Abstract

ABSTRAK Uji fitokimia dan uji aktivitas antibakteri ekstrak metanol batang Melicope glabra (BI.) T.G. Hartley telah dilakukan. Ektraksi sampel batang Melicope glabra dilakukan dengan metode maserasi menggunakan pelarut metanol. Uji aktivitas antibakteri dilakukan menggunakan metode difusi agar. Hasil uji fitokimia menunjukkan ekstrak metanol batang Melicope glabra mengandung senyawa metabolit sekunder flavonoid, fenolik, triterpenoid dan steroid. Nilai MIC (Minimum Inhibitory Concentration) ekstrak metanol batang Melicope glabra (BI.) T.G. Hartley untuk bakteri Salmonella typhi adalah pada konsentrasi 1,25%, sedangkan untuk bakteri Staphylococcus aureus adalah pada konsentrasi 2,5%. Ekstrak metanol batang Melicope glabra (BI.) T.G. Hartley memiliki aktivitas antibakteri dengan spektrum luas. Kata Kunci : Melicope glabra (BI.) T.G. Hartley, aktivitas antibakteri, metabolit sekunder

Abstract

Lipase is an enzyme that can hydrolyze triglycerol into glycerol and free fatty acids. The aim of this research is to obtain lipase-producing bacteria and determine the potential of lipase from water bacteria in the water of the Benanga Lempake Dam, North Samarinda for biodetergent applications through stability test of lipase activity against several commercial detergents. Screening of lipase-producing bacteria was carried out on nutrient agar media containing olive oil and Rhodamine B. The results showed that seven isolates were capable of producing lipase. Based on the stability test of lipase on detergent, it can be seen that the activity of lipase decreased by more than 70% at the addition of 1% detergent.

Abstract

Phytochemical test and antibacterial activity of ethanol extract of Mahoni seed (Swietenia mahagoni (L.) Jacq) have been done. The extraction of Mahoni seed samples was carried out by the maceration method using ethanol as a solvent. An antibacterial activity test was carried out using the agar diffusion method. Phytochemical tests results show that ethanol extract of Mahoni seeds has secondary metabolites of alkaloid, triterpenoid, and phenolic compounds. The antibacterial activity test results obtained MIC values for Streptococcus mutans bacteria were 1.25-2.5% and for Salmonella typhi bacteria at 0.625-1.25%. Ethanol extract of Mahoni seeds has broad-spectrum antibacterial activity.

Abstract

The research about in vitro vasodilatation response of methanol extract of Macaranga lamellata whitmore leaves was carried out. This research was conducted to determine the vasodilatation response of methanol extract of Macaranga lamellata Whitmore leaves on changes in aortic tone of Wistar rat. The methods used include the in vitro methods by using separated aortic organ of Wistar rat. Methanol extract were given cumulatively in bath organs. Positive control is used methacholine and negative control is used DMSO solvent. T-test statistical was done to determine the significant differences between groups. Phytochemical test results showed that total methanol extract contained alkaloids, polyphenol, steroids, flavonoids and saponins. Percentage change in aortic tone by using methanol extract concentration of 0.3; 1 and 3 mg/mL respectively were -7.422%; -9.641% and -10.593%. Methanol extract of Macaranga lamellata Whitmore leaves have antihypertensive potential. Keywords: Antihypertensive, Aortic, In vitro, Macaranga lamellata Whitmore, Vasodilatation

Abstract

Raw starch-degrading amylase (RSDA) is a hydrolytic enzyme that can degrade raw starch into glucose. This study was conducted to obtain raw starch-degrading amylase-producing bacteria from the soil of the Bukit Pinang Landfill, Samarinda City. Isolation of bacteria using the spread plate method. Screening of RSDA-producing bacteria using nutrient media to contain raw starch. Isolation of bacteria obtained 10 single colonies of bacteria. The isolated bacteria were selected for their ability to produce RSDA. All single colonies of bacteria could produce RSDA and their activity was tested quantitatively using the 3,5 Dinitrosalicylic Acid (DNS) method. Bacteria with the TPA 1 colony code had the highest activity of 3.837 U/mL.

Abstract

Bacteria are one of the microorganisms that have the potential to produce hydrolytic enzymes. This research was carried out to isolate bacteria from the soil of the Bukit Pinang Samarinda Final Disposal Site and select bacteria that produce cellulase, protease and lipase enzymes. Bacteria from landfill soil were isolated using the spread plate method and 10 single colony bacterial isolates were selected which were then coded TPA 1 – TPA 10. The results of the selection of bacteria producing hydrolase enzymes showed that 9 isolates were capable of producing protease enzymes, 6 isolates were capable of producing cellulase enzymes and 4 isolates were capable of producing cellulase enzymes. The isolate was able to produce the lipase enzyme.

Abstract

Pacing plants (Costus sp.) Potentially as medicinal plants, but there is no research on the endophytic bacteria they contain. This research was conducted to filter endophytic bacteria from Pacing stems (Costus sp.) And to find out the secondary metabolites contained by each endophytic bacteria. Endophytic bacteria obtained from Pacing Stems (Costus sp.) Totaled 17 colonies. Crude extracts from all bacterial colonies containing alkaloids, 15 crude extracts containing saponins. Steroids are only contained by 3 types of bacterial extracts and triterpenoids are only contained by 2 types of bacterial extract.

Abstract

Uji Fitokimia dan aktivitas antibakteri ekstrak metanol daun salam (Syzygium polyanthum) telah dilakukan. Penelitian ini bertujuan untuk mengetahui metabolit sekunder yang terkandung pada ekstrak metanol daun salam dan konsentrasi hambat minimum (KHM) ekstrak tersebut. Uji aktivitas antibakteri dilakukan dengan metode difusi menggunakan kertas cakram. Ekstrak metanol daun salam (Syzygium polyanthum) metanol daun salam mengandung steroid, flavonoid dan fenolik. Konsentrasi hambat minimum ekstrak metanol daun salam (Syzygium polyanthum) terhadap Staphylococcus aureus sebesar 0-1% dan terhadap Salmonella typhi sebasar 1-2%.

Abstract

Isolasi bakteri penghasil lipase dari sumber air panas Long Kali dilakukan dengan menyeleksi koloni bakteri pada media Nutrient Agar (NA) yang mengandung Rhodamin B dan minyak zaitun. Hasil isolasi menunjukkan 5 dari 14 isolat bakteri mampu menghasilkan lipase. lima isolat tersebut diuji aktivitas lipase dengan metode titrimetri, isolat dengan kode 14 memiliki aktivitas tertinggi dengan aktivitas 2,20 Unit/mL.

Abstract

Endophyte bacteria isolate from the leaves of Macaranga hullettii King ex Hook.f. has potential to producer lipase. In the study was conducted to filter lipase producing endophyte bacteria from Macaranga hullettii King ex Hook.f. and determine the optimum production time. The results of screening endophyte bacteria showed 6 isolates of endophyte bacteria that had the potential to produce lipase and 1 isolate was chosen which is number 5 to determine the optimum production time. The optimum production time of lipase from Macaranga hullettii King ex Hook.f. leaves for 72 hours with activity that is 2.92 U/mL.
