Abstract

Bunga Telang (Clitoria ternatea L.) dapat digunakan untuk berbagai pengobatan penyakit, salah satunya sebagai antidiabetes. Pengujian keamanan dari bunga telang masih terbatas untuk pengembangan obat baru. Tujuan penelitian ini untuk mengetahui gejala toksisitas serta nilai LD50 dari uji toksisitas akut. Metode OECD menggunakan hewan uji tikus betina galur wistar, yang dibagi menjadi kelompok kontrol negatif (Na-CMC) dan kelompok perlakuan (ekstrak etanol 70% bunga telang dengan dosis 5000 mg/kgBB). Hewan uji diamati gejala-gejala toksisitas dan berat badannya selama 14 hari. Pengamatan yang dilakukan meliputi kulit dan bulu, mata, tremor (gemetar), konvulsi (kejang), letargi (lesu), diare, dan mati. Hasil menunjukkan ekstrak mengandung alkaloid, fenol, flavonoid, saponin, tanin dan triterpernoid. Positif flavonoid dipertegas dengan identifikasi menggunakan kromatografi lapis tipis. Eluen yang digunakan n-heksan:etil asetat (4:2) dengan penampak bercak AlCl3 5%, nilai Rf yang didapat yaitu 0,77 dan 0,92. Berdasarkan hasil pengamatan, hewan uji tidak mengalami gejala-gejala toksisitas dan tidak mengalami perubahan bermakna pada berat badan hewan uji kontrol negatif dan perlakuan dengan nilai sig (2- tailed) pada Independent T-Test lebih dari 0,05. Hasil uji toksisitas akut menunjukkan nilai LD50 ekstrak etanol 70% bunga telang adalah >5000 mg/kgBB. Berdasarkan klasifikasi BPOM, nilai tersebut termasuk dalam kategori 5 yaitu praktis tidak toksik. Telang Flower (Clitoria ternatea L.) can be used for various treatments of diseases, like antidiabetic. Telang flower needs to be carried about the level of safety samplefor the development of new drugs. This study aimed to determine the symptoms and LD50 value of the acute toxicity with OECD 425 method. The test animals were female Wistar rats divided into controls (Na-CMC) and group with 70% ethanolic extract of telang flower at a doses of 5000 mg/kgBW. The test animals were observed for the symptoms of toxicity and body weight for 14 days. Observations were included skin, eyes, tremors, convulsions, lethargy, diarrhea, and death. The result obtained on alkaloids, phenols, flavonoids, saponins, tannins and triterpenoids. The flavonoids was confirmed by identification using thin layer chromatography with n-hexane: ethyl acetate (4:2) on AlCl3 5% spots, and the RF values obtained were 0.77 and 0.92. The result showed not experience significant changes in the body weight of controls (Na-CMC) and group at a doses of 5000 mg/kgBW with a sig (2-tailed) value on the Independent T-Test of more than 0.05. LD50 value showed of the 70% ethanolic extract of telang flower was > 5000 mg/kgBW. Based in category BPOM classification, it is include in category 5, practically non toxic.

Abstract

A wide range of compounds has been isolated from Lansium domesticum Corr. Secondary metabolites are commonly utilised as valuable resources for potential drug for research and development. An exemplary drug candidate must possess efficacy against the therapeutic target, safety, and favourable pharmacokinetic characteristics. The objective of this study was to determine the pharmacokinetic characteristics and toxicity potentials of 23 compounds derived from Lansium domesticum Corr. utilising the pKCSM online tool and the drug-likeness using swissADME online tool. Based on pKCSM prediction, compounds 14 (methyl angolensate) and 22 (7,14(27)-Onoceradiene-3,21-dione) from Lansium domesticum Corr. are identified as having favourable pharmacokinetic properties and are not expected to exhibit mutagenic or hepatotoxic effects. The LD50 values for compounds 14 and 22 were 2.983 and 1.813 (mol/kg), respectively, indicating their lethal dosages. In conclusion, only compound 14 that also met all the Lipinski’s Rule of Five.

Abstract

Indonesia is rich in biodiversity. One of the plants that grows widely in Indonesia is spinach (Amaranthus viridis). The aim of this research was to determine the secondary metabolite content of the ethanol extract of spinach leaves (A. viridis) qualitatively. This research consisted of 2 stages. The first stage is making simplicia and the second stage is screening phytochemical simplicia. The yield obtained from making simplicia was 20.83%. Based on the results, it can be concluded that spinach leaves (A. viridis) contain secondary metabolites of alkaloids, tannins, phenols, and saponins. The suggestion for this research is that the secondary metabolites can be isolated and then continued with activity tests.

Abstract

Overexpression of Human Epidermal Receptor-2 (HER-2) is the cause of the development of breast cancer, so HER-2 can be a target for anticancer therapy. Trastuzumab is the drug of choice in inhibiting the HER-2 receptor and tyrosine kinase, but the weakness of this drug is that drug resistance can occur. Naringin has potential as a breast cancer therapy. The research aims to determine the inhibiting overexpression of the HER-2 receptor by naringin using in silico method. Molecular docking has been carried out by optimizing naringin and trastuzumab, preparing the HER-2 protein target (PDB ID 3PP0), validating the molecular docking protocol, and docking naringin and trastuzumab on HER-2. The research produced a binding energy of -8,66 kcal/mol, while the binding energies of the original ligand and trastuzumab to HER-2 were -9,94 kcal/mol and -5,87 kcal/mol, respectively. The binding energy shows that naringin has potential as an anti-breast cancer agent as indicated by its stronger affinity for HER2.

Abstract

Non-steroidal anti-inflammatory drugs (NSAIDs) commonly used have associated side effects, necessitating the search for safer alternatives based on natural ingredients. This study aims to evaluate the effectiveness of a 70% ethanol extract emulgel of mulberry leaves (Morus alba L.) as an anti-inflammatory agent. The methodology employed is an experimental study involving the formulation of emulgel, which was tested on Wistar rats. The extraction process utilized maceration of mulberry leaves with 70% ethanol, yielding a concentrated extract that was then formulated into an emulgel. Anti-inflammatory activity was assessed using carrageenan induction and measuring edema volume in the test animals. The results indicated that the mulberry leaf extract emulgel exhibited significant anti-inflammatory activity, with a p-value of <0.023, demonstrating a meaningful difference compared to the positive control. The formula with a concentration of 0.32% showed the best efficacy at 180 minutes. The conclusion of this study highlights the potential of the 70% ethanol extract emulgel of mulberry leaves as a safe and effective alternative for anti-inflammatory therapy, recommending further safety testing to ensure its safe use in the community.

Abstract

Kalimantan Island has an endemic plants that holds potential as a medical one of them namely hiyung cayenne pepper fruit (Capsicum frutescens L. var hiyung) the local commodity of Tapin, South Kalimantan and known as the hottest pepper in Indonesia. It containts secondary metabolites and capsaicin that has potential to lower cholesterol. This study aims to identify secondary metabolite compounds and cholesterol-lowering activity of 70% ethanol extract of hiyung cayenne pepper fruit. Cholesterol lowering test was conducted in vitro by cholesterol complex formation method with FeCl3 using UV-Vis Spectrophotometer at maximum wavelength of 525 nm. The results of the phytochemical screening test showed positively contained alkaloids, triterpenoids, phenols, saponins, tannins and flavonoids. The result of cholesterol reduction test from concentrations of 20, 40, 60, 80, and 100 µg/mL were 53.62%, 58.30%; 64.19%; 70.02%; and 74.91%, respectively. So, the 70% ethanol extract of hiyung cayenne pepper fruit has the potential to reduce cholesterol with the lowest concentration being able to reduce cholesterol by more than 50%.
