Abstract

Pesticides are one of the components of agri input needed to protect plants from plant pests. However, massive use of pesticides can cause various negative impacts on plants, the environment, and human health. Excessive use of pesticides in rice cultivation is still a problem that is often found in Sukorame Village, Trenggalek. Seeing this condition, there needs to be more intensive education efforts for farmers. Seeing this problem, community service activities were carried out by the Faculty of Agriculture team at Kadiri University. Community service activities were carried out with the aim of providing education to farmers regarding more environmentally friendly and sustainable pest management. This program is also expected to reduce dependence on chemical pesticides and introduce Integrated Pest Management (IPM) techniques that can help farmers increase productivity without damaging the environment. Community service activities were carried out using the Participatory Aural Appraisal (PRA) approach. This method emphasizes community participation and involvement starting from the preparation, education, training and mentoring stages. Overall, the main objectives of the program to increase farmer awareness of the dangers of pesticides and introduce the IPM method have been achieved. The community began to understand the importance of environmentally friendly pest management. After the program was implemented, farmers began to provide follow-up. It is seen that some farmers have started to adopt the IPM method as a sustainable agricultural system.

Abstract

Abstract

Household organic waste is a major component of waste generation in densely populated residential areas, especially in urban areas with limited land and time. In RT 1 RW 7, Sumbersari Village, waste management is not optimally carried out, as the waste transportation system still relies on the door-to-door method, with no processing at the household level. This community service program aims to increase community awareness and involvement, especially among PKK mothers, in managing organic waste through bioconversion technology using Black Soldier Fly (BSF) maggots. The activity was carried out through a participatory approach in the form of a Focus Group Discussion (FGD) that was aligned with the PKK's regular meeting agenda. Residents were educated on how to use maggots, starting with sorting organic and non-organic waste. PKK mothers were also shown the decomposition process using BSF maggots firsthand, thereby convincing them of the program's benefits. Regular monitoring and evaluation were conducted to assess the program's effectiveness and changes in community behavior in sorting and processing waste. Monitoring results showed that most residents were quite enthusiastic about implementing the organic waste conversion program with maggots.

Abstract

Anthracnose disease caused by Colletotrichum gloeosporioides remains a major constraint in horticultural production, particularly in tropical regions. The cutinase enzyme plays an essential role in fungal pathogenicity by facilitating penetration of the host cuticle during the early stages of infection. Volatile organic compounds (VOCs) produced by Bacillus velezensis have been reported to exhibit antifungal activity. Inhibiting the cutinase enzyme disrupts the biochemical mechanism used by C. gloeosporioides to breach the plant’s waxy cuticle. By blocking cutin degradation, this anti-virulence strategy prevents initial infection and tissue invasion. However, information regarding their molecular interactions with cutinase is still limited. This study aimed to evaluate the potential of B. velezensis-derived VOCs as inhibitors of the cutinase enzyme from C. gloeosporioides using an in-silico approach. Antifungal bioactivity was predicted using PASS Server analysis, while binding affinity and inhibition constant (Ki) values were determined through molecular docking simulations with AutoDock Vina. Molecular interactions between VOC ligands and the target enzyme were analyzed based on their involvement with key active-site residues. The Pa–Pi analysis indicated that most VOCs, particularly terpenoids and medium-chain alcohols, exhibited higher predicted antifungal activity than inactivity. Molecular docking results showed that several VOCs interacted stably with the active site of cutinase, involving key residues Ser57, Ser136, and Gln137. Among the evaluated compounds, β-ionone exhibited the strongest binding affinity, as indicated by the lowest binding energy and inhibition constant. These results suggested that VOCs produced by B. velezensis, especially terpenoid compounds, had potential as biocontrol-based antifungal agents through inhibition of cutinase activity and required further validation through in vitro and in vivo studies.
