Abstract

β-CGTase (β-siklodekstrin glikosil transferase) adalah enzim yang mengkonversi pati menjadi CD (siklodekstrin). CD adalah oligosakarida yang bernilai tinggi digunakan untuk modifikasi pangan, tekstil, kimia, obat-obatan, kosmetik hingga bioteknologi. Penelitian ini bertujuan untuk mendapatkan isolat bakteri penghasil β-CGTase dari tanah perkebunan ubi jalar. Skrining dilakukan menggunakan media Horikoshi yang mengandung indikator warna fenolftalein dan metil jingga, bakteri penghasil β-CGTase akan memberikan yellow hollow zone jika tumbuh pada media tersebut. Pengukuran aktifitas CGTase diukur menggunakan spektrofotometer UV-Vis. Didapatkan tiga isolat penghasil CGTase diberi kode BSP6A, BSP6B, dan BSP6C, yang memiliki karakteristik Gram positif, berbentuk basil, menghasilkan spora, bersifat motil, uji katalase positif, dan dapat memfermentasikan asam. Ketiga isolat ini mampu menghasilkan enzim CGTase optimum jika diinkubasi selama 18 dan 24 jam pada suhu 37°C dan pH 10, serta mampu mengubah pati menjadi CD jika diinkubasi selama 30 menit pada suhu 37°C dan pH 7.

Abstract

Cyclodextrin glycosyltransferase (CGTase) is an extracellular enzyme produced by several microorganisms, particularly bacteria, that converts starch into cyclodextrin (CD). CD has various applications in fields such as food, textiles, pharmaceuticals, chemistry, and biotechnology. The objective of this study was to isolate b-CGTase-producing bacteria from soil in a potato plantation located in Kerinci, and to conduct their characterization. Screening was conducted using Horikoshi agar media with phenolphthalein and methyl orange color indicators. The results indicated that one isolate, coded CK-2, produced the optimum b-CGTase enzyme when incubated for 30 hours at 37 ºC and pH 10. The CGTase enzyme converted starch to CD when incubated for 60 minutes at 37 ºC and pH 7. The CK-2 isolate was identified as monobacilli Gram-positive bacteria, having spores, positive in the catalase test, and motile. The study concluded that the bacteria producing b-CGTase enzyme are found in the soil of potato plantations in Kerinci, which is believed to originate from the genus Bacillus sp.

Abstract

Stunting adalah masalah gizi akibat kekurangan asupan jangka panjang, terutama dari masa kehamilan hingga anak berusia 24 bulan. Penelitian ini menganalisis implementasi program penanggulangan stunting di Dinas Kesehatan Kabupaten Kerinci, menggunakan pendekatan deskriptif kualitatif dan studi kasus. Sampel penelitian dipilih secara purposive sampling, melibatkan petugas kesehatan dan masyarakat yang terkait langsung dengan program ini. Data diperoleh melalui wawancara mendalam dan dianalisis secara tematik dengan triangulasi untuk memastikan validitas. Hasil menunjukkan bahwa koordinasi antarinstansi dalam penanggulangan stunting berjalan efektif dengan dukungan komitmen yang kuat. Terdapat 724 kasus stunting pada anak usia 0-59 bulan, atau sekitar 4,24%, dengan prevalensi tertinggi di Kecamatan Kuliling Danau mencapai 11,80%. Inovasi dalam program penanggulangan stunting telah diterapkan dan mendapatkan respon positif dari masyarakat, meski partisipasi masih perlu ditingkatkan. Implementasi program telah optimal, namun terdapat beberapa tantangan, seperti rendahnya pengetahuan dan kondisi ekonomi kelompok sasaran, keterbatasan anggaran, serta kurangnya publikasi dan konvergensi lintas sektor.

Abstract

Papaya leaves (Carica papaya L.) are rich in secondary metabolites with pharmacological potential as antioxidants, anti-inflammatories, and antimicrobials. This study aimed to identify bioactive compounds in papaya leaves, develop simplicia and extracts as a basis for capsule formulation, and evaluate the characteristics and stability of the resulting capsules. The methods used included simplicia preparation, extraction using maceration with 70% ethanol, phytochemical analysis using specific reagent tests, and capsule formulation with appropriate excipients. The results showed that papaya leaf simplicia had a moisture content of ≤ 5%, extraction yielded a rendement of 9.6%, and active compounds such as alkaloids, flavonoids, and polyphenols were detected. Capsule formulations containing papaya leaf extract showed good characteristics, with a dissolution time reaching 80% within 30 minutes, and good stability during storage. Papaya leaf extract can be developed as a stable, effective, and pharmaceutically standardized herbal phytopharmaceutical capsule preparation to support natural medicine.
