Abstract

Moringa leaf extract contains flavonoids confirmed as anti-inflammatory at 200 mg/Kg b.w. The patch was chosen for topical anti-inflammatory preparation because it is continuous for an extended treatment period. Previous research has developed a patch formula using a polymer combination of polyvinyl alcohol (PVA) and alpha-cellulose (1: 2) that meets the requirements for good patch preparation. However, the effect of temperature resistance on physicochemical properties and anti-inflammatory activity needs to be confirmed. Moringa leaves were macerated in 96% ethanol and dispersed into a patch matrix of PVA and alpha-cellulose. Patches were treated at various storage temperatures, namely 4°C, 25°C, and 40°C each for 8 hours for six cycles. Mice were divided into three groups: blank patch, brand patch product, and moringa leaf patch. Mice were given 0.05 mL of carrageenan suspension intraplanar on the sole of the left foot, and the edema volume was measured using a plethysmometer. Statistical analysis uses One-Way ANOVA and T-test. The results showed that temperature affected the pH, thickness, and weight properties but did not affect the moisture, folding endurance, and organoleptic properties. The average edema volume of mice in the brand product group was not significantly different from the moringa leaf patch group, with a significance value of 0.066 (p > 0.05). The Moringa leaf ethanolic extract patch confirmed it can reduce the volume of mouse foot edema by 5 hours.

Abstract

Inulin from Gembili has been identified as an effective bioreductor for forming nanosilver with a size of 481.4 nm, stable for 30 days when stored at 4°C. Inulin nanosilver exhibits immunomodulatory properties and has been proven safe through acute toxicity evaluation at a dose of 4 mg/kgBB. A drug delivery system needs to be developed for its use as a supplement. Syrup was chosen due to its alcohol-free nature, better taste, and ease of measuring the active substance compared to elixirs, solutions, and suspensions. Simplex syrupus, used as a syrup base, influences stability by potentially forming crystals during storage. This research aims to determine how varying concentrations of simplex syrupus affect the physicochemical properties of inulin nanosilver syrup. The study involved the biosynthesis process using Gembili's inulin, nanosilver characterization, formulation, and stability testing. Inulin nanosilver syrup was prepared with simplex syrupus concentrations of 20%, 40%, and 60%. The physicochemical stability of the syrup, including organoleptic properties, pH, and viscosity, was tested before and after storage at 4°C and 40°C over six cycles. The selected formula was evaluated for sugar reduction content and FT-IR profile. Data analysis was performed using SPSS 21.0 for Windows with One-way ANOVA and Paired T-Test. Results indicated that higher concentrations of simplex syrupus led to increased consistency, pH, and viscosity. A 60% concentration of simplex syrupus met the physicochemical stability requirements, with a medium-thick consistency, pH of 5.25±0.03, and viscosity of 92±2.6 cps. The reducing sugar content was 20.59% ±0.002, and the FT-IR profile confirmed the presence of inulin nanosilver, indicated by Ag-N groups compared to silver nitrate solution. This product has the potential to be developed as a health supplement.

Abstract

Parijoto (Medinilla speciosa, Reinw. ex. Bl.), a tropical plant native to Southeast Asia, contains flavonoids, tannins, and saponins, which have the potential as an anticancer. Gold nanoparticle-based drug formulations are applied to increase the anticancer effectiveness of herbal medicines. The compounds in the stalk of parijoto have the potential to be bioreductor in the biosynthesis of gold nanoparticles. This study aims to determine the physicochemical characterization, cytotoxic activity, and expression of protein caspase-9 after treatment with nanogold parijoto (AuNPs-PR) on HeLa cell. The nanogold biosynthesis process was done by reacting 1 mM HAuCl4 with parijoto aqueous extract (EP). Physicochemical characterization measure of particle size, Polydisperse Index (PdI), and zeta potential of AuNPs-PR was carried out using a particle size analyzer. The cytotoxic effect and viability cell of AuNPs- PR were carried out using the MTT assay. The expression of caspase-9 was observed by immunocytochemistry assay. Physicochemical characterization of AuNPs-PR shows that the particle size value is 160.8 nm with PdI and zeta potential values of 0.430 and -4.56 mV respectively. In the MTT assay, both AuNPs-PR and EP demonstrated a reduction in the viability of Hela cells after 24 h in a dose-dependent manner, yielding IC50 values of 3.28 μg/mL and 19.22 μg/mL, respectively. AuNPs-PR and EP showed low cytotoxic activity against Vero normal cells, with IC50 values of over 500 μM. Further, the immunocytochemistry assay indicated that there was upregulation of caspase-9 by their expression. These results indicate that AuNPs-PR could effectively induce apoptosis in HeLa cells by upregulating caspase-9.Keywords: caspase-9, HeLa cells, MTT, nanogold, parijoto.
