Abstract

Inulin from Gembili has been identified as an effective bioreductor for forming nanosilver with a size of 481.4 nm, stable for 30 days when stored at 4°C. Inulin nanosilver exhibits immunomodulatory properties and has been proven safe through acute toxicity evaluation at a dose of 4 mg/kgBB. A drug delivery system needs to be developed for its use as a supplement. Syrup was chosen due to its alcohol-free nature, better taste, and ease of measuring the active substance compared to elixirs, solutions, and suspensions. Simplex syrupus, used as a syrup base, influences stability by potentially forming crystals during storage. This research aims to determine how varying concentrations of simplex syrupus affect the physicochemical properties of inulin nanosilver syrup. The study involved the biosynthesis process using Gembili's inulin, nanosilver characterization, formulation, and stability testing. Inulin nanosilver syrup was prepared with simplex syrupus concentrations of 20%, 40%, and 60%. The physicochemical stability of the syrup, including organoleptic properties, pH, and viscosity, was tested before and after storage at 4°C and 40°C over six cycles. The selected formula was evaluated for sugar reduction content and FT-IR profile. Data analysis was performed using SPSS 21.0 for Windows with One-way ANOVA and Paired T-Test. Results indicated that higher concentrations of simplex syrupus led to increased consistency, pH, and viscosity. A 60% concentration of simplex syrupus met the physicochemical stability requirements, with a medium-thick consistency, pH of 5.25±0.03, and viscosity of 92±2.6 cps. The reducing sugar content was 20.59% ±0.002, and the FT-IR profile confirmed the presence of inulin nanosilver, indicated by Ag-N groups compared to silver nitrate solution. This product has the potential to be developed as a health supplement.

Abstract

Potassium azeloyl diglycinate is an ingredient for moisturizing the skin. In this research, potassium-azeloyl-diglycinate was formulated into a gel-based hydrating serum. A good serum gel can be identified by its physicochemical properties and stability, which are determined by the composition of the ingredients used, one of which is CMC-Na, which is used as a gelling agent. The aim of this research was to test the effect of CMC-Na concentrations (1%, 1.25%, and 1.5%) on the physicochemical stability of serum and to find a formulation with the best stability by testing the water content and safety. The evaluation results show that increasing the concentration of CMC-Na has an impact on increasing viscosity, pH, and adhesion, thereby providing a different gel consistency in each formula and reducing its spreadability. The best formulation is Formula-2, which is then tested for water content on respondents. The research results show that this formula has the ability to hydrate the skin, as seen from an increase in oil content of 8.39% and an increase in water content of 8.86%. Initial acute dermal toxicity test results show that the serum is safe to use on the skin and does not cause side effects or toxicity reactions.

Abstract

Indonesia has a tropical climate, and it causes skin diseases such as acne. One of the bacteria causes acne is Staphylococcus aureus. Acne is caused by clogged skin pores so that oil secretion becomes clogged and then enlarges and dries. Antibacterial agents can be applied to the skin with the help of topical preparations, one of which is lotion. Lotion with kaffir lime leaf oil as the main ingredient contains high flavonoid compounds that have an antibacterial activity that can prevent acne, and chicken leg bone gelatin, contains collagen which can moisturize the skin. This research is experimental laboratory research. Lotion is made with different concentrations of kaffir lime leaf oil 0.5%, 1%, and 1.5% w/v. The lotion was tested for physical (organoleptic, homogeneity, spreadability, adhesion, viscosity) and chemical (pH) properties. Antibacterial activity test was carried out by well diffusion method. The research data were processed using SPSS 21 with the Kruskal Wallis Test. The test results showed that the lotion preparation with the strongest antibacterial activity was the preparation with 1.5% kaffir lime leaf oil, with a yield of 11.26 mm. The more composition of kaffir lime leaf oil, the stronger the antibacterial inhibition. The more composition of kaffir lime leaf oil causes the viscosity to decrease, the pH to increase, the adhesive power to decrease, the dispersion power to increase, and the bacterial inhibitory power to increase.

Abstract

Parijoto (Medinilla speciosa, Reinw. ex. Bl.), a tropical plant native to Southeast Asia, contains flavonoids, tannins, and saponins, which have the potential as an anticancer. Gold nanoparticle-based drug formulations are applied to increase the anticancer effectiveness of herbal medicines. The compounds in the stalk of parijoto have the potential to be bioreductor in the biosynthesis of gold nanoparticles. This study aims to determine the physicochemical characterization, cytotoxic activity, and expression of protein caspase-9 after treatment with nanogold parijoto (AuNPs-PR) on HeLa cell. The nanogold biosynthesis process was done by reacting 1 mM HAuCl4 with parijoto aqueous extract (EP). Physicochemical characterization measure of particle size, Polydisperse Index (PdI), and zeta potential of AuNPs-PR was carried out using a particle size analyzer. The cytotoxic effect and viability cell of AuNPs- PR were carried out using the MTT assay. The expression of caspase-9 was observed by immunocytochemistry assay. Physicochemical characterization of AuNPs-PR shows that the particle size value is 160.8 nm with PdI and zeta potential values of 0.430 and -4.56 mV respectively. In the MTT assay, both AuNPs-PR and EP demonstrated a reduction in the viability of Hela cells after 24 h in a dose-dependent manner, yielding IC50 values of 3.28 μg/mL and 19.22 μg/mL, respectively. AuNPs-PR and EP showed low cytotoxic activity against Vero normal cells, with IC50 values of over 500 μM. Further, the immunocytochemistry assay indicated that there was upregulation of caspase-9 by their expression. These results indicate that AuNPs-PR could effectively induce apoptosis in HeLa cells by upregulating caspase-9.Keywords: caspase-9, HeLa cells, MTT, nanogold, parijoto.
