Abstract

Snakehead fish (Channa striata) is a highly nutritious food product. However, unhygienic smoking processes may lead to contamination by pathogenic microorganisms, including Staphylococcus aureus, which has the potential to cause food poisoning. This study aimed to determine the level of bacterial contamination in smoked snakehead fish (ikan gabus salai) sold in traditional markets, particularly to identify the presence of Staphylococcus aureus and to determine the total bacterial count based on the SNI 7388:2009 standard. This study employed a laboratory experimental method using smoked snakehead fish samples collected from four vendors at Simpang Limun Traditional Market. Bacterial identification was conducted through a series of tests, including growth on the selective medium Mannitol Salt Agar (MSA), Gram staining, and biochemical tests such as motility, catalase, coagulase, and Voges-Proskauer (VP) tests. In addition, the Total Plate Count (TPC) was determined using the pour plate method on Plate Count Agar (PCA) medium. The results showed that yellow-colored colonies on MSA medium indicated the growth of Staphylococcus aureus. Gram staining revealed purple-colored cocci arranged in grape-like clusters, which are characteristic of Gram-positive bacteria. The biochemical tests further supported these findings, showing positive reactions for catalase, coagulase, and VP tests. The TPC values obtained from several samples showed that the number of colonies did not exceed the maximum limit specified by SNI, namely 5 × 10⁵ colonies/g, indicating that although S. aureus was detected in the tested smoked snakehead fish, the products were still within the permissible bacterial contamination limit and considered safe for consumption.

Abstract

Moisturizers are a type of cosmetic product that function to hydrate the skin by reducing water evaporation from the skin surface and attracting water from the air into the dehydrated stratum corneum. Ingredients that reduce water evaporation are known as occlusives, while those capable of attracting water into the skin layers are referred to as humectants. This study employed an experimental method with a purposive sampling technique. The samples used were aqueous extracts of cucumber fruit (Cucumis sativus L.) combined with tomato fruit (Solanum lycopersicum L.) juice, obtained from fresh fruits, followed by phytochemical screening to determine their secondary metabolite content. The preparation was formulated as a moisturizer gel with varying concentrations of the combined cucumber and tomato fruit extracts of 5%, 10%, and 15%. Physical quality tests were subsequently conducted, including organoleptic, homogeneity, pH, adhesion, spreadability, and irritation tests, as well as a moisturizer effectiveness test using a Skin Analyzer on volunteers. The results of the phytochemical screening showed that the combination of cucumber and tomato fruit extracts contained alkaloids, flavonoids, tannins, saponins, steroids/triterpenoids, and glycosides. The moisturizer gel preparations containing the combination of cucumber and tomato fruit extracts demonstrated good stability, spread easily on the skin, did not cause irritation, and had a pH suitable for the skin. The effectiveness test showed that the highest increase in skin moisture was obtained at a concentration of 15%, with an average increase in moisture content of 72.11 ± 12.23%.

Abstract

Diuretics are drugs used to increase urinary excretion; however, long-term use of synthetic diuretics may cause adverse effects. Therefore, alternatives derived from natural sources are needed, one of which is sambau grass (Eleusine indica (L.) Gaertn.), which has potential diuretic activity. This study aimed to determine the secondary metabolite content and the effectiveness of sambau grass ethanol extract as a diuretic in male white rats (Rattus norvegicus). The study employed a laboratory experimental method using a Posttest Only Control Group Design. The experimental animals were divided into five groups: a negative control group (0.5% CMC-Na), a positive control group (furosemide), and three groups receiving sambau grass ethanol extract at doses of 100, 200, and 300 mg/kg body weight. The observed parameter was cumulative urine volume over 24 hours, which was analyzed using One-Way ANOVA followed by the Tukey HSD test. The results showed that sambau grass ethanol extract contained alkaloids, flavonoids, saponins, tannins, steroids/triterpenoids, and glycosides. The mean cumulative urine volumes at doses of 100, 200, and 300 mg/kg body weight were 8.14 ± 2.77 mL, 7.80 ± 2.25 mL, and 9.56 ± 2.47 mL, respectively. All three doses demonstrated relatively similar diuretic activity. Thus, sambau grass ethanol extract exhibits diuretic activity in male white rats (Rattus norvegicus).

Abstract

Lips are part of the face that have thinner skin compared to other parts of the body, making them more susceptible to dryness and loss of moisture. Dry lips can cause discomfort, so a preparation that can help maintain lip moisture is needed. One cosmetic preparation that can be used is lip balm. Carrot (Daucus carota L.) and orange sweet potato (Ipomoea batatas L.) are natural ingredients that have the potential to be utilized as moisturizers. This study aimed to determine whether a combination of ethanol extracts of carrot and orange sweet potato could be formulated into a lip balm preparation and to determine the concentration that provides the best moisturizing effect. The study was conducted experimentally using the maceration method with 80% ethanol. The extracts were formulated at concentrations of 2%, 4%, and 6%, along with a blank formulation. The evaluation included organoleptic, homogeneity, pH, stability, irritation, spreadability, adhesion, and moisture tests. Phytochemical screening showed the presence of alkaloids, flavonoids, tannins, triterpenoids, glycosides, and saponins. In particular, flavonoids act as antioxidants that help protect the lips from free radicals. The results showed that the preparation was semi-solid, homogeneous, stable for 8 weeks, and did not cause irritation. The moisture test showed that the 6% formulation provided the best result, increasing moisture from 30% to 48% after three days of use. Based on these results, the combination of ethanol extracts of carrot and orange sweet potato can be formulated into lip balm as a moisturizer, with the 6% formulation providing the best moisturizing effect. The preparation had a spreadability of 6.5–7.0 cm and an adhesion time of 4.0–4.8 seconds, meeting the established requirements.
