Abstract

Penelitian ini dilakukan untuk melihat hubungan antara motivasi memperoleh penghasilan tambahan di luar jam dinas dengan loyalitas prajurit TNI. Adapun hipotesis yang diajukan ialah ada hubungan negatif antara motivasi memperoleh penghasilan tambahan di luar jam dinas dengan loyalitas anggota TNI. Subyek dalam penelitian ini sebanyak 64 prajurit TNI yang masih aktif dan tinggal di Surabaya dan sekitarnya. Pengumpulan data dilakukan menggunakan skala motivasi memperoleh penghasilan tambahan dan skala loyalitas prajurit yang selanjutnya dianalisis dengan teknik korelasi product moment dengan program Statistic Package for Sosial Science for Windows (SPSS) 20.0 untuk membuktikan hipotesis. Hasil analisis uji hipotesis diperoleh koefisien korelasi Pearson (rxy) =  -0,452 pada taraf signifikansi (p) = 0,000 (p<0.01). Hal tersebut membuktikan bahwa ada hubungan negatif, antara motivasi memperoleh penghasilan tambahan di luar jam dinas dengan loyalitas anggota TNI. Hasil kuadrat pada nilai koefisien korelasi diperoleh nilai koefisien determinasi sebesar 0,204 yang berarti bahwa motivasi memperoleh penghasilan tambahan pada prajurit TNI memberikan sumbangan efektif terhadap loyalitas prajurit TNI sebesar 20,7%.Kata kunci: motivasi, loyalitas, penghasilan tambahan

Abstract

Exposure to cadmium (Cd) could increase of reactive oxygen species (ROS) and changes in expression of antioxidant genes. Gynura procumbens is a medicinal plant that is rich in phenolic and flavonoid compounds. The aimed of study to evaluate the hepatoprotective effect of G. procumbens adventitious root (GAR) extract against Cd toxicity, especially expression rate of hepatic antioxidant genes. Twenty-five male mice were treated as follows: P1 (control), P2 (Cd100mg/L), P3 (GAR100mg/L + Cd100mg/L), P4 (GAR200mg/L + Cd100mg/L), and P5 (GAR300mg/L + Cd100mg/L). The samples (blood and liver) were collected for analysis of malondialdehyde (MDA) levels, superoxide dismutase (SOD) and catalase (CAT) activities, and their relative gene expression were determined. The hematological assay showed Cd-treated administered with GAR extract increased the number of red blood cells (RBC), haemoglobin concentration (HGB), hematocrit (HCT), mean corpuscular volume (MCV), and mean corpuscular haemoglobin (MCH), but reduced the level of alanine aminotransferase (ALT) and aspartate aminotransferase (AST). In addition, the GAR extract decreased the MDA production, but increased the activities of SOD and CAT. These enzymatic activities were positively correlated with their respective gene transcripts. Our study revealed that GAR extract administration showed marked hepatoprotective effects against Cd-induced oxidative stress.

Abstract

Common carp (Cyprinus carpio Linnaeus 1758) is grouped into Family Cyprinidae and Order Cypriniformes. One of Indonesian common carp which is commonly domesticated in Indonesia to fulfil consumer needs of animal protein is “Majalaya”. Even though the common carp “Majalaya” is extensively domesticated to meet market demands, any comprehensive study on the fish chromosomes has not been investigated. Chromosome study on the common carp “Majalaya” is quite important related to breeding program including certification process. The objective of this study was to study karyotype of the common carp “Majalaya” collected from UKBAT Cangkringan, Yogyakarta. Chromosome preparation of the common carp “Majalaya” investigated in this study was a splash method with blood cell cultures. The results revealed that the common carp “Majalaya” has diploid chromosome number (2n) = 98 classified as 12 (6 pairs) metacentric chromosomes and 86 (43 pairs) telocentric chromosomes displaying karyotype formula 2n = 2x = 98 = 12 m + 86 t. However, sex chromosomes of the common carp “Majalaya” could not be identified in this study. In addition, the variation of total length chromosomes of the fish is between 1,052 μm and 3,299 μm. Abstract

The valuable extract of bioactive compounds from Gynura procumbens has been widely manufactured into various health products. The demand for these compounds is continuously increasing, but production through conventional farming methods is insufficient due to limited agricultural land and environmental stresses. An alternative to producing plant biomass is in vitro cultivation methods. This method requires less space and enables biomass propagation in a controlled condition that can facilitate stable and efficient production of plant secondary metabolites. This study evaluated the effect of inoculum subculture periods and culture methods on G. procumbens biomass and secondary metabolite production in a bioreactor. The 3-L airlift balloon type-bubble bioreactors was modified in this study to adopt the treatment of 1st-5th subculture periods and fed- and batch-cultivation strategies. We found the G. procumbens adventitious root culture was optimally derived from the 1st subculture produced biomass of 148.02±1.45 g FW and 8.59±0.12 g DW, and TPC (14.48±1.08 mg GAE/g DW) and TFC (116.89±0.44 mg KE/g DW and 33.97±0.13 mg QE/g DW). Additionally, the fed method after 28 days of culture using double distilled water replenishment improved adventitious root biomass (213.75±35.00 g FW and 11.21±0.18 g DW), while nutrient replenishment improved TFC (52.14±0.44 mg KE/g DW and 14.54±0.13 mg QE/g DW). These results can be used to optimize the cultivation of G. procumbens adventitious roots in a large-scale bioreactor.

Abstract

Pandemi COVID-19, mengharuskan masyarakat untuk membiasakan hidup sehat. Selain dengan memakai masker, masyarakat juga dianjurkan untuk menggunakan hand sanitizer dan mengkonsumsi minuman suplemen secara rutin untuk menjaga dan meningkatkan imunitas tubuh. Saat ini telah beredar berbagai jenis hand sanitizer dan minuman suplemen di pasaran, namun dengan harga yang tidak murah dan kadang langka. Hal ini menyebabkan masyarakat enggan untuk membeli serta menggunakan hand sanitizer dan minuman suplemen. Kegiatan pengabdian masyarakat ini bertujuan untuk melatih mitra supaya dapat membuat hand sanitizer, minuman herbal dan budidaya tanaman obat. Metode yang digunakan terdiri atas empat tahap yaitu pretest, materi tentang pembuatan hand sanitizer, minuman herbal, budidaya jahe dan sereh serta manajemen keuangan. evaluasi berupa postest dan tahap terakhir adalah praktek pembuatan hand sanitizer dari bahan herbal, membuat minuman herbal serta budidaya jahe dan sereh. Hasil dari kegiatan ini menunjukkan bahwa terdapat peningkatan pengetahuan terhadap materi yang telah diberikan, hal ini dapat dilihat dari hasil post test yang menunjukkan peningkatan nilai menjadi 91,11, dari nilai awal pretest 75,33. Ada kenaikan nilai sebesar 20,94%. Kegiatan ini diharapkan dapat membantu pencegahan penyebaran COVID-19.Kata kunci—COVID-19, hand sanitizer, herbal

Abstract

Annonaceae is one of the most important and economically valuable plant families in Malesia region. Taxonomical studies could contribute to further utilization and conservation of the studied taxa. Due to their importance, an update in the morphological characters and taxanomical position of Annonaceae is required. This study aims to characterize the taxonomical position and relationships of 24 species belongs to the family Annonaceae found in the Purwodadi Botanic Gardens. In this work, we reported the taxonomical position and the phenetic clustering of 24 Annonaceae species collected from the Purwodadi Botanic Garden, East Java, Indonesia. The taxonomical study was performed based on morphological and phenetic approach where 81 characters (leaves, stem, and flower) were used in the analysis. A dendogram was produced following the scoring of the character using statistical program PAST ver. 3.0. (Hierarchial Cluster Analysis using Jukes Cantor Similarity approach). The  dendogram showed clear separation between members of sub-family Annonoideae and Malmeoideae; however Alphonsea javanica and Sageraea lanceolata were not grouped into their respective tribe or sub-family. Further studies are required to evaluate these taxonomical separations. Overall, from the 24 studied species, we found that they grouped into 2 sub-family (Annonoideae and Malmeoideae), 3 tribes (Annoneae, Miliuseae, and Uvarieae), and 13 genus (Alphonsea, Annona, Desmos, Fissistigma, Miliusa, Mitrephora, Orophea, Polyalthia, Popowia, Pseuduvaria, Sageraea, Stelechocarpus, dan Uvaria).

Abstract

Secondary metabolites in the medicinal plant Gynura procumbens have various beneficial pharmacological activities. However, the limited availability of these beneficial compounds has constrained the possibilities for therapeutic applications. Exogenously applied methyl jasmonate (MeJa) as an elicitor has been reported to trigger the accumulation of secondary metabolites in plant tissues cultured in vitro. This study aimed to assess how varying periods of MeJa elicitation affect the growth and accumulation of phenolic and flavonoid compounds in adventitious root cultures of G. procumbens within a balloon-type bubble bioreactor (BTBB). Adventitious roots were induced from leaf and stem explants obtained from established in vitro shoots. Upon transfer to the BTBB, the cultures were supplemented with 100 µM MeJa and sampled at 1, 2, 3, and 4 weeks. The results revealed that including MeJa in the culture medium significantly boosted growth and the accumulation of phenolic and flavonoid compounds in G. procumbens. The highest biomass and concentrations of phenolic and flavonoid compounds were observed after 3 weeks of elicitation. These findings highlight the potential of MeJa elicitation for enhancing the production of valuable bioactive compounds in G. procumbens adventitious root cultures, showcasing its utility for industrial applications.

Abstract

The endemic Diospyros celebica and widely spread D. blancoi are two valuable species of Diospyros (the genus of persimmons and ebonies) that have been conserved at botanic gardens in Indonesia. Understanding the biological contrasts between endemic and widely spread species is essential for plant conservation. This study aims to compare the phenotypic and estimated genetic variability between the endemic D. celebica and the widely spread D. blancoi to support their conservation. Fifty-three morphological characters from fifteen selected living plant specimens at Purwodadi Botanic Garden were measured directly or using variable encoding methods. Unpaired two-samples Wilcoxon test and hierarchical cluster analysis were used to compare morphological variations between both species. Further, genetic variabilities were estimated from encoded morphological characters using GenAlEx 6.503 software. The two Diospyros’ morphological characters were significantly different, except for the lamina length, pairs of leaf vein, number of flowers in each inflorescence, and the fruit sulci presence. The cluster analysis successfully distinguished D. celebica from D. blancoi according to thirteen comparable morphological characters. The endemic D. celebica has lower genetic variability (female/bisexual %P = 86.21%, He = 0.259, uHe = 0.277; male P = 54.29%, He = 0.195, uHe = 0.234) than the widely spread D. blancoi (female/bisexual %P = 86.76, He = 0.311, uHe = 0.355). Due to the importance of genetic diversity in plant conservation, both species’ population size needs to be maintained, if not increased, to preserve the existing phenotypic and genetic variabilities within the cultivated populations at the botanic garden.

Abstract

Komunitas peternak sapi perah di Junrejo, Batu menghadapi tantangan dalam meningkatkan produktivitas dan kualitas susu akibat rendahnya kesehatan ternak dan keterbatasan akses terhadap teknologi pakan yang efisien. Program pengabdian masyarakat ini bertujuan untuk meningkatkan pemahaman dan keterampilan peternak dalam memanfaatkan probiotik dan Single Cell Protein (SCP) sebagai pakan tambahan, yang diharapkan dapat meningkatkan kualitas kesehatan dan produktivitas ternak. Program ini menggunakan pendekatan partisipatif yang melibatkan pelatihan langsung dan sesi diskusi untuk memastikan penerapan yang efektif dan berkelanjutan. Probiotik dan SCP berperan dalam memperbaiki mikrobiota usus ternak, meningkatkan efisiensi pencernaan, dan memberikan sumber protein berkualitas tinggi. Hasil pelatihan menunjukkan bahwa setelah mengikuti kegiatan ini, seluruh peserta (100%) memahami dasar-dasar probiotik dan SCP, serta mampu menerapkannya secara mandiri di peternakan mereka. Di samping itu, penggunaan molase sebagai media perbanyakan probiotik mendukung prinsip ekonomi sirkuler, mengurangi limbah produksi gula, dan menciptakan solusi pakan yang ramah lingkungan. Program ini berhasil meningkatkan kapasitas komunitas peternak lokal, sehingga dapat berkontribusi terhadap kesejahteraan ekonomi mereka melalui peningkatan hasil susu dan biaya operasional yang lebih efisien.

Abstract

The environmental microbiome plays an important role in various ecosystems around the world, influencing nutrient cycling, disease dynamics and ecosystem stability. This bibliometric analysis provides a comprehensive overview of the current state and future directions of environmental microbiome research from a global perspective from 2009–2024. Through systematic examination of Scopus publications, 2154 documents were found. The results show a significant increase in the number of publications since 2017 and a peak in 2024. The most cited document was "The hidden world within plants: Ecological and evolutionary considerations for defining functioning of microbial endophytes" with 1887 citations and “Structure and function of the global ocean microbiome” with 1843 citations. The most productive countries in environmental microbiome research are the United States (n = 748), China (n = 533), India (n = 308), Germany (n = 172) and the United Kingdom (n = 157). Microbiome, microbial community, microfolora, microbiota, microbiology, bacteria, and bacterium are the most popular topics that will continue to develop in the future. Although the United States has published the highest total number of papers, more recent studies have predominantly been published from China, indicating shift in the centre of study from the United States to China. Insights gained from this analysis contribute to a deeper understanding of the environmental microbiome research landscape, guiding future research priorities and collaborations in the field.
