Abstract

Spirulina is Cyanobacteria containing active components which is potentially showing anticancer activity. The purposes of this study were to determine anticancer activity and selectivity of crude extractsof Spirulina cultured using Walne and organic media, and to detect the apoptosis. The stages of this study included cultivation and harvesting of Spirulina, active components extraction, anticancer test and apoptosisdetection. Anticancer activity was determined using MTT assay. The crude extracts of Spirulina from Walne and organic cultures contained active components of alkaloids, flavonoids, steroids, and saponins. Theseextracts were not toxic to normal breast cells (MCF-12a), but showed cytotoxic activity in breast cancer cells (MCF-7). The crude extract of Spirulina from Walne culture had IC50 value of 36.23 ppm and selectivity index 30.07, while the IC50 of organic culture was 117.78 ppm and selectivity index 7.17. Detection of apoptosis with Hoechst dye 33342 showed the apoptotic activity of Spirulina crude extract against MCF-7 cells.

Abstract

Differences in snakehead fish species can be detected through several markers, including DNA banding patterns. Isolation to obtain high-quality DNA is one of the basic rules that must be met in molecular studies, especially in DNA fingerprinting. DNA isolation methods can be done conventionally with CTAB (Cationic Hexadecyl Trimethyl Ammonium Bromide) or by using kits that have been provided by several companies to facilitate DNA isolation such as Qiagen and Thermoscientific. Therefore, it is necessary to compare the results of DNA isolation from various isolation methods so as to produce good DNA extraction. The results showed that the CTAB method was the best method. This is evidenced by the use of 2% CTAB as an extraction buffer has been able to break down cells and produce good quality DNA indicated by the presence of genomic DNA bands that match the target and have a high level of effectiveness.

Abstract

Indonesia is a maritime country and an archipelagic country. Indonesia has abundant and diverse fish resources. Abundant fish resources make fish easy to obtain and cheap. However, there are still many people who do not like to consume fish and do not know the benefits contained in the body of fish. This activity aims to provide education about eating fish through puppet stories, so that students understand the benefits of fish in body growth. Community service activities were carried out on November 2, 2022 at the 178 Pekanbaru Elementary School, which were attended by 152 students. The method used is pretest and posttest design. From the results of the activity, it appears that in the pretest activities only a few students already know about the types of fish, the nutritional content of fish, the benefits of eating fish, and processed fish products. Then during the posttest, many students were able to answer and provide explanations regarding the types of fish, fish content, benefits of fish, and processed fish products. This service activity is expected to increase students insight and knowledge about types of fish, nutritional content, benefits of eating fish, processed fish products so that it is hoped that in the future students will like to eat fish.

Abstract

Indonesia, as an archipelagic country with substantial fishery potential, faces challenges related to low fish consumption levels among its population. Muntai Barat Village in Bantan District, Bengkalis Regency, Riau Province, possesses significant fishery resources that remain underutilized. In response, a community service initiative comprising Fish Consumption Promotion Counseling and Fishery Product Demonstration was conducted to enhance public understanding of the importance of fish consumption and its health benefits, and to stimulate entrepreneurship and the creative economy by leveraging local fishery potential. The methods employed included interactive counseling and demonstrations of processed fish products, specifically mackerel and shrimp dumplings. Participants included 15 members of the village's women's group (PKK). The results indicated a significant increase in participants' knowledge of the nutritional content and benefits of fish, with a 45.34% improvement in post-test scores compared to pre-test results. All participants (100%) successfully created fish dumpling products, reflecting improved skills in processing fish into economically valuable products. Support from the village government in the form of business fund access for participants is expected to encourage the establishment of 3-5 fishery-based business units within six months. In conclusion, this activity effectively increased community awareness and skills in utilizing local fishery resources, potentially fostering the growth of the creative economy in the village.

Abstract

Edible film is a primary packaging in the form of a thin layer and functions as a coating for food ingredients, which can be made from hydrocolloid compounds. Carrageenan polysaccharide is one of the hydrocolloids used to make edible films because it produces strong gel properties. Eucheuma cottonii is a seaweed that produces relatively high levels of kappa carrageenan, namely around 50%. This research aims to determine the physical and mechanical characteristics of edible film with carrageenan flour from E. cottonii extracted with different base solvents. The research method was carried out experimentally with an experimental design, namely a non-factorial Completely Randomized Design (CRD) with three levels of treatment, namely differences in carrageenan flour extracted with KOH, NaOH and Ca(OH)2 solvents. The parameters of this research are physical and mechanical analysis of edible film including testing tensile strength, thickness, percent elongation, and water vapor transmission rate. The research results showed that the best results were in the treatment of carrageenan flour extracted with KOH solvent, where the physical and mechanical analysis of the edible film showed a tensile strength value of 9.49±0.50 MPa, a thickness of 0.03±0.003 mm, a water vapor transmission rate of 30.13±0.41 g/m2/day, and a percent elongation 9.86±0.41%.
