graptophyllum pictum (L.)Griff)} were showed by uterus histology examinations of ovariectomy female mice. Female mice aged 3 months, were treated by gavage with handeuleum leaf extracts, ethinyl estradiol, aqua dest, and olive oil. The results showed that the treatment of handeuleum leaf extracts (with concentration 0.5 mg/0.05 ml olive oil) could increase uterus diameter, mucous layer thickness, lumen epithelial cell height, and glandular epithelial cell height if it was compared with aquadest treatments. Compared with ethinyl estradiol as standard estrogen, handeuleum leaf extracts have lower effects than ethinyl estradiol.

Abstract

Kelurahan Pacar Kembang terletak di wilayah Surabaya Timur. Permasalahan yang dihadapi oleh mitra adalah terdapat kampung yang kumuh dan belum tertata, juga pemuda karang taruna dan ibu-ibu PKK yang belum mempunyai kegiatan edukasi untuk dapat mewujudkan kampung yang bersih dan asri. Kegiatan ini bertujuan untuk mewujudkan kampung yang bersih, asri dan aman. Berdasarkan permasalahan tersebut di atas, maka permasalahan mitra yang harus segera ditangani adalah aspek lingkungan dan budidaya dengan memberikan pelatihan mural dan budidaya lidah buaya dilanjutkan dengan kegiatan praktek terhadap materi yang diberikan. Berdasarkan hasil evaluasi terhadap materi yang telah disampaikan menunjukkan bahwa rerata nilai pelatihan mural adalah 76 dan rerata nilai pelatihan budidaya lidah buaya adalah 82.
