Abstract

Anchovies (Stolephorus commersonnii) are a good source of calcium and protein. Intake of these nutrients is associated with child height. According to the Ministry of Health of the Republic of Indonesia's Nutrition Status Study Report, the prevalence of stunting among Indonesian children in 2022 was 21.6%, which is still higher than the World Health Organization's (WHO) standard of <20%. This study aims to optimize the formulation of anchovy extract gummy candies that meet quality standards and contain high levels of calcium and protein. Anchovy powder contains 328.64 mg/kg of calcium, 28.46% protein, 0.12% ash, and 5.3% moisture. The anchovy extract was prepared using the maceration method with a 70% ethanol solvent. Four gummy candy formulations were made based on variations in anchovy extract concentration: 10%, 7.5%, 3%, and 1.5%. Formulation 4 (F4) with 1.5% anchovy extract produced the best physical quality, with a standard deviation (SD) of <5% for the weight uniformity test. The organoleptic test results for F4 are as follows: the anchovy taste is mild, the color is dark purple, the smell is slightly aromatic, the shape is consistent with the mold, and the texture is chewy. F4 gummy candies contain 1,490.64 mg/kg of calcium, 27.49% protein, 9.17% ash, and 3.96% moisture.  A serving size of one gummy (1.5 g) contains 2.24 mg of calcium, 0.41 g of protein, 0.06 g of total fat, 0.07 g of carbohydrates, 0.55 g of sugar, and 4.18 mg of sodium. F4 meets quality standards and is high in calcium and protein.

Abstract

Public awareness of the importance of health is increasingly developing towards a better direction. One method is by consuming supplements and herbal medicines to increase the body's resistance to avoid certain diseases. The consumption of safe, high-quality and useful herbal supplements and medicines needs to be cultivated and preserved by the community. Therefore, community service was carried out which consisted of 3 stages of activities, namely distributing questionnaires before counseling, counseling, and distributing questionnaires after counseling to see whether there was an influence of counseling on knowledge. The aim of this activity is to increase public awareness of the misuse and wrong use of supplements and herbal medicines. Community service was carried out at Harapan Sehati Hospital in the Bojonggede area, Bogor Regency with a total of 30 participants.   are used for the counselling. Data were collected using a questionnaire consisting of demographics, knowledge and behavior of participants regarding the use of supplements and herbal medicines. Data processing uses descriptive analysis methods, and the non-parametric Wilcoxon Signed Rank Test. The majority of participants were women (83.3%) and aged 17-25 years (43.3%). As many as 66.7% of participants were college graduates and 96.7% worked as private employees. The percentage of participants who had good knowledge about supplements and traditional medicine before (pre) counseling was 43.3%. Good knowledge becomes 80% after (post) counseling. The data shows a p-value of 0.001 (<0.05), which means there is an influence of counseling, or there is a significant difference between knowledge before and after being given education. It can be concluded that this series of activities is effective in increasing knowledge about the use of supplements and herbal medicines, so it is hoped that it can prevent misuse.

Abstract

Response to angiotensin-converting enzyme inhibitors (ACEIs)-based antihypertensive therapy varies between individuals, which is largely influenced by genetic factors. The ACE gene has several polymorphisms that can affect enzyme levels and therapeutic efficacy. This study aims to explore the relationship between genetic variations in the ACE gene and response to captopril, lisinopril, ramipril, and enalapril in hypertensive patients. This study used a bioinformatics and pharmacogenomics approach by analyzing data from PharmGKB, Ensembl, and GTEx Portal. Genetic polymorphisms were analyzed to evaluate their association with ACEI efficacy using a descriptive statistical approach. Results: Four single nucleotide polymorphisms (SNPs) in the ACE gene were found to be associated with response to ACEI. Variants rs4291 and rs1799752 were associated with captopril efficacy, where the AA genotype showed a decrease in the severity of renal failure. The rs1799752 variant was also associated with lisinopril and enalapril, with the DD genotype providing greater blood pressure reduction. In addition, rs4359 and rs4344 were correlated with the efficacy of ramipril, especially in the CC+TT and AA+GG genotypes. Genetic variation in the ACE gene plays a role in determining the response to ACEI therapy. Pharmacogenetic approaches have the potential to improve the efficacy and safety of antihypertensive treatment.

Abstract

Bidara leaves (Ziziphus mauritiana) are a herbal plant that is known to be rich in bioactive compounds, especially flavonoids, which have antioxidant, anti-inflammatory and antimicrobial activities. This study aims to determine the total flavonoid content in bidara leaf extract and evaluate the potential of its secondary metabolites for pharmaceutical and cosmetic applications. Phytochemical screening was carried out to identify secondary metabolite content, which included flavonoids, alkaloids, saponins, tannins and phenolics. Extraction was carried out using the maceration method with 96% ethanol solvent, followed by filtering and evaporation of the solvent using a rotary vacuum evaporator at a temperature of 40°C to produce a thick extract. Determination of total flavonoid levels was carried out using the UV-Vis spectrophotometric method with aluminum chloride reagent at a wavelength of 441 nm, using quercetin as a standard. The results showed that the total flavonoid content in bidara leaf extract reached 3202.43 mg QE/100 g dry extract. This high flavonoid content supports antioxidant activity through the mechanism of capturing free radicals, chelating transition metal ions, and inhibiting lipid peroxidation. In addition, the flavonoids in the extract have the potential to be anti-inflammatory by inhibiting pro-inflammatory enzymes and modulating cytokines, as well as antimicrobial through disrupting microbial membranes. The conclusion of this research is that bidara leaf extract has great potential to be developed as an active ingredient in pharmaceutical and cosmetic formulations.
