Abstract

Isolate J7 is a bacterial strain separated from the rhizosphere of Zingiber officinale Roscoe var. Rubrum that potentially produces antibacterial compounds against Escherichia coli. The study aimed to determine the antibacterial activity of the most active fraction of this isolate from its capacity to induce leakage in E. coli cells. The secondary metabolites were extracted from Isolate J7 using ethyl acetate solvent and then fractionated with different ratios of hexane and ethyl acetate solvents―1:0, 9:1, 8:2, 7:3, 6:4, 5:5, 4:6, 3:7, 2:8, 1:9, and 0:1 (v/v), ethyl acetate and methanol with 1:1 (v/v) ratio, and methanol 100%. Fractions were identified based on the spotting on the Thin-Layer Chromatography (TLC) plate. Only the most active fraction was tested to define its ability to cause leakage of cellular components like nucleic acid and protein. The leakage was scanned with a UV-Vis spectrophotometer at wavelengths of 260 and 280 nm. The results showed that F2 was the fraction that prevented the growth of E. coli most actively because it created a zone of inhibition sized 11.58±0.95 mm in diameter with the lowest MIC among the other fractions (10%). Also, based on the spectrophotometric analysis, the addition of F2 at concentrations of 10% and 20% resulted in higher levels of protein, DNA, and RNA than the negative control. In conclusion, F2 can cause plasma membrane leakage in E. coli at a concentration of 10%. Another fraction that exhibited antibacterial activity was F3. In the spotting analysis of the TLC plate, F3 appeared to have a spot profile and Rf that were similar to F2 but considerably different from the inactive fractions (i.e., showing no antibacterial activity). Observed with multiple wavelengths, the Rf values of F2 and F3 spots varied between 0.56-0.57 and 0.61-0.62 (254 nm) and 0.47-0.48 and 0.56-0.57 (366 nm), respectively. Because these spot profiles did not appear in the inactive fractions, compounds with this range of Rf values are, thereby, suspected as the active substances that inhibit the growth of E. coli.

Abstract

Bangsa Indonesia secara turun temurun menggunakan tanaman yang dimanfaatkan sebagai obat tradisional untuk menanggulangi masalah kesehatan. Pada umumnya penggunaan obat tradisional dinilai lebih aman daripada penggunaan obat modern. Hal ini disebabkan karena obat tradisional memiliki efek samping yang relatif lebih sedikit daripada obat modern. Akan tetapi tetap diperlukan ketepatan penggunaan obat tradisional untuk meminimalisir efek sampingnya. Oleh karena itu, dilakukan edukasi tentang keamanan penggunaan obat herbal kepada masyarakat agar masyarakat dapat terhindar dari efek yang tidak diinginkan. Kegiatan pengabdian dilakukan dengan sosialisasi atau pemberian materi secara tatap muka langsung dengan masyarakat. Untuk materi mengenai edukasi keamanan obat herbal diberikan oleh dosen pengusul dan teknisnya dibantu oleh mahasiswa. Partisipasi mitra dalam pengabdian ini dapat dilihat dengan ikut terlibatnya mitra dalam sesi diskusi maupun tanya jawab yang diadakan. Kegiatan pengabdian secara keseluruhan dikatakan baik dan berhasil, dilihat dari keberhasilan target jumlah peserta pelatihan, ketercapaian tujuan pelatihan, ketercapaian target materi yang telah direncanakan, dan kemampuan peserta dalam penguasaan materi. Kegiatan pengabdian ini merupakan implementasi dari mata kuliah pengembangan obat tradisional. Selain itu target luaran dari kegiatan pengabdian ini adalah berupa artikel dalam bentuk publikasi jurnal terkait pengabdian yang dilaksanakan

Abstract

ABSTRACT. Indonesian have used plants as traditional medicine to treat health problems. In general, the use of traditional medicine is considered safer than the use of modern medicine. This is because traditional medicine has relatively fewer side effects than modern medicine. However, it is still necessary to use traditional medicines accurately to minimize side effects. Empowering housewives in using Family Medicinal Plants can be used to support improving family welfare and health. For this reason, this service activity aims to: (1) increase knowledge about the scientific properties of Family Medicinal Plants among housewives, (2) increase knowledge about the procedures for planting Family Medicinal Plants among housewives, and (3) improve skills for processing Family Medicinal Plants among housewives. stairs so that it can be beneficial for health. Service activities are carried out using the lecture method to explain the scientific benefits of Family Medicinal Plants, planting Family Medicinal Plants, and processing Family Medicinal Plants. Overall service activities can be said to be good and successful, seen from the success of the target number of training participants, achievement of training objectives, achievement of planned material targets, and participant's ability to master the material to meet predetermined targets. The Empowerment of Housewives in the Use of Family Medicinal Plants community service program in Pangkah Hamlet, Bantul, Yogyakarta was able to increase participants' interest and knowledge regarding the use of family medicinal plants. Abstract

Background: Kaempferia parviflora (Black ginger) has phytochemical compounds such as flavonoids, terpenoids, essential oils, and phenols that have the potential to treat various diseases. To maximize the total extracted flavonoids needs to optimize the extraction duration.Objective: Knowing the optimum maceration duration on the total flavonoid content using the UV-VIS Spectrophotometric method on black ginger ethanol extract.Method: Black ginger is extracted using the maceration method with ethanol (70%) solvent in maceration duration of 12, 24, 36,48, and 60 hours. The extract will be tested qualitatively and quantitative to determine the total levels of flavonoids using the UV-Vis Spectrophotometric method.Result: Qualitative test of black ginger ethanol extract gave positive results for flavonoids, phenolics, alkaloids, steroids, and tannins. The total level of flavonoids in black ginger ethanol extract was 7,8988  0,1223 mgEQ/g (12 hours); 8,5705  0,1529 mgEQ/g (24 hours); 9,8102  0,0416 mgEQ/g (36 hours); 10,6161  0,0768 mgEQ/g (48 hours); 7,2594  0,1244 mgEQ/g (60 hours). The statistical analysis indicates a significant difference (p < 0,05) in the total flavonoid content at each maceration time. Conclusion: The duration of maceration time significantly affects the total flavonoid content produced with the optimum maceration time results at 48 hours

Abstract

Bacteria are one of the organisms that cause infectious diseases. One of the bacteria that can cause infection is Staphylococcus aureus. Treatment of bacterial infections is by using antibiotics, but using the antibiotics for a long term can cause resistance. It is necessary to look for alternative compounds that can inhibit the growth of Staphylococcus aureus. In the bioprospecting process of medicinal plants, it is not only necessary to rely on empirical information but also to have scientific evidence proven through scientific testing. One of the plants with potential as a medicine is black ginger (Kaempferia parviflora). The aim of this study is to determine the effectiveness of Kaempferia parviflora rhizome in inhibiting the growth of Staphylococcus aureus bacteria. The concentrations of the Kaempferia parviflora extract tested were 25%, 50%, 75%, and 100%. Based on the results, it can be concluded that the ethanol extract of Kaempferia parviflora rhizome has antibacterial activity against the growth of Staphylococcus aureus ATCC 25923 at concentrations of 25%, 50%, 75%, and 100%, with strong antibacterial inhibition. This study is expected to contribute to the field of health in general and pharmacy in particular by developing the potential of Indonesian medicinal plants. The findings of this study will be directed toward the development of formulations with antibacterial activity.

Akan tetapi tetap diperlukan ketepatan penggunaan obat tradisional untuk meminimalisir efek sampingnya. Pemberdayaan masyarakat dalam pemanfaatan dan pengolahan tanaman jahe untuk kesehatan salah satunya dapat digunakan untuk mendukung peningkatan kesejahteraan dan kesehatan keluarga. Untuk itu kegiatan pengabdian ini bertujuan untuk: meningkatkan pengetahuan tentang khasiat tanaman jahe secara ilmiah, meningkatkan pengetahuan tentang tata cara menanam tanaman jahe, dan meningkatkan keterampilan untuk mengolah tanaman jahe menjadi minuman yang bermanfaat bagi kesehatan. Kegiatan pengabdian dilakukan dengan metode ceramah dan demonstrasi. Metode ceramah untuk menjelaskan tentang khasiat tanaman jahe dan mempraktekkan pengolahan jahe menjadi bahan minuman yang berkhasiat bagi kesehatan. Kegiatan pengabdian secara keseluruhan berjalan dengan baik dan berhasil, dilihat dari keberhasilan target jumlah peserta pelatihan, ketercapaian tujuan pelatihan, ketercapaian target materi yang telah direncanakan, dan kemampuan peserta dalam penguasaan materi memenuhi target yang telah ditetapkan. Program pengabdian masyarakat tentang Edukasi untuk Peningkatan Pengetahuan tentang Tanaman Obat Keluarga (TOGA) terhadap Pemanfaatan tanaman Jahe.

Abstract

Background: Bacteria are one of the organisms that cause infectious diseases. One of the bacteria that can cause infection is Staphylococcus aureus. Treatment of bacterial infections is by using antibiotics, but using the antibiotics for a long term can cause resistance. It is necessary looking for alternative compounds that can inhibit the growth of Staphylococcus aureus. Cymbopogon nardus L. Rendle is a plant that has antibacterial compound, that compounds are flavonoids and tannins.Objective: To determine the antibacterial activity of Cymbopogon nardus ethanol extract against the growth of Staphylococcus aureus ATCC 25923 using the well method.Method: Cymbopogon nardus is extracted using the maceration method with 70% ethanol solvent. A phytochemical screening test was carried out and continued with an antibacterial activity test using the well method with concentrations of 5%, 10%, 15% and 20%.Results: Phytochemical screening show that Cymbopogon nardus ethanol extract contains saponins, flavonoids, steroids and tannins. The results of the antibacterial test showed that the average inhibitory zone value of Cymbopogon nardus ethanol extract at concentrations of 10%, 15% and 20% was 23.00; 23.77; 24.96 mm, whereas at concentration of 5% it did not show an inhibition zone for the Staphylococcus aureus ATCC 259323 bacteria.Conclusion: Cymbopogon nardus ethanol extract has antibacterial activity against the growth of Staphylococcus aureus ATCC 259323 with a minimum inhibitory concentration (MIC) which is able to inhibit the growth of Staphylococcus aureus ATCC 259323 at concentration of 10%, for concentrations of 10%, 15% and 20% it has an inhibitory power of very strong category.
